Quantitative RT-PCR profiling of miRNA expression of non-small cell lung cancer patients against healthy controls

ABSTRACT: Lung cancer (LC) is one of the major cancers, with survival of patients dictated by the time of diagnosis. Cell-free circulating miRNAs have been proposed as candidate biomarkers for non-small cell lung cancer. The goal of this study was to profile the miRNAs in blood plasma of lung cancer patients diagnosed with squamous cell carcinoma (SCC, n=14) or adenocarcinoma (AD, n=6), and healthy individuals (HD) using miRCURY LNA miRNA qPCR Serum/Plasma Panel (Exiqon). Average Cq of detected assays was used for normalization of miRNA expression.

Lung cancer is one of major cancers, and survival of lung cancer patients is dictated by the timely detection and diagnosis. Cell-free circulating miRNAs were proposed as candidate biomarkers for lung cancer. These RNAs are frequently deregulated in lung cancer and can persist in bodily fluids for extended periods of time, shielded from degradation by membrane vesicles and biopolymer complexes. Project description:We used microarrays to perform systematic profiling of human microRNAs in plasma from nasopharyngeal carcinoma (NPC) patients to find potential biomarkers. By comparing the plasma microRNA profiles of the NPC patients and healthy donors, potential biomarkers for NPC were investigated. A total of 39 microRNAs were aberrantly expressed based on 50 Agilent microarrays containing 887 human microRNAs 50 microarrays containing 887 human microRNAs were used to screen for potential biomarkers with significant differential expression levels between 31 NPC patients and 19 healthy donors.

Project description:To evaluate the impact of the RNA purification method on extracellular RNA (exRNA) sequencing, 8 different RNA purification kits were compared by applying Small RNA sequencing (Illumina) to exRNA from human healthy donor plasma. Minimum and maximum plasma input volumes recommended by the manufacturers were tested in triplicate. Due to donor privacy concerns the raw data for this study have been submitted to the controlled-access archive EGA under the accession EGAS00001005263.

Changes in exosomal circulating microRNAs in grazing cattle

Project description:To investigate the potential effect of grazing movement on miRNA circulation in cattle, here we profiled miRNA expression in centrifugally prepared exosomes from the plasma of both grazing and housed Japanese Shorthorn cattle. Microarray analysis of the c-miRNAs resulted in detection of a total of 231 bovine exosomal miRNAs in the plasma, with a constant expression level of let-7g across the duration and cattle groups. Expression of muscle-specific miRNAs such as miR-1, miR-133a, miR-206, miR-208a/b, and miR-499 were undetectable, suggesting the mildness of grazing movement as exercise. Changes in miRNA expression in plasma exosome of cattle was measured at during 0, 1, 2 and 4 months of grazing or housing. Plasma exosome samples prepared from three cattle for each treatment were collected and mixed within the treatment at each time for microarray analysis .

Real-time PCR analysis of microRNA expression in differentiating human skeletal muscle cells

Project description:Human skeletal myoblast cell line LHCN-M2 (Zhu et al., Aging Cell (2007), vol. 6, pp 515-523) were plated on collagen-coated 6-well Falcon cell culture plates at 500 000 cells/well, differentiation was induced 24h later by switching to DMEM supplemented with 0,01 mg/ml insulin and 0,1 mg/ml transferrin. Cells were collected at 0, 3, and 7 days of differentiation time points. Total RNA was extracted and miRNA expression evaluated by quantitative real-time PCR. MicroRNAs differentially present in the plasma of HIV elite controllers

Project description:Elite controllers maintain HIV-1 viral loads below the limit of detection. The mechanisms responsible for this phenomenon are poorly understood. As microRNAs (miRNAs) are regulators of gene expression and some of them modulate HIV infection, we have studied the miRNA profile in plasma from HIV elite controllers and chronically infected individuals and compared against healthy donors. Several miRNAs correlate with CD4+ T cell count or with the known time of infection. No significant differences were observed between elite controllers and healthy donors; however, 16 miRNAs were different in the plasma of chronic infected versus healthy donors. In addition, levels of hsa-miR-29b-3p, hsa-miR-33a-5p and hsa-miR-146a-5p were higher in plasma from elite controllers than chronic infected and hsa-miR-29b-3p and hsa-miR-33a-5p overexpression significantly reduced the viral production in MT2 cells. Therefore, levels of circulating miRNAs might be of diagnostic and/or prognostic value for HIV infection. Additionally, hsa-miR-29b-3p and miR-33a-5p may be used in therapeutic strategies. An exploratory cross-sectional study of microRNA levels in EDTA plasma samples. Plasma samples were obtained from 24 subjects and were classified in 3 groups, 9 Elite Controllers (defined as individuals with plasma viral load (PVL) < 50 copies/ml, CD4 count >350/ml), 9 chronic HIV patients (CH) under anti-retroviral treatment and 6 healthy HIV negative donors (HD). This study was approved by the HuM-CM-)sped Foundation Ethics Committee and informed consent was obtained from all subjects.
