Microarray experiment on M. incognita infected and uninfected material in A.thaliana Col-0 plants and A. thaliana ERF6 mutated plants

ABSTRACT: In this experiment we investigate the transcription profile of M. inocognita infection at the start of the infection and 7 days later in A. thalinana Col-0 plants. This was compared to M. incognita infected material of the plant line erf6-1, a T-DNA insertion mutant in ERF6. The data showed the role of ERF6 during early M. incognita infection in Arabidaaaaaaaaaaopsis besides more insight in the transcription regulated by M. incognita in Col-0 the wildtype plant. Samples are taken from whole root systems of 14 (0 days after infection) or 21 (7 days after infection) day old plants.

Project description:In this experiment we measured the transcriptional response of ten tomato cultivars when infected by the plant-parasitic nematode M. incognita. The ten cultivars showed differential levels of susceptibility to M. incognita infection. Ten-days old plants were exposed to nematodes and harvested 1, 2, 3, 4, 7, or 10 days post infection. Galls or representative uninfected tissues were harvested and used for RNA sequencing. The data was used to investigate the link between susceptibility to M. incognita infection and gene expression in tomato.

The aim of the experiment was to determine whether the tcp mutatants affected gene expression patterns induced by nematode infection.

Project description:In this experiment we evaluate the transcriptional response to beet cyst nematode infection in wild-type (Col-0) and a upl3 knocked-down Arabidopsis thaliana. The data indicates that the upl3 knock-down alone does not induce a strong transcriptional regulation in the plant. However, the interaction of the mutation and infection by H. schachtii results in a strong transcriptional differential regulation, likely involving the plant’s immune responses. Samples were taken from whole root systems of 14 day-old seedlings (0 days post-infection) and mock or H. schahtii-infected 21 day-old seedlings (7 days post-infection). Each sample contained at least 18 plants and 4 biological replicates of each sample were collected.
