RNA-seq of mouse myeloma cell line SP 2/0 overexpressed with Gm40600 against vector control

ABSTRACT: We found that some novel genes such as Gm40600 mRNA significantly changed in plasma cells. Because of plasmablast-like, mus musculus myeloma SP 2/0 cell line was selected to test the effect of Gm40600 overexpression on plasmablast/plasma cells. Gm40600 cDNA was cloned into lentiviral vector LV201 to generate Gm40600 protein. Gm40600-expressing LV201 was then transfected into SP 2/0 cells, and stable transfectants were identified by drug selection. To identify the effect of Gm40600 overexpression on gene expression, we determined mRNA profiles in Gm40600-overexpressed and vector-transduced SP 2/0 cells by RNA-sequencing. RNA-seq was done with an Illumina HiSeq 2500 instrument at GENEWIZ, Suzhou, China.

Because of plasmablast-like, mus musculus myeloma SP 2/0 cell line was selected to test the effect of BC094916 overexpression on plasmablast/plasma cells. BC094916 cDNA (General Biosystems, Anhui, China) was cloned into lentiviral vector LV122 or LV201 (Fugene Corp., Guangzhou, China) to generate BC094916 and BC094916-EGFP fusion protein, respectively. BC094916-expressing LV122 was then transfected into SP 2/0 cells, and stable transfectants were identified by drug selection (Puromycin, Sigma, 10 μg/ml). BC094916 overexpression suppressed SP 2/0 cell proliferation by inducing cell apoptosis. Importantly, BC094916 overexpression effectively suppressed tumor progression in the SP 2/0 xenograft mouse model. In addition, we found that BC094916 is a suppressive transcriptional factor. To verify its target genes, we determined mRNA profiles in BC094916-overexpressed and vector-transduced SP 2/0 cells by RNA-seq. Self-maintenance of rat C6 glioma stem cells (GSCs) by their non-GSC progenies

Project description:Analysis of Hoechst 33342 dye-effluxing side population cells (SP cells defined as glioma stem cells, GSCs) and dye-retaining main population cells (MP cells defined as non-GSCs) that were FACS-sorted from the C6 glioma cell line stably expressing EGFP (C6-eGFP). ECM-related genes, such as Col4a1 and Col4a2, and the iron carrier gene Tf are upregulated in MP cells. Results provide the insight into molecular basis underlying the maintenance of GSCs by non-GSCs. Gene expression profiles were compared between SP and MP cells just after FACS-sorting from the whole C6-eGFP cells based on their Hoechst-effluxing abilities.

Functional Connection with Cardiac Tissues Confers Maturation Phenotypes in hPSC-Derived PHOX2B::GFP Expressing Sympathetic Neurons

Project description:Neurons derived from human pluripotent stem cells (hPSCs) are a remarkable tool for modeling human neural development and diseases. However, it remains largely unknown whether the hPSC-derived neurons can be functionally coupled with their target tissues in vitro, which is essential for understanding inter-cellular physiology and further translational studies. Here, we demonstrate that hPSC-derived sympathetic neurons can be obtained from hPSCs and that the resulting neurons form physical and functional connections with cardiac muscle cells. By use of multiple hPSC reporter lines, we recapitulated human autonomic neuron development in vitro, and successfully isolated PHOX2B::eGFP+ neurons exhibiting sympathetic marker expression, electrophysiological properties, and norepinephrine secretion. With pharmacological and optogenetic manipulations, the PHOX2B::eGFP+ neurons controlled the beating rates of cardiomyocytes, and their physical interaction led to neuronal maturation. Our study lays a foundation for the specification of human sympathetic neurons and for the hPSC-based neuronal control of end organs in a dish. Using the four genetic reporter systems (OCT4::eGFP, SOX10::eGFP, ASCL1::eGFP, and PHOX2B::eGFP reporter hESC lines), we were able to purify discrete cell populations at four differentiation stages, recapitulating the sympathoadrenal differentiation process in vitro with purified and defined populations in four specific differentiation stages. We performed transcriptome analysis of OCT4::eGFP+ cells (3 biological replicates, representing undifferentiated hESCs), SOX10::eGFP+ cells (3 biological replicates, multi-potent neural crest), ASCL1::eGFP+ cells (3 biological replicates, putative sympathoadrenal progenitors), and PHOX2B::eGFP+ cells (2 biological replicates, putative sympathetic neuronal precursors).

SP analysis and sorting were done using a FACSVantage SE.The breast cancer cells were sorted into SP and non-SP cells.Total RNA was isolated from the FACS-sorted SP or non-SP cells, and gene expression signiture was detected by microarray.
