RNA-Seq of chicken liver samples treated with chronic heat stress

ABSTRACT: To study the effect of chronic heat stress on producing laying type hens, RNA-Seq was performed on liver samples during a 4-week heat stress experiment. RNA-Seq samples were collected 3-hour, 2-weeks, and 4-weeks post heat from both control (unheated) and treatment (heated) birds. RNA was extracted from the tissues with mirVana miRNA Isolation Kit, treated with DNase, and mRNA cDNA libary were prepared with Illumina TrueSeq Stranded mRNA Library Prep Kit. Sequencing was done on 4 lanes with 12 libraries per lane on Illumina HiSeq 3000 platform.

Immunomodulatory effects of heat stress and lipopolysaccharide on the bursal transcriptome in two chicken lines

Project description:For this study, bursal transcriptome responses to an acute heat stress and/or lipopolysaccharide (LPS) were investigated in a broiler line (heat and disease susceptible) and an inbred Fayoumi line (heat and disease resistant) of chickens. In a 2 x 2 design, 22 day-old birds were exposed to heat stress (35°C for 7 hours), lipopolysaccharide (100 µg/kg average body weight per line), or both stressors. Thermoneutral temperature (25°C) and phosphate buffered saline were used as the respective controls. Tissue samples were collected from the bursa of Fabricius and used to isolate high quality RNA. cDNA libraries (n = 31) were constructed and sequenced (2 technical replicates per library; 62 total datasets) on the HiSeq 2500.

Gene Expression Profiling of Three Tissues in Chicken with Heat Stress by Affymetrix Microarray

Project description:Heat stress is a very complex physiological process. At present, there are some reports about physiological function and pathological changed on chicken heat stress. We use DNA microarrays to detail the global programme of gene expression underlying heat stress and identified distinct classes of differentially expressed (DE) genes during this process. Chicken were randomly divided into two groups to study the effects of heat stress on gene expression of brain, liver and leg muscle. Twenty-four birds (12 male and 12 female) were allocated to heat stress or post-heat stress groupM-oM-<M-^H40M-BM-11M-BM-0C,3hM-oM-<M-^I and control or pre-heat stress groupM-oM-<M-^H28M-BM-11M-BM-0CM-oM-<M-^I.

mRNA-seq of the jejunum mucosa in LSL and LB laying hens following the variations of dietary calcium and phosphorus levels

Project description:Calcium (Ca) and phosphorus (P) are essential micronutrients that are linked to a wide set of biological processes. In laying hens there is still uncertainty about the optimal Ca/P ratio in feed and further strategies for the reliable mineral restriction in poultry diets are required. The dataset is based on Lohmann Selected Leghorn (LSL) and Lohmann Brown (LB) laying hens sampled at peak performance. The experimental design comprises four dietary groups differed in Ca (recommended vs. 15 % reduction) and mineral P (adequate vs. 20 % reduction) levels; 1) control diet (Con; Ca=34.4g/kg, P=5.3 g/kg and Ca/P ratio=7.45), 2) Low Ca and P diet (LCaP), 3) low Ca diet (LCa), and low P diet (LP). Jejunal transcriptome profiles were assessed by mRNA sequencing in a total of 80 animals (10 hens per experimental group for each laying line) at sampling age of 31 weeks) to approximate the consequences of variable Ca and P supply.

Differential gene expression in breast muscle is similar in heat stressed layers and boilers at control temperatures

Project description:In total 32 Male broiler and 32 male layer birds at the age of 6wk were subjected to two treatment: control (21C temperature and 50% RH) and heat-stress (32C temperature and 80% RH) in four climate chambers. After exposing the birds to said temperature and humidity the breast tissues samples were taken to get RNA and perform microarray experiment. RNA samples from two birds, from same treatment, same breed, and same chamber were pooled before hybridization.

Transcription profiling of mouse preshocked hsp70(-/-) and (+/+) cells to assess thermotolerance

Project description:From preliminary experiments, HSP70 deficient MEF cells display moderate thermotolerance to a severe heatshock of 45.5 degrees after a mild preshock at 43 degrees, even in the absence of hsp70 protein. We would like to determine which genes in these cells are being activated to account for this thermotolerance. Experiment Overall Design: Two cell lines are analyzed - hsp70 knockout and hsp70 rescue cells. 6 microarrays from the (-/-)knockout cells are analyzed (3 Pretreated vs 3 unheated controls). For the (+/+) rescue cells, 4 microarrays are used (2 pretreated and 2 unheated controls). Cells were plated at 3k/well in a 96 well plate, covered with a gas permeable sealer and heat shocked at 43degrees for 30 minutes at the 20 hr time point. The RNA was harvested at 3hrs after heat treatment.

In laying hens, the optimal Ca/P ratio in feed is inconsistent but necessary for reliable schemes of mineral restriction in poultry diets. This study investigates the effects of dietary treatments varying in the Ca and P levels in two laying hen strains (Lohmann Brown-Classic and Lohmann LSL-Classic) at the peak of egg production (31 weeks of age). Alterations in Tissue microRNA after Heat Stress in the Conscious Rat: Potential Biomarkers of Organ-Specific Injury

Project description:Heat illness, which remains an occupational and environmental hazard, can be caused by excessive strain or heat load in combination with other factors. In the 10-year period from 1999 to 2009, an average of 658 annual heat-related deaths occurred in the United States. While heat stress at the cellular level has been studied, a paucity of risk assessment and injury biomarkers as well as therapeutic interventions remains. To identify novel biomarkers and to further understand the molecular mechanisms of heat stress, we identified differentially expressed microRNAs (miRNAs) in the heart, liver, and kidney from a conscious rat model at three time points. We distinguished the effect in animals with histopathological evidence of heart injury from those without evidence of organ injury. In animals without evidence of injury, we identified a total of 45 unique modulated miRNAs, whereas in the three animals with evidence of injury, we identified 171 unique differentially expressed miRNAs. The majority of the perturbed miRNAs were both time and tissue specific. Using the data from a microarray companion study, we identified the mRNAs that are the predicted targets of the differentially expressed miRNAs and performed pathway enrichment analysis. The enrichment analysis suggested that the perturbed miRNAs are involved in biological pathways related to energy metabolism, the unfolded protein response, and organ injury. These miRNAs may serve as organ-specific heat stress biomarkers of exposure or effect, as well as identify potential targets of heat illness prevention. Heart, liver, lung and kidney tissue was collected from rats at Tc,max and at 24 and 48 h for both heat exposed rats (n=4 to 6) or time matched, unheated controls (n= 4 to 6)
