RNA-Seq of adipose tissue from mice infected with Trypanosoma brucei

ABSTRACT: The goal of this experiment was to study the alterations that occur in the adipose tissue of mice during a T. brucei infection. Gonadal adipose tissue from C57BL/6J mice infected with T. brucei AnTat 1.1E 90-13 parasites was collected at day 0 and 26 post-infection. Total RNA was extracted from 3 samples of non-infected adipose tissue and 2 samples of adipose tissue on day 26 post-infection. Day 6 post-infection adipose tissue samples (n=3) were already published (E-MTAB-4061).

Project description:We asked whether the in-vivo transcriptome of T. brucei AnTat 1.1E 90-13 parasites collected from adipose tissue and blood is different. We collected gonadal adipose tissue of infected mice on day 6 post-infection, when a high number of parasites is expected, and blood on day 4 post-infection when slender-forms are the most abundant form of the parasite. Parasites from blood were purified over a DEAE column. Total RNA was extracted from two independent samples of blood and three independent samples of fat and subjected to RNA-sequencing.

IL-17 signalling is critical for controlling subcutaneous adipose tissue wasting and parasite burden during chronic Trypanosoma brucei infection [single-cell RNA-seq]

Project description:In the skin, Trypanosoma brucei colonises the subcutaneous white adipose tissue (WAT) and harbours a pool of parasites competent for forward transmission. The interaction between parasites, adipose tissue, and the local immune system is likely to drive adipose tissue wasting and weight loss observed in cattle and humans infected with T. brucei. However, mechanistically, this process is not fully understood. Here, using several complementary approaches including mass cytometry by time of flight, bulk and single cell transcriptomics, we found that T. brucei infection drives a local expansion of several IL-17A-producing cells in the murine WAT, including TH17 and V6+ T cells. We also found that global IL-17 deficiency, or mice lacking IL-17 receptor expression specifically on adipocytes, were protected from infection-induced WAT wasting and weight loss. Unexpectedly, we found that abrogation of IL-17 signalling on adipocytes results in higher burden of extravascular parasites in the WAT. Taken together, our study highlights the central role of IL-17 signalling on adipocytes in controlling WAT responses to infection, suggesting that adipocytes are a critical coordinator of the tissue immune response to T. brucei infection.

Interleukin-17 drives sex-dependent weight loss and changes in feeding behaviour during Trypanosoma brucei infection

Project description:African trypanosomes, the causative agents of Human and Animal African trypanosomiasis or sleeping sickness, reside in tissue niches proposed to be important for disease outcome and transmission. Here, we demonstrate that parasites in the inguinal white adipose tissue (iWAT) niche induce sexually dimorphic physiological and immunological responses. Following chronic Trypanosoma brucei infection, male mice experience weight loss, reduced adipose tissue mass and altered tissue function, as well as changes in feeding behaviour, whereas females do not. We identified that interleukin-17 (IL-17), a cytokine that we show is elevated in sleeping sickness patients, orchestrates a sex-specific response to T. brucei infection in experimental infections. Deletion of murine IL-17a/f abolishes infection-associated weight loss, alters feeding behaviour, and limits adipose tissue wasting in male mice only. We propose that these effects might be triggered locally in adipocytes via engagement of IL-17 with its cognate receptor leading to lipolysis and tissue wasting, and/or systemically, via IL-17 signalling in the hypothalamus, potentially suggesting that IL-17 signalling coordinates brain-adipose tissue communication during sleeping sickness. Our findings also suggest a key sex-dependent role for the IL-17 isoforms IL-17A and IL-17F in regulating adipose tissue and energy balance during infection. Altogether, the results presented here open new directions to understand energy balance and brain-adipose tissue communication during chronic infection.

RNA-seq of gonadal adipose tissue from male and female Trim28-WT and Trim28 adipose specific KO mice

Project description:Many genes have been implicated in WAT lipid metabolism, including tripartite motif containing 28 (Trim28), a gene proposed to primarily influence adiposity via epigenetic mechanisms in embryonic development. We set out to determine if adipose specific deletion of Trim28 led to changes in adipose tissue function and molecular phenotype. We performed transcriptomics analysis on adipose tissue taken from WT and adipose specific Trim28 KO mice to investigate their molecular phenotype, and to identify pathways altered in KO animals.

Rosiglitazone-induced molecular changes in adipose progenitors

Project description:White adipose tissue regulates metabolism; the importance of this control is highlighted by the ongoing pandemic of obesity and associated complications such as diabetes, atherosclerosis, and cancer. White adipose tissue maintenance is a dynamic process, very little is known about how pharmacologic stimuli affect such plasticity. Combining in vivo lineage marking and BrdU labeling strategies, we found that rosiglitazone, a member of the thiazolidinedione class of glucose-lowering medicines, markedly increases the evolution of adipose progenitors into adipocytes. Notably, chronic rosiglitazone administration disrupts the adipogenic and self-renewal capacities of the stem cell compartment and alters its molecular characteristics. These data unravel unknown aspects of adipose dynamics and provide a basis to manipulate the adipose lineage for therapeutic ends. The goal of this gene expression array was to identify changes in molecular expression in adipose progenitors isolated from mice that underwent two-month rosiglitazone treatment. Adipose SV GFP+ cells (adipose progenitors) were FACS-isolated from adult AdipoTrak mice that had been treated with or without rosiglitazone (0.0075%) for 2 months. RNAs isolated from these cells were used for microarray. Each cohort contains 3-4 mice, each experimental group (-TZD and +TZD) contains 3 cohorts.

Slow growing behavior in African trypanosomes during adipose tissue colonization

Project description:When Trypanosoma brucei parasites, the causative agent of sleeping sickness, colonize the adipose tissue, they rewire gene expression. Whether this adaptation affects population behavior and disease treatment remained unknown. By using a mathematical model, we estimate that the population of adipose tissue forms (ATFs) proliferates slower than blood parasites. Analysis of the ATFs proteome, measurement of protein synthesis and proliferation rates confirm that the ATFs divide on average every 12hr, instead of 6hr in the blood. Importantly, the population of ATFs is heterogeneous with parasites doubling times ranging between 5hr and 35hr. Slow-proliferating parasites remain capable of reverting to the fast proliferation profile in blood conditions. Intravital imaging shows that ATFs are refractory to drug treatment. We propose that in adipose tissue, a subpopulation of T. brucei parasites acquire a slow growing behavior, which contributes to disease chronicity and treatment failure.

We analyzed differential gene expression, exon usage and RNA editing. RNA sequencing on hippocampus of wild-type C57BL/6 mice, congenic B6.129-PrnpZH1/ZH1 and coisogenic C57BL/6-PrnpZH3/ZH3 mice (3 month-old males, n=4 per genotype).

Tissue-autonomous regulation of adipose expansion by core clock protein REVERBα

Project description:The circadian clock coordinates energy metabolism, with REVERBα considered a dominant regulator of lipid metabolism. REVERBα is a core clock protein, which demonstrates robust rhythmic expression across metabolic tissues. Here, we explore its role in white adipose metabolism, and propose that REVERBα plays a key part in the response of adipose to obesity. Mice globally lacking Reverbα (Reverbα-/-) display adiposity, increased susceptibility to diet-induced obesity, and up-regulation of the lipogenic proteome in white adipose depots. When Reverbα is deleted selectively in adipose (ReverbαFlox2-6AdipoCre), however, we observe only modest phenotypic and transcriptomic changes in the basal state, with altered regulation of clock and extracellular matrix (ECM) pathways. Thus the adipose-autonomous actions of REVERBα are shaped by context. On high-fat diet challenge, ReverbαFlox2-6AdipoCre mice accumulate more adipose mass than littermate controls, and are spared obesity-related inflammation. Gene expression analysis reveals pathways of lipid handling to be up-regulated. These findings therefore highlight a critical role for REVERBα in the regulation of adipose tissue expansion. Further, when transcriptomic data is integrated with the REVERBα cistrome, we detect strong associations of REVERBα binding sites, not only with the clock and ECM targets de-repressed under normal conditions, but with the broad range of metabolic genes unmasked as REVERBα targets by obesity. Thus our data favour a new interpretation of REVERBα function, not as conferring rhythmicity to lipogenesis, but as buffering against lipogenic cues asynchronous to the normal temporal routine.
