Brain endothelium gene expression during group B streptococcus (GBS) infection

ABSTRACT: The aim of this study was to investigate the response of human brain endothelial cells to bacterial (group B streptococcus, GBS) infection. Results: GBS WT strain infection results in a specific gene induction pattern that is different from the pilA mutant, but not other mutants such as pilB and srr-1. Conclusion: These findings suggest that the GBS PilA protein contributes to gene induction in brain endothelium.

Project description:30-day-old Cftr-/- homozygous for the S489X, B6.129P2-Cftr(tm1Unc), mutation or sibling wild-type control mice were treated with standard chow or 20 mg/kg/d rosiglitazone for 5 days. Colonic epithelial cells were extracted and mRNA isolated. Cftr-/- mice suffer from a severe intestinal phenotype including ileo-colonic obstruction resulting from the accumulation of mucin and inspissated material in the lumen.

Transcriptome changes in Streptococcus agalactiae due to deletion of both the CiaR and LiaR Transcriptional Regulators

Project description:The CiaRH and LiaFSR two-component regulatory systems in Streptococcus agalactiae (Group B Streptococcus, GBS) are essential mediators of the organism s response to biologically important sources of environmental stress, and positive regulators of GBS virulence. Transcriptional profiling of CiaR mutant GBS and LiaR mutant GBS reveals that LiaR is positively-regulated by CiaR, and the individual mutant transcriptomes share a number of commonly-regulated genes. To determine the GBS response to loss of both of these key regulatory systems, we constructed a GBS mutant strain with non-polar deletions in both ciaR and liaR, and performed transcriptional profiling using DNA microarray analysis, comparing wild-type GBS to CiaR/LiaR double mutant GBS under non-stressed conditions. Two separate RNA samples were extracted for each condition. One flip-dye replicate (2 hybridizations) was obtained for each pair of RNA samples for 4 hybridizations total.

Transcription profiling of mouse skin treated with ovalbumin to induce atopic dermatitus and then treated with a test drug or ramatroban or dexamethasone

Project description:A highly specific and potent small molecule antagonist of CRTH2 (chemoattractant receptor) was used to investigate the role of this Prostaglandin D2 (PGD2) receptor in chronic models of cutaneous inflammation and the underlying immune response. These RNA samples are from mice that were in a chronic (50 day) model of atopic dermatitis. Each mouse had a section of back skin patched with a gauze, and the RNA was from the patched area of skin; the control mice had the gauze soaked in PBS, whereas all the other test mice had the gauze soaked in ovalbumin dissolved in PBS. The ovalbumin acts as an allergen and induces the experimental atopic dermatitis. The six groups of mice that received the OVA patch were treated in different ways:some received just the drug vehicle during the OVA patching, others received the test drug (compound A) at 3 different concentrations: 10 mg/kg 1 mg/kg and 0.1 mg/kg. Two other cohorts received positive control drugs: ramatroban and dexamethasone .Each different sample represents a different animal.

LiaR Transcriptome in Streptococcus agalactiae

Project description:Streptococcus agalactiae (Group B Streptococcus, GBS) is a leading cause of early-onset neonatal bacterial infection. Evasion of innate immune defenses is critical to neonatal GBS disease pathogenesis. Effectors of the innate immune system such as antimicrobial peptides, as well as numerous antibiotics, target the peptidoglycan layer of the gram positive bacterial cell wall. The intramembrane-sensing histidine kinase class of two-component regulatory systems has recently been identified as important to the gram-positive response to cell wall stress. We identified and characterized the GBS homolog of LiaR, the response regulator component of the LiaFSR system and constructed site-directed, non-polar deletion mutations in the regulator gene liaR. GBS LiaR deletion mutant strains are more susceptible to cell wall active antibiotics (vancomycin and bacitracin) as well as antimicrobial peptides (colistin, nisin and the human cathelicidin LL-37) compared to isogenic wild-type GBS. LiaR mutant GBS are significantly attenuated in mouse models of both GBS sepsis and GBS pneumonia. To determine the genes regulated by LiaR that account for these defects, transcriptional profiling was performed using DNA microarray analysis, comparing wild-type GBS to LiaR mutant GBS under non-stressed conditions. Two separate RNA samples were extracted for each condition. One flip-dye replicate (2 hybridizations) was obtained for each pair of RNA samples for 4 hybridizations total.

To investigate whether glucose-dependent regulation of gene expression was modulated by the CsrRS system, we also compared transcriptome analysis of an isogenic Delta_csrRS mutant and of the wild type strain incubated in a glucose-rich medium.

Group sizes were five mice per group, and therefore five arrays were used per group. Whole mouse bladder collected at 24h following transurethral infection for RNA extraction and hybridization on Affymetrix microarrays. Non-pooled samples were used, with one array per mouse to expand power of expression profiles. Control mice for each time point received only PBS as a mock infection.
