Effect of progesterone in biofilm formation by Candida albicans

ABSTRACT: In this work the effect of the hormone progesterone in the ability of C. albicans SC5314 to form biofilms was investigated as well as the transcriptomic response of the yeast to this compound. The analysis was performed always comparing the genomic expression of the biofilm cells cultivated in the presence and absence of progesterone with the transcriptome of planktonic cells (in mid-exponential phase) which was used to reference the analysis.

Project description:From a clinical and molecular perspective, colon cancer (CC) is a heterogeneous disease but to date no classification based on high-density transcriptome data has been established. The aim of this study was to build up a robust molecular classification ofÂ mRNA expression profiles (Affymetrix U133Plus2) ofÂ a large series of 443 CC and to validate it on an independent serie of 123 CC and 906 public dataset.Â We identified and validated six molecular subtypes in this large cohort as a combination of multiple molecular processes that complement current disease stratification based on clinicopathological variables and molecular markers. The biological relevance of these subtypes was consolidated by significant differences in survival. These insights open new perspectives for improving prognostic models and targeted therapies. Among a large series of colon cancers collected for the Cartes d'IdentitÃ© des Tumeurs (CIT) program from the French Ligue Nationale Contre le Cancer (http://cit.ligue-cancer.net), 566 were analyzed for mRNA expression profiles using Affymetrix U133plus2 chip and, among theses, 463 could also be analyzed for DNA alteration profiles using the CGH Array (Â CIT-CGHarray V6). The 566 tumors was divided into a discovery dataset of 443 CC and a validation dataset of 123 CC.

IRF-5 and IRF-8 govern the response of Human Plasmacytoid Dendritic Cells following TLR9 ligation

Project description:Synthetic oligonucleotides (ODNs) containing CpG motifs stimulate human plasmacytoid dendritic cells (pDCs) to produce type-1 interferons (IFN) and pro-inflammatory cytokines. Previous studies demonstrated that interferon regulatory factors (IRFs) played a central role in mediating CpG-induced pDC activation. This work explores the inverse effects of IRF-5 and IRF-8 on CpG dependent gene expression. Results from RNA interference and microarray studies indicate that IRF-5 up-regulates TLR9-driven gene expression whereas IRF-8 down-regulates the same genes. Several findings support the conclusion that IRF-8 inhibits TLR9 dependent gene expression by directly blocking the activity of IRF-5. First, the inhibitory activity of IRF-8 is only observed when IRF-5 is present. Second, proximity ligation analysis shows that IRF-8 and IRF-5 co-localize within the cytoplasm of resting human pDC and co-translocate to the nucleus after CpG stimulation. Taken together, these findings suggest that two transcription factors with opposing functions control TLR9 signaling in human pDCs. CAL-1 cells were transfected with siRNA targeting IRF-5 (IRF-5si) and left unstimulated (n=4, technical repeats) or stimulated with K-type CpG ODN (n=4, technical repeats). CAL-1 cells were transfected with siRNA targeting IRF-8 (IRF-8si) and left unstimulated (n=4, technical repeats) or stimulated with K-type CpG ODN (n=4, technical repeats). CAL-1 cells were transfected with control siRNA (Contsi) and left unstimulated (n=4, technical repeats) or stimulated with K-type CpG ODN (n=4, technical repeats).

Gene expression profile induced by two different variants of street rabies virus in mice

Project description:It is well established that the pathogenicity and pathology of rabies virus (RABV) varies according to the variant, but the exact mechanism for this is still not completely known. In this study, the gene expression profile in brains of mice infected with virus isolated from a human case of dog rabies (V2) or vampire bat-acquired rabies (V3) were analyzed in experimental condition. In total, 138 array probes associated with 120 genes were differentially expressed between mice inoculated with V2 and the control mice at day 10 post-inoculation. A single probe corresponding to an unannotated gene was identified in V3 versus control mice. Gene ontology (GO) analysis revealed that all of the genes up-regulated in mice inoculated with V2 were involved in the biological process of immune defense against pathogens. Although both variants being considered pathogenic, inoculation in exactly same condition generated particular results regarding gene expression, more likely to differences in pathogenesis between dog and bat-related variants, already considered in other studies. This study was the first to demonstrate the global gene expression in experimental rabies infection due to V3 wild-type rabies virus, which reservoir is the vampire bat Desmodus rotundus, an important transmitter of rabies for humans and production animals in Latin America.

Project description:Plants have the ability to shed organs that are no longer in use. In Arabidopsis thaliana abscission of floral organs involves cell wall remodeling and cell expansion prior to cell wall dissolution. IDA encodes a secreted peptide that signals through the leucine-rich repeat receptor-like kinases (LRR-RLKs) HAESA (HAE) (At4g28490) and HASEA-LIKE2 (HSL2) (At5g65710). Arabidopsis thaliana (ecotype Colombia-0) plants were kept in growth chambers with a 16/8 h (light/dark) photoperiod at 22 M-BM-0C, and 100 mE m-2 s-1 light intensity. 4 biological replicates were prepared from each sample, each containing abscission zone regions of siliques position 4 to 8 (when counting from the flowe at anthesis at the top of the inflorescence) from plants with at least 20 siliques. Differences in transcriptional responses were measured by comparing genes expression in abscission zones of ida-2 plants (SALK_133209) against abscission zones from control plants.
