Transcriptional changes induced by fulvestrant and anastrozole treatment in ER-positive breast cancers

ABSTRACT: Analysis of breast cancer tumors following fulvestrant treatment for 4 weeks. Fulvestrant is a highly specific ER antagonist used to treat postmenopausal women with breast cancer. Data provide insight into the molecular mechanism of action of fulvestrant on whole genome expression.

Purpose
Endocrine therapies include aromatase inhibitors and the selective estrogen receptor (ER) downregulator fulvestrant. This study aimed to determine whether the reported efficacy of fulvestrant over anastrozole, and high- over low-dose fulvestrant, reflect distinct transcriptional responses.
Experimental design
Global gene expression profiles from ERα-positive breast carcinomas before and during presurgical treatment with fulvestrant (n = 22) or anastrozole (n = 81), and cor ...[more]
