Transcriptional characterisation of hemopoietic populations in GATA1s ESC-derived Megakarypoieisis


ABSTRACT: Embryonic Stem Cells expressing the BirA biotin ligase (BirA) together with a biotinylable version of the full length GATA1 (bioG1) or the truncated GATA1s (bioG1s) transcription factor were differentiated into Embryoid Bodies for 6 days. kithiCD41+ cells were FACS-sorted and grown onto OP9 cells layer for 2 days in the presence of TPO. The kithiCD41lo (P1), kitloCD41lo (P2), kitloCD41hi (P3) and kit-CD41hi (P4) subpopulations were then FACS-sorted and analysed by RNA sequencing
