Impact of Neospora caninum infection on the transcriptome of cerebrovascular endothelial cells

ABSTRACT: We also used Affymetrix array technology to investigate the global alteration in the gene expression of human brain microvascular endothelial cells (hBMECs) and rat brain microvascular endothelial cells (rBMVECs) in response to N. caninum infection at 24 hours post infection.

Transcription profiling of human primary brain microvascular endothelial cell to investigate the effect of treatment with Plasmodium falciparum infected erythrocytes

Project description:Cerebral malaria is a multifactorial condition that begins with high numbers of infected erythrocytes binding to human brain endothelium without invasion into the brain. Here we investigated the global transcriptional gene response of primary human brain endothelial cells after incubation with high numbers of infected erythrocytes; High or low parasite binding phenotypes did not alter gene response. Predominate amongst signaling pathways were the NF-kB proinflammatory response. The inflammatory pathways were validated by direct measurement of proteins. This study delineates the strong inflammatory component of human brain endothelium contributing to cerebral malaria. Experiment Overall Design: Total of 8 samples (4 control and 4 treated) were analyzed. 4 control samples included two normal RBC control and two medium controls. 4 treated samples includes 2 exposed to low binding Pf-IRBC and 2 exposed to high binding Pf-IRBC (Pf-IRBC-P). Medium and RBC controls were finally used as four replicates of control and all four Pf-IRBC or Pf-IRBC-P exposed endothelial cells were used as 4 separate treated controls.

Streptococcus suis 05ZY: control vs contact with hBMEC 3 h

Project description:The interactions of Streptocococcus suis (S.suis) with human microvascular endothelial cells (hBMEC) are important process for S.suis passage across human blood brain barrier (BBB). S.suis has evolved precise mechanisms to alter gene expression depending on the distinct challenges posed by particular disease sites. Herein, a whole-genome DNA microarray was used to investigate the change of gene expression profile of S.suis after contact with hBMEC 3 h. comparison of RNA isolated from hBMEC-associated S.suis with RNA derived from control bacteria revealed significant differential changes for 175 S.suis genes including 123 up-regulated genes and 52 down-regulated genes at 3 h post infection. A cDNA microarray imprinted with 2156 genes representing about 98% of Streptococcus suis serotype 2 genome was used for transcriptome analysis . For two-sample (reference vs. test) microarray hybridization, Two-condition experiment, control vs contact with hBMEC 3 h, four replicates (two biological replicates, two technical replicates) at each condition.

Different PfEMP1-expressing Plasmodium falciparum variants induce divergent endothelial transcriptional responses during cytoadherence

Project description:The pathogenesis of severe malaria is complex and involves several pathways that influence host inflammation and endothelial function. The human malaria parasite Plasmodium falciparum is responsible for the majority of mortality and morbidity caused by malaria infection and differs from other human malaria species in the degree of accumulation of parasite-infected red blood cells in the microvasculature, known as cytoadherence or sequestration. In P. falciparum, cytoadherence is mediated by a protein called PfEMP1 which, due to its exposure to the host immune system, undergoes antigenic variation resulting in the expression of different PfEMP1 variants on the infected erythrocyte membrane. These PfEMP1s contain various combinations of adhesive domains, which allow for the differential engagement of a repertoire of endothelial receptors on the host microvasculature, with specific receptor usage associated with severe disease. Cytoadherence results in perturbation of the micro-circulation as well as direct effects on endothelial cells promoted by receptor-mediated signalling. We used a co-culture model of cytoadherence incubating human brain microvascular endothelial cells with erythrocytes infected with two parasite lines expressing different PfEMP1s; IT4var14 (long-form; ups B) that binds strongly to human brain microvascular endothelial cells mainly via ICAM-1, and IT4var 37 (short-form; ups C) that does not bind brain endothelium but shows high levels of binding to human dermal microvascular endothelial cells via CD36. We determined the transcriptional profile of the endothelial cells following different incubation periods with infected erythrocytes, identifying different transcriptional profiles of pathways involved in the pathology of severe malaria, such as inflammation, apoptosis and barrier integrity, induced by the two PfEMP1 variants.
