MiRNA arrays on human bone marrow-derived mesenchymal stem/stromal cells

ABSTRACT: Human MSCs were cocultured for 18 hours with macrophages which had been differentiated from human peripheral blood-derived monocytes by PMA and sequentially stimulated by 2 μg/mL LPS (InvivoGen) for 3 hours and 5 mM ATP (InvivoGen) for 45 minutes. Normal MSCs without macrophage coculture and MSCs cocultured with activated macrophages were assayed by miRNA arrays.

Mast Cell Maturation is Driven via a Novel Group III Phospholipase A2-Prostaglandin D2-DP1 Receptor Paracrine Axis

Project description:Microenvironment-based alterations in mast cell phenotypes influence the susceptibility to anaphylaxis, yet the mechanisms underlying proper maturation of mast cells toward an anaphylaxis-sensitive phenotype are incompletely understood. Here we report that PLA2G3, a mammalian homolog of anaphylactic bee venom phospholipase A2, regulates this process. PLA2G3 secreted from mast cells is coupled with fibroblastic lipocalin-type PGD2 synthase (L-PGDS) to provide PGD2, which facilitates mast cell maturation via PGD2 receptor (DP1). Mice lacking PLA2G3, L-PGDS or DP1, mast cell-deficient mice reconstituted with PLA2G3- or DP1-null mast cells, or mast cells cocultured with L-PGDS-ablated fibroblasts exhibited impaired mast cell maturation and anaphylaxis. Thus, we describe a lipid-driven PLA2G3-L-PGDS-DP1 loop that drives mast cell maturation. IFN-γ-driven iNOS induction in macrophages mediates CAR T cell resistance in B cell lymphoma

Project description:Chimeric antigen receptor (CAR) T cell therapies have revolutionized B cell malignancy treatment, but subsets of patients with large B cell lymphoma (LBCL) experience primary resistance or relapse after CAR T cell treatment. To uncover tumor microenvironment (TME)-induced resistance mechanisms, we examined patients’ intratumoral immune infiltrates and observed that elevated levels of immunoregulatory macrophages in pre-infusion tumor biopsies are correlated with poor clinical responses. In murine models, CAR T cell-produced interferon-gamma (IFN-g) promotes the expression of inducible nitric oxide synthase (iNOS, NOS2) in immunoregulatory macrophages, impairing CAR T cell function. Mechanistically, proteomics analysis of CAR T cells revealed that iNOS-expressing macrophages promote the upregulation of genes mediating apoptosis and cell cycle arrest in CAR T cells, while downregulating ribosome biogenesis and protein synthesis. Furthermore, CAR T cell metabolism is compromised by the depletion of glycolytic intermediates and rewiring of the TCA cycle. Pharmacological inhibition of iNOS enhances the CAR T cell treatment efficacy in B cell tumor-bearing mice. Notably, elevated levels of iNOS+CD14+ monocytes were observed in leukaphereses of patients with non-durable response to CAR T cell therapy. These findings suggest that mitigating iNOS in tumor-associated macrophages (TAMs), potentially by modulating IFN-g expression in CAR T cells, could improve outcomes for LBCL patients.

CDK4/6 Inhibition Induces CD8+ T Cell Antitumor Immunity via MIF-Induced Functional Orchestration of Tumor-Associated Macrophages

Project description:Cyclin-dependent kinases 4 and 6 (CDK4/6) regulate cell cycle progression from the G1 to S phase. Recent findings have demonstrated that CDK4/6 inhibition (CDK4/6i) enhances antitumor immunity, as evidenced by increased tumor infiltration of CD8+ T cells, though the underlying mechanism remains unclear. Our current study reveals that CDK4/6i enhances intratumoral CD8+ T cell infiltration in breast tumors through the functional reprogramming of tumor-associated macrophages (TAMs), facilitating indirect interactions between tumor cells and CD8+ T cells. Mechanistically, CDK4/6i enhances the proliferation and activation of M1 macrophages and promote the polarization of M2 to M1 macrophages via the macrophage migration inhibitory factor (MIF)-CD44/CD74 axis between tumor cells and macrophages. CDK4/6i-trained M1 TAMs increase and activate CD8+ T cells through MHC-I antigen presentation machinery. Inhibition of MIF in tumor cells or loss of MIF reverses the immunostimulatory effects of CDK4/6i on macrophages and subsequent CD8+ T cell antitumor immunity. Therefore, CDK4/6i-trained M1 TAM supernatant therapy surmounts the immunosuppressive tumor microenvironment and invokes a tumor response to low-dose PD-1 immune checkpoint blockade therapy in breast cancers.
