Stranded Total RNA-seq of whole blood cells collected from Holstein Friesian cows

ABSTRACT: Bovine mastitis is an inflammatory disease of the mammary gland with serious economic implications for dairy industries worldwide. We performed total RNA sequencing using whole blood cells collected from multiparous Holstein Friesian dairy cows with naturally occurring mastitis to investigate the changes in systemic gene expression and their association with inflammatory responses. Some related sequencing data are deposited in E-MTAB-9347 and E-MTAB-9348.

Project description:Homeorhetic mechanisms assist dairy cows to transition from pregnancy to lactation. In some cows this is less successful and they develop severe negative energy balance, placing them at risk of metabolic and infectious disease and reduced fertility. We placed Holstein Friesian cows into metabolically BALANCED or IMBALANCED clusters using as biomarkers measurements of plasma nonesterified fatty acids, β-hydroxybutyrate, glucose and IGF-I collected at 14 and 35 day in milk (DIM). To determine underlying mechanisms associated with their metabolic and immune dysfunction, we performed stranded total RNA sequencing using whole blood cells collected from some of the animals.

Project description:Supplementation of a Saccharomyces cerevisiae fermentation product modulates dairy cows health by reducing incidence and severity of mastitis, one of the most common and economically important diseases of the dairy industry. However, mechanisms remain unclear. We conducted a comprehensive molecular analysis, along with physiological data, on dairy cows supplemented for 45 days with NutriTek, a commercially available S. cerevisiae fermentation product, and then subjected to a mastitis challenge . Preliminary cell culture models for subclinical mastitis in sheep

Project description:Ovine mastitis is defined as the inflammation of the sheep udder, most commonly caused in response to intramammary infections. Based on the occurrence of clinical signs, mastitis is characterized as either clinical or subclinical (SCM). The impact of ovine SCM on the overall sustainability of dairy sheep farms has been documented underpinning the significance of efficient diagnosis. Although SCM can be detected in cows, the performance and the validity of the methods used do not transfer in dairy sheep. This fact challenges the development of evidence-based ovine udder health management protocols and renders the detection and control of ovine mastitis rather problematic. Currently, cell culture-based models are being successfully used in biomedical studies and have also been effectively used in the case of bovine mastitis. The objective of the present study was to culture ovine primary mammary cells for the development of 2D and 3D cell culture-based models for the study of ovine SCM. Cells were infected by mastitis-inducing pathogens mimicking the pathogenesis of SCM as derived by natural intramammary infections. The secreted proteins were subjected to mass-spectrometry resulting in the identification of 79 distinct proteins. Among those, several had already been identified in healthy or mastitic milk, while others were detected for the first time in the ovine mammary secretome. The development of cell-based models for the early detection and the overall study of SCM has the potential to be applicable and beneficial for the udder health management in dairy sheep.

Gene expression profiling of human whole-blood samples before and after the administration of recombinant human erythropoietin

Project description:As part of cross-platform comparisons of microarray and RNA-seq, this current experiment using the Affymetrix Human Transcriptome Array 2.0 aimed to quantify gene-level expression in subjects administered recombinant human erythropoietin over a 10-week protocol for the identification of gene signatures of blood doping. These results were compared to results obtained from other gene expression quantification platforms using the same experimental cohort, including the Illumina HumanHT-12v4 Expression BeadChips (archived in ArrayExpression; E-MTAB-2874), Illumina NextSeq 500 and MGI DNBSEQ-G400RS.

Expression of exosomal microRNAs in bovine serum with clinical or subclinical mastitis

Project description:Bovine mastitis causes changes in the serum exosomal miRNAs expression. Serum samples from healthy dairy cows (n = 7) were compared to those of cows with subclinical (n = 7 ) using small RAN sequencing. Three hundred fifty-five miRNAs (341 known and 14 novel ones) were identified. There were 42 miRNAs up-regulated in serum-derived EVs from cows with subclinical mastitis, including bta-miR-1246, bta-miR-2431-3p, bta-miR-126-3p, bta-miR-29a, etc. The MAPK signaling pathway was the most affected pathway by clinical mastitis. Thus, miRNA alterations in mastitis serum-derived EVs support the potential regulator role of specific miRNAs as exosomal cargo in clinical mastitis physiology.

Response of bovine mammary tissue to experimental Staphylococcus aureus infection experiment 3.

Project description:Mastitis in dairy cattle can result from infection by a range of microorganisms but is principally caused by coliform bacteria and gram positive bacteria such as Staphylococcus aureus (S. aureus). The former species are often acquired by environmental contamination while S. aureus is particularly problematic due to its resistance to antibiotic treatments and ability to reside within mammary tissue in a chronic, subclinical state. The transcriptional and translational responses within bovine mammary epithelial tissue subjected to intramammary challenge with S. aureus are poorly characterised, particularly at the earliest stages of infection. A Bovine Innate Immune Microarray was employed to measure changes in gene expression occurring in bovine mammary tissues sampled from three dairy cows after a brief and graded intramammary challenge with a virulent strain of S. aureus. Keywords: dose response, disease state analysis Animal infected with vaying doses of S. aureus in different udder locations were compared to a single control uninfected tissue and to the other 2 locations at each infective dose. Dye swaps were performed for each comparison.
