Transcription profiling of Dictyostelium discoideum strains lacking the DimB or MybE transcriptions factors in response to treatment with the polyketide DIF-1


ABSTRACT: The prototypic prestalk-specific gene ecmA is inducible by the polyketide DIF-1. The DimB and MybE transcription factors bind to sites in the ecmA promoter and DIF-1 induction of ecmA in a monolayer assay is greatly diminished in null strains for either of the two factors. We perform genome-wide micro-array analyses, on parental (Ax2), mybE- and dimB- cells, and identify many additional genes that depend on MybE and DimB for their DIF-1 inducibility. Surprisingly, an even larger number of genes are only DIF-inducible in mybE- cells, some genes are only inducible in DimB- cells and some are inducible when either transcription factor is absent. Thus in monolayer assay, where MybE and DimB function as inducers of ecmA, many other genes fall under negative control by the same two transcription factors. There is presumably therefore a partially or wholly alternate DIF-1 activation pathway for the latter genes.
