Microarray of isolated adipocytes from subcutaneous adipose tissue of severely obese patients compared to lean controls

Project description:The aim of the project was to compare global gene expression in adipocytes from obese patients and lean controls. Subcutaneous adipose tissue was collected from severely obese patients undergoing bariatric surgery (average body-mass index (BMI) of 45.5 kg/m2 (n = 12, thereof 4 men) and healthy lean patients undergoing hernia repairs (average BMI of 24.2 kg/m2 (n = 12, thereof 7 men), between 27 and 56 years of age. Adipocytes were isolated by collagenase treatment of adipose tissue, followed by filtering and centrifugation. Floating adipocytes were lysed in Qiazol before RNA purification and microarray analysis.

miRNA profiling by array of human paired adipose tissue (subcutaneous and omentum) before and after gastric-bypass and weight loss

Project description:Objective: Adipose tissue plays a key role in obesity related metabolic dysfunction. MicroRNA (miRNA) are gene regulatory molecules involved in inter-cellular and inter-organ communication. We hypothesised that miRNA levels in adipose tissue would change after gastric bypass surgery and that this would provide insights into their role in obesity-induced metabolic dysregulation. Methods: miRNA-profiling (Affymetrix_Gene-Chip_miRNA2.0_Arrays) of omental and subcutaneous adipose (n=15 females) before, and after, gastric bypass surgery. Results: One omental, and thirteen subcutaneous adipose miRNAs were significantly, differentially expressed after gastric bypass, including down-regulation of miR-223-3p and its antisense relative, miR-223-5p, in both adipose tissues. mRNA levels of miR-223-3p targets NLRP3 and GLUT4 were increased and decreased respectively following gastric bypass in both adipose tissues. Significantly more NLRP3 protein was observed in omental adipose after gastric bypass (P=0.02). Significant hypomethlyation of NLRP3 and hypermethylation of miR-223 was observed in both adipose tissues after gastric bypass. In subcutaneous adipose significant correlations were observed between both miR-223-3p and miR-223-5p and glucose, and between NLRP3 mRNA and protein levels and blood lipids. Conclusions: This is the first report detailing genome-wide miRNA-profiling of omental adipose before and after gastric bypass, and further highlights a link between miR-223-3p and the NLRP3 inflammasome in obesity.

Adipose Tissue MicroRNAs as Regulators of CCL2 Production in Human Obesity (differentiation data)

Project description:We used an unbiased systems biology approach to study the regulation of gene expression in human adipose tissue focusing on inflammation. We show that microRNAs play a major role as regulators of CCL2 production in obesity. Subcutaneous adipose tissue was obtained from healthy subjects undergoing cosmetic liposuction (n=12). There was no selection for age, sex or BMI. From the tissue, we isolated preadipocytes and in vitro differentiated them to adipocytes. The cells were lysed at day 4/5 (early), 8 (middle) and 12 (late) of differentiation. From the samples, we prepared and hybridised biotinylated complementary RNA to GeneChip Human Gene 1.0 ST Arrays (Affymetrix Inc., Santa Clara, CA). Pre-processing was performed using the Affymetrix Expression Console version 1.1 and the following settings: Summarization: PLIER; Background Correction: PM-GCBG; Normalization: Global Median.
