Investigation on Centrifugally Spun Fibrous PCL/3-Methyl Mannoside Mats for Wound Healing Application.

ABSTRACT: Fibrous structures, in general, have splendid advantages in different forms of micro- and nanomembranes in various fields, including tissue engineering, filtration, clothing, energy storage, etc. In the present work, we develop a fibrous mat by blending the bioactive extract of Cassia auriculata (CA) with polycaprolactone (PCL) using the centrifugal spinning (c-spinning) technique for tissue-engineered implantable material and wound dressing applications. The fibrous mats were developed at a centrifugal speed of 3500 rpm. The PCL concentration for centrifugal spinning with CA extract was optimized at 15% w/v of PCL to achieve better fiber formation. Increasing the extract concentration by more than 2% resulted in crimping of fibers with irregular morphology. The development of fibrous mats using a dual solvent combination resulted in fine pores on the fiber structure. Scanning electron microscope (SEM) images showed that the surface morphology of the fibers in the produced fiber mats (PCL and PCL-CA) was highly porous. Gas chromatography-mass spectrometry (GC-MS) analysis revealed that the CA extract contained 3-methyl mannoside as the predominant component. The in vitro cell line studies using NIH3T3 fibroblasts demonstrated that the CA-PCL nanofiber mat was highly biocompatible, supporting cell proliferation. Hence, we conclude that the c-spun, CA-incorporating nanofiber mat can be employed as a tissue-engineered construct for wound healing applications.

Project description:Development of natural protein-based fibrous scaffolds with tunable physical properties and biocompatibility is highly desirable to construct three-dimensional (3D), fully cellularized scaffolds for wound healing. Herein, we demonstrated a simple and effective technique to construct electrospun 3D fibrous scaffolds for accelerated wound healing using a photocrosslinkable hydrogel based on gelatin methacryloyl (GelMA). We found that the physical properties of the photocrosslinkable hydrogel including water retention, stiffness, strength, elasticity and degradation can be tailored by changing the light exposure time. We further observed that the optimized hydrogel fibrous scaffolds which were soft and elastic could support cell adhesion, proliferation and migration into the whole scaffolds, facilitating regeneration and formation of cutaneous tissues within two weeks. Such tunable characteristics of the fibrous GelMA scaffolds distinguished them from other reported substrates developed for reconstruction of wound defects including glutaraldehyde-crosslinked gelatin or poly (lactic-co-glycolic acid) (PLGA), whose physical and chemical properties were difficult to modify to allow cell infiltration into the 3D scaffolds for tissue regeneration. We anticipate that the ability to become fully cellularized will make the engineered GelMA fibrous scaffolds suitable for widespread applications as skin substitutes or wound dressings.Statement of significanceIn present study, we generate three-dimensional photocrosslinkable gelatin (GelMA)-based fibrous scaffolds with tunable physical and biological properties by using a combined photocrosslinking/electrospinning approach. The developed GelMA fibrous scaffolds can not only support cell viability and cell adhesion, but also facilitate cell migration and proliferation, accelerating regeneration and formation of cutaneous tissues. In addition, the physical properties of the engineered fibrous GelMA hydrogel including water retention capability, mechanical properties and biodegradability can be tuned to accommodate different patients' needs, making it a promising candidate for skin tissue engineering.

Project description:Liposomes are employed for the delivery of molecular cargo in several classes of systems. For instance, the embedding of loaded liposomes in polymeric fibrous scaffolds has enabled the creation of hybrid materials that mimic biological membranes. Liposomes with unmodified surfaces have been predominantly integrated into fibers, which leads to instabilities due to interfacial incompatibility. In addition, electrospinning has been almost exclusively employed for fiber fabrication, which limits the potential for scale-up production. Here, we present the fabrication of hybrid biomimetic materials by fusing polymer-coated liposomes to force-spun microfibers to increase the stability of the hybrid materials and enhance the sustained release of the cargo. l-α-Phosphatidylcholine liposomes were coated with chitosan or polyethylene glycol (PEG). The nano-differential scanning calorimetry results confirm that polymer coating does not affect the phase transition temperature (T m) of the liposomes, where only the model drug, quercetin, reduced T m. Centrifugal spinning was employed to fabricate hydrophobic polycaprolactone (PCL) microfibers at various polymer concentrations and using various solvents and spinning parameters to increase the yield at the lowest fiber diameter. The highest microfiber production rate obtained occurred at a 20% (w/v) PCL concentration in 50 : 50 (v/v) chloroform and methanol solution with an average fiber diameter of 584.85 ± 26.30 nm. The non-chemical fusion of the polymer-coated liposomes and the fibrous scaffolds was promoted by immersion at T > T m, under ultrasonication. We hypothesize that the fusion is driven by hydrophobic interactions between the liposomes and the fibers, which merge the materials through the lipid bilayer. The fused hybrid material solved the burst release problem observed when adhering plain liposomes to nanofibers. Both PEG and chitosan yielded a sustained release, where the release rate with the former was faster. These results demonstrate that the fusion of polymer-coated liposomes and microfibers enables more effective blending of the loaded carriers into the polymer microfibers. Ultimately, the fused liposome/microfiber hybrids are stable matrices and enhance the sustained release of molecular cargo.
