Project description:Because of severe abiotic limitations, Antarctic soils represent simplified ecosystems, where microorganisms are the principle drivers of nutrient cycling. This relative simplicity makes these ecosystems particularly vulnerable to perturbations, like global warming, and the Antarctic Peninsula is among the most rapidly warming regions on the planet. However, the consequences of the ongoing warming of Antarctica on microorganisms and the processes they mediate are unknown. Here, using 16S rRNA gene pyrosequencing and qPCR, we report a number of highly consistent changes in microbial community structure and abundance across very disparate sub-Antarctic and Antarctic environments following three years of experimental field warming (+ 0.5-2°C). Specifically, we found significant increases in the abundance of fungi and bacteria and in the Alphaproteobacteria-to-Acidobacteria ratio. These alterations were linked to a significant increase in soil respiration. Furthermore, the shifts toward generalist or opportunistic bacterial communities following warming weakened the linkage between bacterial diversity and functional diversity. Warming also increased the abundance of some organisms related to the N-cycle, detected as an increase in the relative abundance of nitrogenase genes via GeoChip microarray analyses. Our results demonstrate that soil microorganisms across a range of sub-Antarctic and Antarctic environments can respond consistently and rapidly to increasing temperatures, thereby potentially disrupting soil functioning.

Project description:De facto marine protected areas (DFMPAs) are regions of the ocean where human activity is restricted for reasons other than conservation. Although DFMPAs are widespread globally, their potential role in the protection of marine habitats, species, and ecosystems has not been well studied. In 2012 and 2013, we conducted remotely operated vehicle (ROV) surveys of marine communities at a military DFMPA closed to all civilian access since 2010 and an adjacent fished reference site at San Clemente Island, the southernmost of California's Channel Islands. We used data extracted from ROV imagery to compare density and biomass of focal species, as well as biodiversity and community composition, between the two sites. Generalized linear modeling indicated that both density and biomass of California sheephead (Semicossyphus pulcher) were significantly higher inside the DFMPA. Biomass of ocean whitefish (Caulolatilus princeps) was also significantly higher inside the DFMPA. However, species richness and Shannon-Weaver diversity were not significantly higher inside the DFMPA, and overall fish community composition did not differ significantly between sites. Demonstrable differences between the DFMPA and fished site for two highly sought-after species hint at early potential benefits of protection, though the lack of differences in the broader community suggests that a longer trajectory of recovery may be required for other species. A more comprehensive understanding of the potential conservation benefits of DFMPAs is important in the context of marine spatial planning and global marine conservation objectives.

Project description:Glacial retreat is one of the most conspicuous signs of warming in Antarctic regions. Glacier soils harbor an active microbial community of decomposers, and under the continuous retraction of glaciers, the soil starts to present a gradient of physical, chemical, and biological factors reflecting regional changes over time. Little is known about the biological nature of fungi in Antarctic glacier soils. In this sense, this work aimed at studying the behavior of fungal community structure from samples of glacier soil collected after glacial retreat (Collins Glacier). A total of 309 fungi distributed in 19 genera were obtained from eleven soil samples. Representatives of the genera Pseudogymnoascus (Ascomycota) and Mortierella (Mortierellomycota) were the most abundant isolates in all samples. The data revealed the presence of filamentous fungi belonging to the phylum Basidiomycota, rarely found in Antarctica. Analysis of the generalized linear models revealed that the distance from the glacier as well as phosphorus and clay were able to modify the distribution of fungal species. Environmental variations proved to have influenced the genera Pseudogymnoascus and Pseudeutorium.

Project description:Understanding how microbial communities survive in extreme environmental pressure is critical for interpreting ecological patterns and microbial diversity. Great Gobi A Strictly Protected Area represents an intriguing model for studying the bacterial community since it is a protected and intact wild area of the Mongolian desert. In this work, the composition of a bacterial community of the soil from four oases was characterized by extracting total DNA and sequencing through the Illumina NovaSeq platform. In addition, the soil's chemical and physical properties were determined, and their influence on shaping the microbial communities was evaluated. The results showed a high variability of bacterial composition among oases. Moreover, combining specific chemical and physical parameters significantly shapes the bacterial community among oases. Data obtained suggested that the oases were highly variable in physiochemical parameters and bacterial communities despite the similar extreme climate conditions. Moreover, core functional microbiome were constituted by aerobic chemoheterotrophy and chemoheterotrophy, mainly contributed by the most abundant bacteria, such as Actinobacteriota, Pseudomonadota, and Firmicutes. This result supposes a metabolic flexibility for sustaining life in deserts. Furthermore, as the inhabitants of the extreme regions are likely to produce new chemical compounds, isolation of key taxa is thus encouraged.

We conducted in situ warming experiments for three years using open-top chambers (OTCs) at one sub-Antarctic (Falkland Islands, 52ºS) and two Antarctic locations (Signy and Anchorage Islands, 60ºS and 67ºS respectively) (see Supplementary Fig. 1 for a map). OTCs increased annual soil temperature by an average of 0.8°C (at a depth of 5 cm), resulting in 8-43% increase in positive-degree days annually and a decrease in freeze-thaw cycle frequency by an average of 15 cycles per year (8). At each location, we included densely vegetated and bare fell-field soils in the experimental design for a total of six environments. Densely vegetated and bare environments represent two contrasting environments for Antarctic soil microorganisms, with large differences in terms of C and N inputs to soils. Massively parallel pyrosequencing (Roche 454 GS FLX Titanium) of 16S rRNA gene amplicons was used to follow bacterial diversity and community composition [GenBank Accession Numbers: HM641909-HM744649], and functional gene microarrays (GeoChip 2.0)(11) were used to assess changes in functional gene distribution. Bacterial and fungal communities were also quantified using real-time PCR.

Project description:BackgroundIn the COVID-19 pandemic, in Switzerland people aged 65+ years were categorized as a high-risk group, as they are at higher risk of severe COVID-19 due to higher rates of underlying health conditions. This focused attention on older adults, mostly with respect to frailty and helplessness. What is lacking is older adults' subjective views, including their state of mind and their experiences.ObjectiveThis study explores the subjective view of older adults and aims to examine their everyday coping during the pandemic, particularly the concrete impacts on their lives, their self-assessment as a vulnerable group, and fears about COVID-19.Material and methodsThe longitudinal study was conducted from April to June 2020 using a mixed methods design comprising both closed and open-ended questions. We conducted guided biweekly interviews by telephone with 40 people (22 were women) aged 65-90 years. The quantitative data included descriptive analyses. The qualitative data were evaluated by structured content analysis with inductive category formation.ResultsThe respondents reported little change in their everyday lives. Overall, a good mood was reported. Social restrictions were experienced as a great burden. The respondents perceived their classification as a risk group as undifferentiated and arbitrary. Fear or worry about contracting the coronavirus was low.DiscussionRespondents appear to have coped much better with the crisis than public discourse suggested. Activities and routines as well as support services may have been protective as strategies in everyday life. The homogeneous classification of older adults as an at-risk group ignores their resources and promotes both negative stereotypes and ageism.

Project description:Soil samples from a remote Japanese island (Kozushima) were processed and investigated for organisms exhibiting antimicrobial activity against pathogenic strains. A Pseudomonas strain demonstrating antimicrobial activity against Staphylococcus aureus (S. aureus) was identified, prompting further investigation. Whole-genome sequencing was employed to identify the species and conduct phylogenetic analysis, followed by in silico molecular analysis. Chemotaxonomic and biochemical analyses were conducted to further characterize the strain. Genomic analysis identified the strain of interest as Pseudomonas batumici (P. batumici), originally isolated from soil of the Black Sea coast of the Caucasus in 1980. P. batumici Koz11 is the second P. batumici strain to be isolated and identified outside its initial area of discovery. Similar to the type strain, P. batumici Koz11 showed antimicrobial activity against various S. aureus strains, including methicillin-resistant S. aureus (MRSA) and vancomycin-resistant S. aureus (VRSA). However, the previously reported 'batumin gene cluster', which synthesizes antimicrobial compounds, was absent from P. batumici Koz11. This study provides new insights into P. batumici. Since the type strain of P. batumici is exclusively deposited in the Ukrainian Collection of Microorganisms, the Koz11 strain may serve as a surrogate to facilitate continued study of P. batumici.

Project description:Locally-established marine protected areas (MPAs) have been proven to achieve local-scale fisheries and conservation objectives. However, since many of these MPAs were not designed to form ecologically-connected networks, their contributions to broader-scale goals such as complementarity and connectivity can be limited. In contrast, integrated networks of MPAs designed with systematic conservation planning are assumed to be more effective--ecologically, socially, and economically--than collections of locally-established MPAs. There is, however, little empirical evidence that clearly demonstrates the supposed advantages of systematic MPA networks. A key reason is the poor record of implementation of systematic plans attributable to lack of local buy-in. An intermediate scenario for the expansion of MPAs is scaling up of local decisions, whereby locally-driven MPA initiatives are coordinated through collaborative partnerships among local governments and their communities. Coordination has the potential to extend the benefits of individual MPAs and perhaps to approach the potential benefits offered by systematic MPA networks. We evaluated the benefits of scaling up local MPAs to form networks by simulating seven expansion scenarios for MPAs in the Verde Island Passage, central Philippines. The scenarios were: uncoordinated community-based establishment of MPAs; two scenarios reflecting different levels of coordinated MPA expansion through collaborative partnerships; and four scenarios guided by systematic conservation planning with different contexts for governance. For each scenario, we measured benefits through time in terms of achievement of objectives for representation of marine habitats. We found that: in any governance context, systematic networks were more efficient than non-systematic ones; systematic networks were more efficient in broader governance contexts; and, contrary to expectations but with caveats, the uncoordinated scenario was slightly more efficient than the coordinated scenarios. Overall, however, coordinated MPA networks have the potential to be more efficient than the uncoordinated ones, especially when coordinated planning uses systematic methods.
