Project description:A novel hydrosoluble near-infrared fluorescence off-on probe has been developed for detecting carboxylesterase activity. The probe was designed by introducing (4-acetoxybenzyl)oxy as a quenching and recognizing moiety to the decomposed product of IR-783, which exhibits excellent near-infrared fluorescence features and good water solubility. The responding mechanism of novel probe 1 to carboxylesterase was investigated. It would lead to the cleavage of the carboxylic ester bond by carboxylesterase catalyze the spontaneous hydrolysis of the probe, resulting in the release of a near-infrared fluorophore. This behaviour leads to the development of a simple and sensitive fluorescent method for assaying carboxylesterase activity, with a detection limit of 3.4 × 10-3 U mL-1. Moreover, the probe displays excellent selectivity toward carboxylesterase over other substances. Notably, the imaging experimental results showed that the probe 1 is cell membrane permeable, and its applicability has been demonstrated for monitoring carboxylesterase activity in HeLa cells.

Project description:Glutathione (GSH), an abundant non-protein thiol, plays a crucial role in numerous biotic processes. Herein, a mitochondria-targeted near-infrared GSH probe (JGP) was synthesized, which displayed desired properties with high specificity and sensitivity, appreciable water solubility, and rapid response time. In the presence of GSH, nearly a 13-fold fluorescence emission growth appeared at 730 nm and the solvent color changed from blue to cyan. The sensing mechanism of JGP and GSH was confirmed by a high-resolution mass spectroscopy analysis. Moreover, good cell penetration enabled JGP to be successfully used for imaging biological samples such as HeLa cells, C. elegans, and especially rat brain slices. Imaging experiments showed that JGP could monitor the GSH concentration changes with a dose-dependent direct ratio in all the tested samples. The successful application of JGP in brain imaging indicates that JGP is a suitable GSH optical probe, which may have wide application value in fields of brain imaging. It also lays a theoretical and practical foundation for the further application of fluorescent probes in brain sciences.

Project description:Aging and oxidative stress are two prominent pathological mechanisms for Parkinson's disease (PD) that are strongly associated with the degeneration of dopamine (DA) neurons in the midbrain. DA and other catechols readily oxidize into highly reactive o-quinone species that are precursors of neuromelanin (NM) pigment and under pathological conditions can modify and damage macromolecules. The role of DA oxidation in PD pathogenesis remains unclear in part due to the lack of appropriate disease models and the absence of a simple method for the quantification of DA-derived oxidants. Here, we describe a rapid, simple, and reproducible method for the quantification of o-quinones in cells and tissues that relies on the near-infrared fluorescent properties of these species. Importantly, we demonstrate that catechol-derived oxidants can be quantified in human neuroblastoma cells and midbrain dopamine neurons derived from induced pluripotent stem cells, providing a novel model to study the downstream actions of o-quinones. This method should facilitate further study of oxidative stress and DA oxidation in PD and related diseases that affect the dopaminergic system.

Project description:Real-time endoscopic rectal lesion characterization employing artificial intelligence (AI) and near-infrared (NIR) imaging of the fluorescence perfusion indicator agent Indocyanine Green (ICG) has demonstrated promise. However, commercially available fluorescence endoscopes do not possess the flexibility and anatomical reach capabilities of colonoscopy while commercial flexible scopes do not yet provide beyond visible spectral imaging. This limits the application of this AI-NIR classification technology. Here, to close this technical gap, we present our development of a colonoscope-compatible flexible imaging probe for NIR-ICG visualization combined with a full field of view machine learning (ML) algorithm for fluorescence quantification and perfusion pattern cross-correlation (including first in human testing). The imaging probe is capable of 133µm minimum object resolution, with a maximum working distance of 50mm and an excitation illumination power of 52mW with 75o average field of illumination (meaning minimum device tip distance from target is 13 mm for a 2 cm polyp). The system demonstrated ex-vivo and in-vivo NIR visualization of clinically relevant concentrations of ICG in both resected and in situ (extracorporeally) colon in patients undergoing colorectal resection. A previously developed AI-NIR perfusion quantification algorithm was applied to videos of a bench model of varying ICG flow captured with the developed flexible system with added ML features generated full field of view pixel-level fluorescence time-series measurements capable of distinguishing distinct ICG flow regions in the image via correlative dynamic fluorescence intensity profiles. Jaccard Index comparison of the AI -generated flow regions against manually delineated flow regions resulted in 79% accuracy. While further clinical validation of the AI-NIR polyp classification method is on-going (in the Horizon Europe Awarded CLASSICA project), other use case applications of NIR colonoscopy include simpler perioperative perfusion assessment in patients undergoing colorectal resection and combination with targeted agents in development thus encouraging continuing development and design optimization of this flexible NIR imaging probe to enable clinical and commercial translation.

Project description:Near-infrared (NIR) light possesses many suitable optophysical properties for medical imaging including low autofluorescence, deep tissue penetration, and minimal light scattering, which together allow for high-resolution imaging of biological tissue. NIR imaging has proven to be a noninvasive and effective real-time imaging methodology that provides a high signal-to-background ratio compared to other potential optical imaging modalities. In response to this, the use of NIR imaging has been extensively explored in the field of immunotherapy. To date, NIR fluorescence imaging has successfully offered reliable monitoring of the localization, dynamics, and function of immune responses, which are vital in assessing not only the efficacy but also the safety of treatments to design immunotherapies optimally. This review aims to provide an overview of the current research on NIR imaging of the immune response. We expect that the use of NIR imaging will expand further in response to the recent success in cancer immunotherapy. We will also offer our insights on how this technology will meet rapidly growing expectations in the future.

Project description:Endometriosis surgery is associated with a high risk of reoperation due to an insufficient recognition of endometriotic lesions. Our aim was to explore the role of near-infrared fluorescence (NIRF) imaging for the visualization and identification of endometriotic lesions next to conventional white light (WL) laparoscopy. Fifteen women scheduled for diagnostic laparoscopy in whom peritoneal endometriosis was suspected were included. Peritoneal exploration was performed in WL, followed by NIRF imaging after ICG administration. Biopsies of all the suspected lesions were taken for histological examination. Subjective evaluations of the equipment and NIRF imaging were also performed. Only 61% (44) of the biopsied lesions contained endometriosis. The positive predictive value (PPV) for the lesions found in WL was 64%. The PPV for the lesions found under NIRF was 69% and the PPV for the lesions found in both modes was 61%. The mean satisfaction of surgeons regarding the surgical procedure and equipment using both imaging modalities was 6.5 (p > 0.05) on a 10 item Likert scale and the mean satisfaction with the quality of the NIRF imaging was 7.4 (p > 0.05). In this study, the additional value of NIRF imaging, although feasible, was found to be limited for the intraoperative detection of endometriotic lesions.

Project description:Systemically injected drug delivery systems distribute into various organs and tissues, including liver, spleen and kidneys. Recent reports pointed out a significant accumulation of systemically injected nanoparticles in the skin. Skin constitutes the largest organ in the body with important immune functions, and accumulation of drug delivery systems could have significant implications for skin toxicity in living subjects. Fiber optic-based near-infrared spectroscopy (FONIRS) setup was developed and tested for measuring of NIR (760nm excitation) emission spectra in the skin. Ex vivo spectral measurements of NIR fluorescence through the skin showed linear response down to 34 femtomole of dye DiR. Following systemic injection of IRDye 800 labeled 500kDa dextran, FONIRS detected an immediate and stable accumulation of fluorescence in the skin. Longitudinal monitoring of skin accumulation and elimination of IRDye 800-labeled therapeutic anti-epidermal growth factor antibody (cetuximab) showed significant signal in the skin after the antibody cleared from circulation. Comparison of skin accumulation of DiR labeled, long-circulating PEGylated liposomes with short-circulating non-PEGylated liposomes showed much higher accumulation of PEGylated liposomes that persisted several days after the liposomes cleared from blood. Measurements with FONIRS enabled to estimate skin concentration of liposomes (percent of injected dose per gram). This simple and practical approach can be used to monitor accumulation of drug delivery systems in preclinical and clinical studies.

Project description:We report what we believe to be the first near-infrared pH-sensitive fluorescence lifetime molecular probe suitable for biological applications in physiological range. Specifically, we modified a known fluorophore skeleton, hexamethylindotricarbocyanine, with a tertiary amine functionality that was electronically coupled to the fluorophore, to generate a pH-sensitive probe. The pK(a) of the probe depended critically on the location of the amine. Peripheral substitution at the 5-position of the indole ring resulted in a compound with pK(a) ? 4.9 as determined by emission spectroscopy. In contrast, substitution at the meso-position shifted the pK(a) to 5.5. The resulting compound, LS482, demonstrated steady-state and fluorescence-lifetime pH-sensitivity. This sensitivity stemmed from distinct lifetimes for protonated (?1.16 ns in acidic DMSO) and deprotonated (?1.4 ns in basic DMSO) components. The suitability of the fluorescent dyes for biological applications was demonstrated with a fluorescence-lifetime tomography system. The ability to interrogate cellular processes and subsequently translate the findings in living organisms further augments the potential of these lifetime-based pH probes.

Project description:BackgroundLymphangiomatosis is a rare disorder of the lymphatic system that can impact the dermis, soft tissue, bone, and viscera and can be characterized by lymphangiomas, swelling, and chylous discharge. Whether disordered lymphangiogenesis in lymphangiomatosis affects the function and anatomy of the entire systemic lymphatic circulation or is localized to specific sites is not fully known.Methods and resultsA 35-year-old Caucasian female diagnosed with whole-body lymphangiomatosis at 2 months of age and who continues to present with progressive disease was imaged with near-infrared fluorescence lymphatic imaging. While the peripheral lymphatics in the extremities appeared largely normal compared to prior studies, we observed tortuous lymphatic vessels, fluorescence drainage from the peripheral lymphatics into lymphangiomas, and extensive dermal lymphatics in the left thigh and inguinal regions where the subject had previously had surgical assaults, potentially indicating defective systemic lymphangiogenesis.ConclusionsFurther research into anatomical and functional lymphatic changes associated with the progression and treatment of lymphangiomatosis could aid in understanding the pathophysiology of the disease as well as point to treatment strategies.

Project description:Accurate detection of H2S is crucial to understanding the occurrence and development of H2S-related diseases. However, the accurate and sensitive detection of H2S in vivo still faces great challenges due to the characteristics of H2S diffusion and short half-life. Herein, we report a H2S-activatable ratiometric near-infrared (NIR) fluorescence liposome nanoprobe HS-CG by the thin-film hydration method. HS-CG shows "always on" fluorescence signal at 816 nm and low fluorescence signal at 728 nm; the NIR fluorescence ratio between 728 and 816 nm (F728/F816) is low. Upon reaction with H2S, the fluorescence at 728 nm could be more rapidly turned on due to strong electrostatic interaction between enriched HS- and positively charged 1,2-dihexadecanoyl-sn-glycero-3-phosphocholine (DPPC) doped in the liposome nanoprobe HS-CG, resulting in a large enhancement of F728/F816, which allows for sensitive visualization of the tumor H2S levels in vivo. This study demonstrates that this strategy of electrostatic adsorption between HS- and positively charged molecules provides a new way to enhance the reaction rate of the probe and H2S, thus serving as an effective platform for improving the sensitivity of imaging.
