Helical fluxionality: numerical frustration drives concerted low-barrier screw motions of a tricopper cluster.

ABSTRACT: Uneven allocation of resources creates frustration, tension, and conflicts. Confronted with an apparent mismatch between the number of donor atoms and the number of metal atoms to be supported, helically twisted ligands cleverly come up with a sustainable symbiotic solution. As an example, we present a tricopper metallohelicate exhibiting screw motions for intramolecular site exchange. A combination of X-ray crystallographic and solution NMR spectroscopic studies revealed thermo-neutral site exchange of three metal centres hopping back and forth inside the helical cavity lined by a spiral staircase-like arrangement of ligand donor atoms. This hitherto unknown helical fluxionality is a superimposition of translational and rotational movements of molecular actuation, taking the shortest path with an extraordinarily low energy barrier without compromising the overall structural integrity of the metal-ligand assembly.

Project description:Highly strained cage hydrocarbons have long stood as fundamental molecules to explore the limits of chemical stability and reactivity, probe physical properties, and more recently as bioactive molecules and in materials discovery. Interestingly, the nitrogenous congeners have attracted much less attention. Previously absent from the literature, azahomocubanes, offer an opportunity to investigate the effects of a nitrogen atom when incorporated into a highly constrained polycyclic environment. Herein disclosed is the synthesis of 1-azahomocubane, accompanied by comprehensive structural characterization, physical property analysis and chemical reactivity. These data support the conclusion that nitrogen is remarkably well tolerated in a highly strained environment. 1-Azahomocubane has been prepared 56 years after the parent hydrocarbon. Introduction of a nitrogen atom into this constrained polycyclic environment resulted in minimal changes to the framework geometry, with s-character of the nitrogen lone pair increasing due to strain.

Project description:Antibodies against poly(ethylene glycol) (PEG) have been found to be the culprit of side reactions and efficacy loss of a number of PEGylated drugs. Fundamental mechanisms of PEG immunogenicity and design principles for PEG alternatives still have not been fully explored. By using hydrophobic interaction chromatography (HIC) under varied salt conditions, we reveal the “hidden” hydrophobicity of those polymers which are generally considered as hydrophilic. A correlation between the hidden hydrophobicity of a polymer and its polymer immunogenicity is observed when this polymer is conjugated with an immunogenic protein. Such a correlation of hidden hydrophobicity vs. immunogenicity for a polymer also applies to corresponding polymer–protein conjugates. Atomistic molecular dynamics (MD) simulation results show a similar trend. Based on polyzwitterion modification and with this HIC technique, we are able to produce extremely low-immunogenic protein conjugates as their hydrophilicity is pushed to the limit and their hydrophobicity is eliminated, breaking the current barriers of eliminating anti-drug and anti-polymer antibodies. The hidden hydrophobicity of conventional polymers widely considered as hydrophilic is closely related to their immunogenicity when they are conjugated to immunogenic proteins.

Project description:Phosphole oxides undergo a highly chemoselective reaction with sulfonyl isocyanates forming sulfonylimino phospholes in high yields. This facile modification proved to be a powerful tool for obtaining new phosphole-based aggregation-induced emission (AIE) luminogens with high fluorescence quantum yields in the solid state. Changing the chemical environment of the phosphorus atom of the phosphole framework results in a significant shift of the fluorescence maximum to longer wavelengths. In this work, we present a powerful synthetic strategy for selectively converting phosphole oxides by reaction with sulfonyl isocyanates into the corresponding sulfonylimino phospholes, which show a decrease or increase in solid-state emission.

Project description:A catalytic asymmetric α-C(sp3)–H functionalization of alkyl silanes with benzosultams was realized by merging photoredox and chiral Lewis acid catalysis. The key to success was the choice of photocatalyst with an appropriate redox potential and non-nucleophilic solvent, providing a novel entry to chiral organosilanes containing two adjacent tri- and tetra-substituted stereocenters with high to efficient diastereo- and enantioselectivity (up to 99% ee, 94 : 6 dr) under mild reaction conditions. Based on the control experiment and spectral analysis, an initial single electron transfer reduction of a benzosultam-triggered simultaneous or stepwise electron transfer/proton transfer process was proposed to rationalize the favored C(sp3)–H functionalization rather than desilylation. A visible-light-induced chemo-, diastereo- and enantioselective α-C(sp3)–H functionalization of alkyl silanes with benzosultams was realized under a bimetallic synergetic catalysis.

Project description:Electron injection is demonstrated to trigger electrocatalytic chain reactions capable of releasing a solvent molecule and forming a redox active guest molecule. One-electron reduction of a hydroxy anthrone derivative (AQH–CH2CN) results in the formation of an anthraquinone radical anion (AQ˙−) and acetonitrile (CH3CN). The resulting fragment of AQ˙− exhibits high stability under mild reducing conditions, and it has enough reducing power to reduce the reactant of AQH–CH2CN. Hence, subsequent electron transfer from AQ˙− to AQH–CH2CN yields the secondary AQ˙− and CH3CN, while the initial AQ˙− is subsequently oxidized to AQ. Overall, the reactants of AQH–CH2CN are completely converted into AQ and CH3CN in sustainable electrocatalytic chain reactions. These electrocatalytic chain reactions are mild and sustainable, successfully achieving catalytic electron-triggered charge-transfer (CT) complex formation. Reactant AQH–CH2CN is non-planar, making it unsuitable for CT interaction with an electron donor host compound (UHAnt2) bearing parallel anthracene tweezers. However, conversion of AQH–CH2CN to planar electron acceptor AQ by the electrocatalytic chain reactions turns on CT interaction, generating a host CT complex with UHAnt2 (AQ ⊂ UHAnt2). Therefore, sustainable electrocatalytic chain reactions can control CT interactions using only a catalytic amount of electrons, ultimately affording a one-electron switch associated with catalytic electron-triggered turn-on molecular recognition. The reactants of AQH–CH2CN are converted into AQ and CH3CN in sustainable electrocatalytic chain reactions, successfully achieving catalytic electron-triggered charge-transfer (CT) complex formation.
