Antioxidant Activity and Photosynthesis Efficiency in Melissa officinalis Subjected to Heavy Metals Stress.

ABSTRACT: The aim of this study was to assess influence of cadmium and zinc treatments on antioxidant activity combined with the photosynthesis efficiency in a popular herb lemon balm (Melissa officinalis L.). Plants were grown under greenhouse conditions by the pot method. The Mn, Cu, Cd, and Zn contents in soil and plants were measured by HR-CS FAAS. The activity of net photosynthesis, stomatal conductance, transpiration rate, intercellular CO₂, and index of chlorophyll in leaves were determined for all investigated species. Reduction of the net photosynthesis was observed for cultivations subjected to either Zn or Cd treatments. Phenolic contents were determined by the chemical Folin-Ciocalteu method, while enhanced voltammetric analysis was applied to assess the antioxidant properties of plant extracts. Both of these approaches yielded similar results. Herbal extracts had exceptional antioxidant capacities and were good scavengers of free radicals and reactive oxygen species. Structural similarity of cadmium and zinc facilitated their mutual structural exchange and prompted substantial expansion of phenolics under the mixed Zn and Cd treatments.

Project description:BackgroundNowadays, light-emitting diodes (LEDs) as a new lighting technology, have been emerged as an alternative source of light for plants due to their wavelength specificity, the narrow width of their bands, small size, solid structure, long lifetime, and low heat generation. Here we investigated the effect of different LED light sources on the essential oil components and antioxidant activity of Melissa officinalis. Two genotypes of lemon balm (Ilam and Isfahan) were subjected to four artificial light treatments, including white, red, blue, red + blue LEDs, and greenhouse light as natural lighting.ResultsThe LED lights significantly increased shoot fresh and dry weights and leaf number in the two genotypes as compared to greenhouse condition. The results showed that the content and composition of essential oil in the two genotypes were variable under different light treatments and the total amount of compounds in the Ilam genotype was higher than the other genotype. The results of analysis of the essential oil by GC/MS indicated that the highest amount of monoterpenes in the genotypes was related to citronellal under red + blue LED lamps (15.3 and 17.2% in Ilam and Isfahan genotypes, respectively) but blue, white, and greenhouse condition had the most effect on sesquiterpenes content in both genotypes. The results showed that the observed variation between the two genotypes in the essentials oil composition was related to the relative percentage of the constituents and not to the appearance or lack of a specific component. Red + blue lighting also provided the highest radical scavenging activity in both genotypes (80.77 and 82.09% for Ilam and Isfahan genotypes, respectively). Based on principal component analyses (PCA), three main groups were identified regarding genotypes and all light treatments.ConclusionsOverall, results indicated that the essentials oil composition of two genotypes of lemon balm was affected both qualitatively and quantitatively by different LED light sources; hence, LED lights might be used to improve monoterpenes, sesquiterpenes, and antioxidant activity in the selected genotypes.

Project description:Lemon balm (Melissa officinalis L.) is a valuable medicinal plant, but its growth can be significantly impacted by drought stress. This study aimed to mitigate the adverse effects of water deficit stress on lemon balm biomass by integrating poultry manure compost, poultry manure biochar, NPK fertilizer, Trichoderma harzianum, Thiobacillus thioparus, and elemental sulfur as soil amendments. The experiment was conducted in a greenhouse using a completely randomized design with a factorial arrangement, consisting of three replicates. It included a water deficit stress factor at three levels (95-100%, 75-80%, and 55-60% of field capacity) and a soil amendment treatment factor with eleven different fertilizer levels. Treatments included control (no amendment), NPK fertilizer, poultry manure compost, poultry manure biochar, and combinations of these with T. harzianum, T. thioparus, and elemental sulfur under various water deficit levels. Water deficit stress significantly reduced photosynthetic pigments, gas exchange parameters, chlorophyll fluorescence, relative water content, and antioxidant enzyme activity, while increasing membrane permeability and lipid peroxidation in lemon balm plants. However, the integrated application of organic, biological, and chemical amendments mitigated these negative impacts. The combined treatment of poultry manure compost, poultry manure biochar, NPK fertilizer, T. harzianum, T. thioparus, and elemental sulfur was the most effective in improving the morpho-physiological properties (1.97-60%) and biomass (2.31-2.76 times) of lemon balm under water deficit stress. The results demonstrate the potential of this holistic approach to enhance the resilience of lemon balm cultivation in water-scarce environments. The integration of organic, biological, and chemical amendments can contribute to sustainable agricultural practices by improving plant morphological and physiological properties and plant performance under drought conditions.
