Alleviation of Hg-, Cr-, Cu-, and Zn-Induced Heavy Metals Stress by Exogenous Sodium Nitroprusside in Rice Plants.

ABSTRACT: The cultivation of rice is widespread worldwide, but its growth and productivity are hampered by heavy metals stress. However, sodium nitroprusside (SNP), a nitric oxide donor, has been found to be effective for imparting heavy metals stress tolerance to plants. Therefore, the current study evaluated the role of exogenously applied SNP in improving plant growth and development under Hg, Cr, Cu, and Zn stress. For this purpose, heavy metals stress was induced via the application of 1 mM mercury (Hg), chromium (Cr), copper (Cu), and zinc (Zn). To reverse the toxic effects of heavy metals stress, 0.1 mM SNP was administrated via the root zone. The results revealed that the said heavy metals significantly reduced the chlorophyll contents (SPAD), chlorophyll a and b, and protein contents. However, SNP treatment significantly reduced the toxic effects of the said heavy metals on chlorophyll (SPAD), chlorophyll a and b, and protein contents. In addition, the results also revealed that heavy metals significantly increased the production of superoxide anion (SOA), hydrogen peroxide (H₂O₂), malondialdehyde (MDA), and electrolyte leakage (EL). However, SNP administration significantly reduced the production of SOA, H₂O₂, MDA, and EL in response to the said heavy metals. Furthermore, to cope with the said heavy metals stress, SNP administration significantly enhanced the activities of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and polyphenol peroxidase (PPO). Furthermore, in response to the said heavy metals, SNP application also upregulated the transcript accumulation of OsPCS1, OsPCS2, OsMTP1, OsMTP5, OsMT-I-1a, and OsMT-I-1b. Therefore, SNP can be used as a regulator to improve the heavy metals tolerance of rice in heavy-metals-affected areas.

Project description:The ASEAN-5 countries (Malaysia, Indonesia, Thailand, Philippines, and Vietnam) of the Association of Southeast Asian Nations as a group is an ever-increasing major economy developmental hub in Asia besides having wealthy natural resources. However, heavy metal (HM) pollution in the region is of increasing environmental and public concern. This study aimed to review and compile the concentrations of Cu, Pb, and Zn in the aquatic sediments of the ASEAN-5 countries published in the literature from 1981 to February 2021. The mean values of Cu, Pb, and Zn in aquatic sediments were elevated and localized in high human activity sites and compared to the earth's upper continental crust and reference values. Based on 176 reports from 113 publications, the ranges of concentrations (mg/kg dry weight) were 0.09-3080 for Cu, 0.37-4950 for Zn, and 0.07-2666 for Pb. The ecological risk (ER) values ranged from 0.02-1077 for Cu, 0.01-95.2 for Zn, and 0.02-784 for Pb. All reports (100%) showed the Zn ER values were categorized as being between 'low potential ecological risk' and 'considerable potential ecological risk'. Almost all Cu ER values (97.7%) also showed similar ranges of the above two risk categories except for a few reports. The highest Cu level (3080 mg/kg dry weight) was reported from a mine-tailing spill in Marinduque Island of the Philippines with 'very high ecological risk'. In addition, drainage sediments in the western part of Peninsular Malaysia were categorized as Cu 'high potential ecological risk'. Almost all reports (96%) showed Pb ER values categorized as between 'low potential ecological risk' and 'moderate potential ecological risk' except for a few reports. Six reports showed Pb ER values of 'considerable potential ecological risk', while one report from Semarang (Indonesia) showed Pb ER of 'very high ecological risk' (Pb level of 2666 mg/kg dry weight). For the ingestion and dermal contact pathways for sediments from the ASEAN-5 countries, all non-carcinogenic risk (NCR) values (HI values 1.0) for Cu, Pb, and Zn reflected no NCR. The ER and human health risk assessment of Cu, Pb, and Zn were compared in an integrative and accurate manner after we reassessed the HM data mentioned in the literature. The synthesis carried out in this review provided the basis for us to consider Cu, Pb, and Zn as being of localized elevated levels. This provided evidence for the ASEAN-5 group of countries to be considered as being a new socio-economic corridor. Beyond any reasonable doubt, an ever-increasing anthropogenic input of HMs is to be expected to a certain degree. We believe that this paper provides the most fundamental useful baseline data for the future management and sustainable development of the aquatic ecosystems in the region. Lastly, we claim that this review is currently the most up-to-date review on this topic in the literature.

Project description:Abscisic acid (ABA) and nitric oxide (NO) are involved in mediating abiotic stress-induced plant physiological responses. Nitraria tangutorum Bobr is a typical salinized desert plant growing in an arid environment. In this study, we investigated the effects of ABA and NO on N.tangutorum seedlings under alkaline stress. Alkali stress treatment caused cell membrane damage, increased electrolyte leakage, and induced higher production of reactive oxygen species (ROS), which caused growth inhibition and oxidative stress in N.tangutorum seedlings. Exogenous application of ABA (15μm) and Sodium nitroprusside (50μm) significantly increased the plant height, fresh weight, relative water content, and degree of succulency in N.tangutorum seedlings under alkali stress. Meanwhile, the contents of ABA and NO in plant leaves were significantly increased. ABA and SNP can promote stomatal closure, decrease the water loss rate, increase leaf surface temperature and the contents of osmotic regulator proline, soluble protein, and betaine under alkali stress. Meanwhile, SNP more significantly promoted the accumulation of chlorophyll a/b and carotenoids, increased quantum yield of photosystem II (φPSII) and electron transport rate (ETRII) than ABA, and decreased photochemical quenching (qP), which improved photosynthetic efficiency and accelerated the accumulation of soluble sugar, glucose, fructose, sucrose, starch, and total sugar. However, compared with exogenous application of SNP in the alkaline stress, ABA significantly promoted the transcription of NtFLS/NtF3H/NtF3H/NtANR genes and the accumulation of naringin, quercetin, isorhamnetin, kaempferol, and catechin in the synthesis pathway of flavonoid metabolites, and isorhamnetin content was the highest. These results indicate that both ABA and SNP can reduce the growth inhibition and physiological damage caused by alkali stress. Among them, SNP has a better effect on the improvement of photosynthetic efficiency and the regulation of carbohydrate accumulation than ABA, while ABA has a more significant effect on the regulation of flavonoid and anthocyanin secondary metabolite accumulation. Exogenous application of ABA and SNP also improved the antioxidant capacity and the ability to maintain Na+/K+ balance of N. tangutorum seedlings under alkali stress. These results demonstrate the beneficial effects of ABA and NO as stress hormones and signaling molecules that positively regulate the defensive response of N. tangutorum to alkaline stress.
