Online Monitoring of Small Volume Reactions Using Compact Liquid Chromatography Instrumentation.

ABSTRACT: A wide variety of analytical techniques have been employed for monitoring chemical reactions, with online instrumentation providing additional benefits compared to offline analysis. A challenge in the past for online monitoring has been placement of the monitoring instrumentation as close as possible to the reaction vessel to maximize sampling temporal resolution and preserve sample composition integrity. Furthermore, the ability to sample very small volumes from bench-scale reactions allows the use of small reaction vessels and conservation of expensive reagents. In this study, a compact capillary LC instrument was used for online monitoring of as small as 1 mL total volume of a chemical reaction mixture, with automated sampling of nL-scale volumes directly from the reaction vessel used for analysis. Analyses to demonstrate short term (~2 h) and long term (~ 50 h) reactions were conducted using tandem on-capillary ultraviolet absorbance followed by in-line MS detection or ultraviolet absorbance detection alone, respectively. For both short term and long term reactions (10 and 250 injections, respectively), sampling approaches using syringe pumps minimized the overall sample loss to ~0.2% of the total reaction volume.

Project description:In untargeted metabolomics, structures of small molecules are annotated using liquid chromatography-mass spectrometry by leveraging information from the molecular retention time (RT) in the chromatogram and m/z (formerly called ''mass-to-charge ratio'') in the mass spectrum. However, correct identification of metabolites is challenging due to the vast array of small molecules. Therefore, various in silico tools for mass spectrometry peak alignment and compound prediction have been developed; however, the list of candidate compounds remains extensive. Accurate RT prediction is important to exclude false candidates and facilitate metabolite annotation. Recent advancements in artificial intelligence (AI) have led to significant breakthroughs in the use of deep learning models in various fields. Release of a large RT dataset has mitigated the bottlenecks limiting the application of deep learning models, thereby improving their application in RT prediction tasks. This review lists the databases that can be used to expand training datasets and concerns the issue about molecular representation inconsistencies in datasets. It also discusses the application of AI technology for RT prediction, particularly in the 5 years following the release of the METLIN small molecule RT dataset. This review provides a comprehensive overview of the AI applications used for RT prediction, highlighting the progress and remaining challenges. SCIENTIFIC CONTRIBUTION: This article focuses on the advancements in small molecule retention time prediction in computational metabolomics over the past five years, with a particular emphasis on the application of AI technologies in this field. It reviews the publicly available datasets for small molecule retention time, the molecular representation methods, the AI algorithms applied in recent studies. Furthermore, it discusses the effectiveness of these models in assisting with the annotation of small molecule structures and the challenges that must be addressed to achieve practical applications.

Project description:Recently, our laboratory has demonstrated the technical feasibility of monitoring dopamine at 1 min temporal resolution with microdialysis and online liquid chromatography. Here, we monitor dopamine in the rat striatum during local delivery of high potassium/low sodium or nomifensine in awake-behaving rats. Microdialysis probes were implanted and perfused continuously with or without dexamethasone in the perfusion fluid for 4 days. Dexamethasone is an anti-inflammatory agent that exhibits several positive effects on the apparent health of the brain tissue surrounding microdialysis probes. Dopamine was monitored 1 or 4 days after implantation under basal conditions, during 10 min applications of 60 mM or 100 mM K+, and during 15 min applications of 10 μM nomifensine. High K+ and nomifensine were delivered locally by adding them to the microdialysis perfusion fluid using a computer-controlled, low-dead-volume six-port valve. Each day/K+/dexamethasone combination elicited specific dopamine responses. Dexamethasone treatment increased dopamine levels in basal dialysates (i.e., in the absence of K+ or nomifensine). Applications of 60 mM K+ evoked distinct responses on days one and four after probe implantation, depending upon the presence or absence of dexamethasone, consistent with dexamethasone's ability to mitigate the traumatic effect of probe implantation. Applications of 100 mM K+ evoked dramatic oscillations in dopamine levels that correlated with changes in the field potential at a metal electrode implanted adjacent to the microdialysis probe. This combination of results indicates the role of spreading depolarization in response to 100 mM K+. With 1 min temporal resolution, we find that it is possible to characterize the pharmacokinetics of the response to the local delivery of nomifensine. Overall, the findings reported here confirm the benefits arising from the ability to monitor dopamine via microdialysis at high sensitivity and at high temporal resolution.
