ABSTRACT: Rhizobacteria are well recognized for their beneficial multifunctions as key promoters of plant development, suppressing pathogens, and improving soil health. In this study, experiments focused on characterizing the plant growth promotion (PGP) and extracellular hydrolase production traits of rhizobacteria, and their impact on Jerusalem artichoke growth. A total of 50 isolates proved capable of either direct PGP or hydrolase-producing traits. Two promising strains (Enterobacter cloacae S81 and Pseudomonas azotoformans C2-114) showed potential on phosphate and potassium solubilization, IAA production, and 1-aminocyclopropane-1-carboxylic acid deaminase activity and hydrolase production. A hydrolase-producing strain (Bacillus subtilis S42) was able to generate cellulase, protease, amylase, β-glucosidase, and phosphatase. These three selected strains also gave positive results for indirect PGP traits such as siderophore, ammonia, oxalate oxidase, polyamine, exopolysaccharide, biofilm, motility, and tolerance to salinity and drought stress. Colonization was observed using a scanning electron microscope and rhizobacteria appeared at the root surface. Interestingly, inoculation with consortia strains (S42, S81, and C2-114) significantly increased all plant parameters, including height, biomass, root (length, surface, diameter, and volume), and tuber fresh weight. Therefore, we recommend that potential consortia of PGP and hydrolase-producing rhizobacteria be employed as a biofertilizer to improve soil and boost crop productivity.

Project description:Helianthus tuberosus L., known as the Jerusalem Artichoke, is a hexaploid plant species, adapted to low nutrient soils, that accumulates high levels of inulin in its tubers. Inulin is a fructose-based polysaccharide used either as dietary fiber or for the production of bioethanol. Key enzymes involved in inulin biosynthesis are well known. However, the gene networks underpinning tuber development and inulin accumulation in H. tuberous remain elusive. To fill this gap, we selected 6,365 ESTs from a H. tuberosus library to set up a microarray platform and record their expression across three tuber developmental stages, when rhizomes start enlarging (T0), at maximum tuber elongation rate (T3) and at tuber physiological maturity (Tm), in “VR” and “K8-HS142”clones. The former was selected as an early tuberizing, the latter as a late-tuberizing clone. We quantified inulin and starch levels, and q-RT PCR confirmed the expression of key genes accounting for inulin biosynthesis. The microarray analysis revealed that the differences in morphological and physiological traits between tubers of the two clones are genetically determined since T0 and that is relatively low the number of differentially expressed ESTs across the stages shared between the clones (93). The expression of ESTs for sucrose:sucrose 1-fructosyltransferase (1-SST) and fructan:fructan 1-fructosyltransferase (1-FFT), the two critical genes for fructans polymerization, resulted to be temporarily synchronized and mirror the progress of inulin accumulation and stretching. The expression of ESTs for starch biosynthesis was insignificant throughout the developmental stages of the clones in line with the negligible level of starch into their mature tubers, where inulin was the dominant polysaccharide. Overall, our study disclosed candidate genes underpinning the development and storage of carbohydrates in the tubers of two H. tuberosus clones. A model according to which the steady state levels of 1-SST and 1-FFT transcripts are developmentally controlled and might represent a limiting factor for inulin accumulation has been provided. Our finding may have significant repercussions for breeding clones with improved levels of inulin for food and chemical industry.

Project description:Plant growth-promoting rhizobacteria (PGPR) are a functionally diverse group of microbes having immense potential as biostimulants and biopesticides. We isolated four PGPR (designated n, L, K, and Y) that confer growth-promoting effects on Arabidopsis thaliana. The present study describes the detailed polyphasic characterization of these PGPR. Classical methods of bacterial identification and biochemical test kits (API20E, API20NE, API ZYM, and API 50CH) revealed their metabolic versatility. All rhizobacterial isolates were positive for 1-aminocyclopropane-1-carboxylate (ACC) deaminase (ACCD) and indole acetic acid production and phosphorous solubilization. PCR analysis confirmed the presence of the nifH gene in strains n, L, and Y, showing their N2-fixation potential. In vitro dual culture methods and bacterial infestation in planta demonstrated that strains n and L exerted antagonistic effects on Pseudomonas syringae pv. tomato DC3000 and Botrytis cinerea 191 and provided protection to Arabidopsis plants against both phytopathogens. Short- or long-term bacterial treatment revealed significant changes in transcript levels of genes annotated to stress response and hormone metabolism in A. thaliana. In particular, the expression of stress-responsive genes in A. thaliana showed an upregulation under salinity stress. MAP kinase 6 (MPK6) was involved in the growth promotion induced by the four bacterial strains. Furthermore, these strains caused a significant increase in root dry weight of maize seedlings under gnotobiotic conditions. We conclude that the four rhizobacteria are good candidates as biofertilizers for enhancing growth of maize, among which strains n and L showed marked plant growth-promoting attributes and the potential to be exploited as functional biostimulants and biopesticides for sustainable agriculture. IMPORTANCE There are pressing needs to reduce the use of agrochemicals, and PGPR are receiving increasing interest in plant growth promotion and disease protection. This study follows up our previous report that the four newly isolated rhizobacteria promote the growth of Arabidopsis thaliana. We test the hypothesis that they have multiple PGP traits and that they can be used as biofertilizers and biopesticides. In vitro assays indicated that these four strains have various PGP properties related to nutrient availability, stress resistance, and/or pest organism antagonism. They significantly influenced the transcript levels of genes involved in stress response and hormone metabolism in A. thaliana. MPK6 is indispensable to the growth stimulation effects. Strains n and L protected A. thaliana seedlings against phytopathogens. Three strains significantly increased maize growth in vitro. In summary, introducing these four strains onto plant roots provides a benefit to the plants. This is the first study regarding the potential mechanism(s) applied by Mucilaginibacter sp. as biostimulants.

Project description:The use of rhizobacteria provide great benefits in terms of nitrogen supply, suppression of plant diseases, or production of vitamins and phytohormones that stimulate the plant growth. At the same time, cyanobacteria can photosynthesize, fix nitrogen, synthesize substances that stimulate rhizogenesis, plant aerial growth, or even suppose an extra supply of carbon usable by heterotrophic bacteria, as well as act as biological control agents, give them an enormous value as plant growth promoters. The present study focused on the in vitro establishment of consortia using heterotrophic bacteria and cyanobacteria and the determination of their effectiveness in the development of tomato seedlings. Microbial collection was composed of 3 cyanobacteria (SAB-M612 and SAB-B866 belonging to Nostocaceae Family) and GS (unidentified cyanobacterium) and two phosphate and potassium solubilizing heterotrophic bacteria (Pseudomonas putida-BIO175 and Pantoea cypripedii-BIO175). The results revealed the influence of the culture medium, incubation time and the microbial components of each consortium in determining their success as biofertilizers. In this work, the most compatible consortia were obtained by combining the SAB-B866 and GS cyanobacteria with either of the two heterotrophic bacteria. Cyanobacteria GS promoted the growth of both rhizobacteria in vitro (increasing logarithmic units when they grew together). While Cyanobacteria SAB-B866 together with both rhizobacteria stimulated the growth of tomato seedlings in planta, leading to greater aerial development of the treated seedlings. Parameters such as fresh weight and stem diameter stood out in the plants treated with the consortia (SAB-B866 and both bacteria) compared to the untreated plants, where the values doubled. However, the increase was more discrete for the parameters stem length and number of leaves. These results suggest that the artificial formulation of microbial consortia can have positive synergistic effects on plant growth, which is of enormous agro-biotechnological interest.
