Dendrobium officinale aqueous extract influences the immune response following vaccination against SARS-CoV-2.

ABSTRACT:

Background

Vaccination is the most effective way to prevent coronavirus disease 2019 (COVID-19). However, it is often less protective and does not significantly increase antibody levels, especially in individuals with impaired immune systems. Nevertheless, the immunocompetence can be enhanced using a natural immunomodulator, such as Dendrobium officinale aqueous extract (DoAE).

Methods

To determine whether DoAE promotes antibody production, we treated healthy volunteers with DoAE during COVID-19 vaccination. Meanwhile, the control volunteers were given a placebo (cornstarch) during the vaccination. Antibody levels were measured at three-week intervals in the DoAE and control groups.

Results

DoAE enhanced immunity and preserved immune cell homeostasis. However, the neutralizing antibody (nAb) levels in the DoAE group were lower than those in the control group. Analysis of the gut microbiota revealed that the abundance of anti-inflammatory flora was increased, while the pro-inflammatory flora was reduced in the DoAE group.

Conclusion

DoAE has immunomodulatory and anti-inflammatory properties. Therefore, DoAE has the potential for COVID-19 prophylaxis, treatment, and recovery from the adverse effects of COVID-19. However, its anti-inflammatory activity affects the production of nAbs. Thus, DoAE may not be recommended for consumption during COVID-19 vaccination.

Project description:IntroductionDendrobium officinale Kimura et Migo (D. officinale) , widely called as "life-saving immortal grass" by Chinese folk, is a scarce and endangered species. The edible stems of D. officinale have been extensively studied for active chemical components and various bioactivities. However, few studies have reported the well-being beneficial effects of D. officinale flowers (DOF). Therefore, the present study aimed to investigate the in vitro biological potency of its aqueous extract and screen its active components.MethodsAntioxidant tests, including 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), the ferric reducing ability of plasma (FRAP), and intracellular reactive oxygen species (ROS) level analyses in primary human epidermal keratinocytes, anti-cyclooxygenase2 (COX-2) assay, anti-glycation assay (both fluorescent AGEs formation in a BSA fructose/glucose system and glycation cell assay), and anti-aging assay (quantification of collagen types I and III, and SA-β-gal staining assay) were conducted to determine the potential biological effects of DOF extracts and its major compounds. Ultra-performance liquid chromatography-electrospray ionisation-quadrupole-time-of-flight-mass spectrometry (UPLC-ESI-QTOF-MS/MS) was performed to investigate the composition of DOF extracts. Online antioxidant post-column bioassay tests were applied to rapidly screen major antioxidants in DOF extracts.Results and discussionThe aqueous extract of D. officinale flowers was found to have potential antioxidant capacity, anti-cyclooxygenase2 (COX-2) effect, anti-glycation potency, and anti-aging effects. A total of 34 compounds were identified using UPLC-ESI-QTOF-MS/MS. Online ABTS radical analysis demonstrated that 1-O-caffeoyl-β-D-glucoside, vicenin-2, luteolin-6-C-β-D-xyloside-8-C-β--D-glucoside, quercetin-3-O-sophoroside, rutin, isoquercitrin, and quercetin 3-O-(6″-O-malonyl)-β-D-glucoside are the major potential antioxidants. In addition, all selected 16 compounds exerted significant ABTS radical scavenging ability and effective AGE suppressive activities. However, only certain compounds, such as rutin and isoquercitrin, displayed selective and significant antioxidant abilities, as shown by DPPH and FRAP, as well as potent COX-2 inhibitory capacity, whereas the remaining compounds displayed relatively weak or no effects. This indicates that specific components contributed to different functionalities. Our findings justified that DOF and its active compound targeted related enzymes and highlighted their potential application in anti-aging.

Project description:Introduction: Numerous studies have demonstrated that the stems of D. officinale have the effect of lowering blood glucose, but the leaves of D. officinale have seldom been investigated. In this study, we mainly studied the hypoglycemic effect and mechanism of D. officinale leaves. Methods: Initially in vivo, male C57BL/6 mice were administered either standard feed (10 kcal% fat) or high-fat feed (60 kcal% fat) along with either normal drinking water or drinking water containing 5 g/L water extract of D. officinale leaves (EDL) for 16 weeks, and changes in body weight, food intake, blood glucose, etc., were monitored weekly. Next in vitro, C2C12 myofiber precursor cells which were induced to differentiate into myofibroblasts and cultured with EDL to detect the expression of insulin signaling pathway related proteins. HEPA cells were also cultured with EDL to detect the expression of hepatic gluconeogenesis or hepatic glycogen synthesis related proteins. Eventually after separating the components from EDL by ethanol and 3 kDa ultrafiltration centrifuge tube, we conducted animal experiments using the ethanol-soluble fraction of EDL (ESFE), ethanol-insoluble fraction of EDL (EIFE), ESFE with a molecular weight of >3 kDa (>3 kDa ESFE), and ESFE with a molecular weight of <3 kDa (<3 kDa ESFE) for intensive study. Results: The results in vivo revealed that the mice fed the high-fat diet exhibited significantly decreased blood glucose levels and significantly increased glucose tolerance after the EDL treatment, whereas the mice fed the low-fat diet did not. The results in vitro showed that EDL activated the expression of protein kinase B (AKT), the phosphorylation of AKT, and the expression of downstream GSK3β in the insulin signaling pathway. EDL treatment of HEPA cells confirmed that EDL did not affect hepatic gluconeogenesis or hepatic glycogen synthesis. In the experiment of studying the composition of EDL, we found that the >3 kDa ESFE displayed the effect of lowering blood glucose. In summary, the effect of EDL in lowering blood glucose may bethanole achieved by activating the insulin signaling pathway to increase insulin sensitivity, and the main functional substance was contained within the >3 kDa ESFE. Discussion: The findings of this study represent a reference point for further exploration of the hypoglycemic effects of D. officinale leaves and may assist in both the identification of new molecular mechanisms to improve insulin sensitivity and the isolation of monomeric substances that lower blood glucose. Furthermore, the obtained results may provide a theoretical basis for the development of hypoglycemic drugs with D. officinale leaves as the main component.
