Two-ring chirality generated by the alignment of two achiral phenylacetylene macrocycles.

ABSTRACT: When two achiral rings are bound mechanically, a chiral source is generated in the assembly. The chiroptical properties could be modulated according to the relative occupation of each ring in the assembly. In fact, we have found that two isomeric assemblies (1 and 2) show unique properties in each assembly with two achiral rings of phenylacetylene macrocycle (PAM). When considering the difference in the chiroptical properties of these two isomeric assemblies (6PAM × 2), no comparison was available based on no activity of the achiral component element itself (6PAM). In this work, we synthesized a two-ring chiral analog (4) by the ring-fusion of two 6PAMs to an 11PAM, and examined the chiroptical properties of 4, since the single helix was imparted as a chiral source. By comparison of the chiroptical properties (molar circular dichroism and molar optical rotation) of 1 and 2 to those of 4, we demonstrated that the disparity was related to the alignment of the two achiral rings.

Chiral diversification through the assembly of achiral phenylacetylene macrocycles with a two-fold bridge.

Project description:We demonstrate so-called "chiral diversification", which is a design strategy to create multiple chiral molecules through the assembly and double-bridging of achiral components. We used phenylacetylene macrocycles (PAMs) as an achiral element. In a molecule, two achiral rings of [6]PAM are stacked one above the other, or bound to each other mechanically. As an alternative, a single enlarged ring of [12]PAM was also assumed to be a doubled form of [6]PAM. In any case, one or two ring(s) are doubly-bridged by covalent bonds to exert chirality. Through intramolecular two-bond formation, these multiple chiral molecules were obtained as a set of products in one reaction. The dynamic chirality generated in molecules with either two helically-stacked rings of [6]PAM or a single helically-folded ring of [12]PAM was characterized by induced Cotton effects with the aid of an external chiral source. Thus, a chiral structure based on [12]PAM could be demonstrated as the first success. Alternatively, enantiomeric separation was achieved for molecules with two interlocked rings of [6]PAM to show remarkable chiroptical properties.

Emergence of single-molecular chirality from achiral reactants.

Project description:The synthesis of enantiopure molecules from achiral precursors without the need for pre-existing chirality is a major challenge associated with the origin of life. We here show that an enantiopure product can be obtained from achiral starting materials in a single organic reaction. An essential characteristic of this reaction is that the chiral product precipitates from the solution, introducing a crystal-solution interface which functions as an asymmetric autocatalytic system that provides sufficient chiral amplification to reach an enantiopure end state. This approach not only provides more insight into the origin of life but also offers a pathway to acquire enantiopure compounds for industrial applications.

Condensation and asymmetric amplification of chirality in achiral molecules adsorbed on an achiral surface.

Project description:The origin of homochirality in nature is an important but open question. Here, we demonstrate a simple organizational chiral system constructed by achiral carbon monoxide (CO) molecules adsorbed on an achiral Au(111) substrate. Combining scanning tunneling microscope (STM) measurements with density-functional-theory (DFT) calculations, two dissymmetric cluster phases consisting of chiral CO heptamers are revealed. By applied high bias voltage, the stable racemic cluster phase can be transformed into a metastable uniform phase consisting of CO monomers. Further, during the recondensation of a cluster phase after lowering down bias voltage, an enantiomeric excess and its chiral amplification occur, resulting in a homochirality. Such asymmetry amplification is found to be both kinetically feasible and thermodynamically favorable. Our observations provide insight into the physicochemical origin of homochirality through surface adsorption and suggest a general phenomenon that can influence enantioselective chemical processes such as chiral separations and heterogeneous asymmetric catalysis.

Improving the reactivity of phenylacetylene macrocycles toward topochemical polymerization by side chains modification.

Project description:The synthesis and self-assembly of two new phenylacetylene macrocycle (PAM) organogelators were performed. Polar 2-hydroxyethoxy side chains were incorporated in the inner part of the macrocycles to modify the assembly mode in the gel state. With this modification, it was possible to increase the reactivity of the macrocycles in the xerogel state to form polydiacetylenes (PDAs), leading to a significant enhancement of the polymerization yields. The organogels and the PDAs were characterized using Raman spectroscopy, X-ray diffraction (XRD) and scanning electron microscopy (SEM).

Multi-level chirality in liquid crystals formed by achiral molecules.

Project description:Complex materials often exhibit a hierarchical structure with an intriguing mechanism responsible for the 'propagation' of order from the molecular to the nano- or micro-scale level. In particular, the chirality of biological molecules such as nucleic acids and amino acids is responsible for the helical structure of DNA and proteins, which in turn leads to the lack of mirror symmetry of macro-bio-objects. To fully understand mechanisms of cross-level order transfer there is an intensive search for simpler artificial structures exhibiting hierarchical arrangement. Here we present complex systems built of achiral molecules that show four levels of structural chirality: layer chirality, helicity of a basic repeating unit, mesoscopic helix and helical filaments. The structures are identified by a combination of hard and soft x-ray diffraction measurements, optical studies and theoretical modelling. Similarly to many biological systems, the studied materials exhibit a coupling between chirality at different levels.

Electronic chirality inversion of lanthanide complex induced by achiral molecules.

Project description:A novel mechanism for chiroptical activity inversion based on the electronic structure of metal complexes without Λ- or Δ-type structure change was demonstrated spectroscopically and theoretically. To demonstrate the mechanism, a europium (Eu(III)) complex with chiral (+)-3-(trifluoroacetyl)camphor (+tfc) and achiral triphenylphosphine oxide (tppo) was prepared. The steric and electronic structures of the Eu(III) complex were adjusted by additional achiral tppo and coordinating acetone molecules, and were characterised by 1H NMR, photoluminescence, and emission lifetime measurements. The optical activity of the Eu(III) complex in solution was evaluated by circularly polarized luminescence (CPL) measurements. CPL sign inversion, which was independent of Λ- or Δ-type structure changes from the spectroscopic viewpoint, and a drastic CPL intensity enhancement were observed depending on the external achiral molecules around Eu(III) ion. These phenomena provide the first clarification of optical activity change associated with electronic structure rather than chiral coordination structure-type (Λ or Δ) under external environments.

Computational design of mixed chirality peptide macrocycles with internal symmetry.

Project description:Cyclic symmetry is frequent in protein and peptide homo-oligomers, but extremely rare within a single chain, as it is not compatible with free N- and C-termini. Here we describe the computational design of mixed-chirality peptide macrocycles with rigid structures that feature internal cyclic symmetries or improper rotational symmetries inaccessible to natural proteins. Crystal structures of three C2- and C3-symmetric macrocycles, and of six diverse S2-symmetric macrocycles, match the computationally-designed models with backbone heavy-atom RMSD values of 1 Å or better. Crystal structures of an S4-symmetric macrocycle (consisting of a sequence and structure segment mirrored at each of three successive repeats) designed to bind zinc reveal a large-scale zinc-driven conformational change from an S4-symmetric apo-state to a nearly inverted S4-symmetric holo-state almost identical to the design model. These symmetric structures provide promising starting points for applications ranging from design of cyclic peptide based metal organic frameworks to creation of high affinity binders of symmetric protein homo-oligomers. More generally, this work demonstrates the power of computational design for exploring symmetries and structures not found in nature, and for creating synthetic switchable systems.

Artificial Macrocycles by Ugi Reaction and Passerini Ring Closure.

Project description:Artificial macrocycles can be convergently synthesized by a sequence of an Ugi multicomponent reaction (MCR) followed by an intramolecular Passerini MCR used to close the macrocycle. Significantly, in this work, the first intramolecular macrocyclization through a Passerini reaction is described. We describe 21 macrocycles of a size of 15-20. The resulting macrocyclic depsipeptides are model compounds for natural products and could find applications in drug discovery.

Effect of Chirality on the Binding of Viologen Guests in Porphyrin Macrocycles.

Project description:As part of a project aimed at the development of chiral processive catalysts that can write information on a polymer chain we describe the synthesis of two optically active porphyrin macrocycles, which are prepared in 3 steps from an achiral precursor compound. Fluorescence and 1H-NMR studies show that one of the macrocycles displays selectivity in the binding of chiral viologen guest molecules.

Spontaneous emergence of chirality in achiral lyotropic chromonic liquid crystals confined to cylinders.

Project description:The presumed ground state of a nematic fluid confined in a cylindrical geometry with planar anchoring corresponds to that of an axial configuration, wherein the director, free of deformations, is along the long axis of the cylinder. However, upon confinement of lyotropic chromonic liquid crystals in cylindrical geometries, here we uncover a surprising ground state corresponding to a doubly twisted director configuration. The stability of this ground state, which involves significant director deformations, can be rationalized by the saddle-splay contribution to the free energy. We show that sufficient anisotropy in the elastic constants drives the transition from a deformation-free ground state to a doubly twisted structure, and results in spontaneous symmetry breaking with equal probability for either handedness. Enabled by the twist angle measurements of the spontaneous twist, we determine the saddle-splay elastic constant for chromonic liquid crystals for the first time.
