ABSTRACT: We present 16S amplicon data derived from the nest materials of three species of Australian stingless bees (Meliponini). This data set reveals the diversity of bacteria associated with these materials. It will serve as a valuable baseline for further study of the nest microbiome and comparison with the stingless bee microbiota.

Contrasting patterns of foraging behavior in neotropical stingless bees using pollen and honey metabarcoding.

Project description:Stingless bees are major flower visitors in the tropics, but their foraging preferences and behavior are still poorly understood. Studying stingless bee interactions with angiosperms is methodologically challenging due to the high tropical plant diversity and inaccessibility of upper canopy flowers in forested habitats. Pollen DNA metabarcoding offers an opportunity of assessing floral visitation efficiently and was applied here to understand stingless bee floral resources spectra and foraging behavior. We analyzed pollen and honey from nests of three distantly related stingless bee species, with different body size and social behavior: Melipona rufiventris, Scaptotrigona postica and Tetragonisca angustula. Simultaneously, we evaluate the local floristic components through seventeen rapid botanical surveys conducted at different distances from the nests. We discovered a broad set of explored floral sources, with 46.3 plant species per bee species in honey samples and 53.67 in pollen samples. Plant families Myrtaceae, Asteraceae, Euphorbiaceae, Melastomataceae and Malpighiaceae dominated the records, indicating stingless bee preferences for abundant resources that flowers of these families provide in the region. Results also reinforce the preference of stingless bees for forest trees, even if only available at long distances. Our high-resolution results encourage future bee-plant studies using pollen and honey metabarcoding in hyper-diverse tropical environments.

Reassessing Hybridisation in Australian <i>Tetragonula</i> Stingless Bees Using Multiple Genetic Markers.

Project description:We re-examined reports of hybridisation in three cryptic stingless bee species in the genus Tetragonula in South East Queensland, Australia (T. carbonaria, T. davenporti and T. hockingsi). Previous studies on this group using microsatellite markers proposed that hybridisation occasionally takes place. In contrast, we find that using 1745 SNPs we could reliably separate the three species, with no evidence of contemporary (or recent) hybridisation. We found identical amplicon sequences of the nuclear gene EF1alpha across most individuals of the three species, but low and moderate species-specific polymorphisms in the nuclear gene Opsin and the mitochondrial 16S rRNA gene, respectively, with no cases of mito-nuclear discordance at these genes. We confirm that nuclear divergence across these species is low, based on 10-26 kb of non-coding sequence flanking EF1alpha and Opsin (0.7%-1% pairwise difference between species). However, we find mitogenomes to be far more diverged than nuclear genomes (21.6%-23.6% pairwise difference between species). Based on these comprehensive analyses of multiple marker types, we conclude there is no ongoing gene flow among the Tetragonula species of South East Queensland, despite their morphological similarity to one another and the low nuclear divergence among them. The higher resolution provided by multiple SNP markers may lead to lower estimates of contemporary hybridisation more generally.

Bacteria, yeasts, and fungi associated with larval food of Brazilian native stingless bees.

Project description:Stingless bees are a diverse group with a relevant role in pollinating native species. Its diet is rich in carbohydrates and proteins, by collecting pollen and nectar supplies the development of its offspring. Fermentation of these products is associated with microorganisms in the colony. However, the composition of microorganisms that comprise this microbiome and its fundamental role in colony development is still unclear. To characterize the colonizing microorganisms of larval food in the brood cells of stingless bees Frieseomelitta varia, Melipona quadrifasciata, Melipona scutellaris, and Tetragonisca angustula, we have utilized molecular and culture-based techniques. Bacteria of the phyla Firmicutes, Proteobacteria, Actinobacteria, and fungi of the phyla Ascomycota, Basidiomycota, Mucoromycota, and Mortierellomycota were found. Diversity analysis showed that F. varia had a greater diversity of bacteria in its microbiota, and T. angustula had a greater diversity of fungi. The isolation technique allowed the identification of 189 bacteria and 75 fungi. In summary, this research showed bacteria and fungi associated with the species F. varia, M. quadrifasciata, M. scutellaris, and T. angustula, which may play an essential role in the survival of these organisms. Besides that, a biobank with bacteria and fungus isolates from LF of Brazilian stingless bees was created, which can be used for different studies and the prospection of biotechnology compounds.

Appetite for self-destruction: suicidal biting as a nest defense strategy in <i>Trigona</i> stingless bees.

Project description:Self-sacrificial behavior represents an extreme and relatively uncommon form of altruism in worker insects. It can occur, however, when inclusive fitness benefits are high, such as when defending the nest. We studied nest defense behaviors in stingless bees, which live in eusocial colonies subject to predation. We introduced a target flag to nest entrances to elicit defensive responses and quantified four measures of defensivity in 12 stingless bee species in São Paulo State, Brazil. These included three Trigona species, which are locally known for their aggression. Species varied significantly in their attack probability (cross species range = 0-1, P < 0.001), attack latency (7.0-23.5 s, P = 0.002), biting duration of individual bees (3.5-508.7 s, P < 0.001), and number of attackers (1.0-10.8, P < 0.001). A "suicide" bioassay on the six most aggressive species determined the proportion of workers willing to suffer fatal damage rather than disengage from an intruder. All six species had at least some suicidal individuals (7-83 %, P < 0.001), reaching 83 % in Trigona hyalinata. Biting pain was positively correlated with an index of overall aggression (P = 0.002). Microscopic examination revealed that all three Trigona species had five sharp teeth per mandible, a possible defensive adaptation and cause of increased pain. Suicidal defense via biting is a new example of self-sacrificial altruism and has both parallels and differences with other self-sacrificial worker insects, such as the honey bee. Our results indicate that suicidal biting may be a widespread defense strategy in stingless bees, but it is not universal.

Resin Use by Stingless Bees: A Review.

Project description:Stingless bees (Meliponini) are highly social bees that are native to tropical and sub-tropical ecosystems. Resin use is vital to many aspects of stingless bee colony function. Stingless bees use resin to build essential nest structures, repel predators, and kill would-be invaders. Furthermore, resin-derived compounds have been found to enrich the cuticular chemical profiles of many stingless bee species, and resin may play an important role in shaping the microbial communities associated with stingless bees and their nests. Despite its importance for colony function, previous reviews of resin use by stingless bees are lacking. This topic grows increasingly urgent as changes in beekeeping and land use practices occur, potentially diminishing stingless bees' ability to incorporate resin into the nest environment. In this article, we review existing literature on resin use by stingless bees and discuss potential areas of future research.
