Project description:Here, we report the development of antibacterial and compostable electrospun polylactic acid (PLA) fibers by incorporation of a multifunctional biobased polymer in the process. The multifunctional polymer was synthesized from the bio-sourced itaconic acid building block by radical polymerization followed by click chemistry reaction with hydantoin groups. The resulting polymer possesses triazole and hydantoin groups available for further N-alkylation and chlorination reaction, which provide antibacterial activity. This polymer was added to the electrospinning PLA solution at 10 wt %, and fiber mats were successfully prepared. The obtained fibers were surface-modified through the accessible functional groups, leading to the corresponding cationic triazolium and N-halamine groups. The fibers with both antibacterial functionalities demonstrated high antibacterial activity against Gram-positive and Gram-negative bacteria. While the fibers with cationic surface groups are only effective against Gram-positive bacteria (Staphylococcus epidermidis and Staphylococcus aureus), upon chlorination, the activity against Gram-negative Escherichia coli and Pseudomonas aeruginosa is significantly improved. In addition, the compostability of the electrospun fibers was tested under industrial composting conditions, showing that the incorporation of the antibacterial polymer does not impede the disintegrability of the material. Overall, this study demonstrates the feasibility of this biobased multifunctional polymer as an antibacterial agent for biodegradable polymeric materials with potential application in medical uses.

Project description:Protein hydrolysis enables production of peptides and free amino acids that are suitable for usage in food and feed or can be used as precursors for bulk chemicals. Several essential amino acids for food and feed have hydrophobic side chains; this property may also be exploited for subsequent separation. Here, we present methods for selective production of hydrophobic amino acids from proteins. Selectivity can be achieved by selection of starting material, selection of hydrolysis conditions, and separation of achieved hydrolysate. Several protease combinations were applied for hydrolysis of rubber seed protein concentrate, wheat gluten, and bovine serum albumin (BSA). High degree of hydrolysis (>50 %) could be achieved. Hydrophobic selectivity was influenced by the combination of proteases and by the extent of hydrolysis. Combination of Pronase and Peptidase R showed the highest selectivity towards hydrophobic amino acids, roughly doubling the content of hydrophobic amino acids in the products compared to the original substrates. Hydrophobic selectivity of 0.6 mol-hydrophobic/mol-total free amino acids was observed after 6 h hydrolysis of wheat gluten and 24 h hydrolysis of rubber seed proteins and BSA. The results of experiments with rubber seed proteins and wheat gluten suggest that this process can be applied to agro-industrial residues.

Project description:The aim of this study is to demonstrate the feasibility of different functionalization methods for electrospun fibers developed using benign solvents. In particular three different approaches were investigated to achieve the functionalization of poly(epsilon caprolactone) (PCL) electrospun fibers with fibronectin. Protein surface entrapment, chemical functionalization and coaxial electrospinning were performed and compared. Moreover, bilayered scaffolds, with a top patterned and functionalized layer with fibronectin and a randomly oriented not functionalized layer were fabricated, demonstrating the versatility of the use of benign solvents for electrospinning also for the fabrication of complex graded structures. Besides the characterization of the morphology of the obtained scaffolds, ATR-FTIR and ToF-SIMS were used for the surface characterization of the functionalized fibers. Cell adhesion and proliferation were also investigated by using ST-2 cells. Positive results were obtained from all functionalized scaffolds and the most promising results were obtained with bilayered scaffolds, in terms of cells infiltration inside the fibrous structure.

Project description:Hypertrophic scars (HTSs) are pathological structures resulting from chronic inflammation during the wound healing process, particularly in complex injuries like burns. The aim of this work is to propose Biofiber PF (biodegradable fiber loaded with Pirfenidone 1.5 w/w), an electrospun advanced dressing, as a solution for HTSs treatment in complex wounds. Biofiber has a 3-day antifibrotic action to modulate the fibrotic process and enhance physiological healing. Its electrospun structure consists of regular well-interconnected Poly-L-lactide-co-poly-ε-caprolactone (PLA-PCL) fibers (size 2.83 ± 0.46 µm) loaded with Pirfenidone (PF, 1.5% w/w), an antifibrotic agent. The textured matrix promotes the exudate balance through mild hydrophobic wettability behavior (109.3 ± 2.3°), and an appropriate equilibrium between the absorbency % (610.2 ± 171.54%) and the moisture vapor transmission rate (0.027 ± 0.036 g/min). Through its finer mechanical properties, Biofiber PF is conformable to the wound area, promoting movement and tissue oxygenation. These features also enhance the excellent elongation (>500%) and tenacity, both in dry and wet conditions. The ancillary antifibrotic action of PF on hypertrophic scar fibroblast (HSF) for 3 days downregulates the cell proliferation over time and modulates the gene expression of transforming growth factor β1 (TGF-β1) and α-smooth muscle actin (α-SMA) at 48-72 h. After 6 days of treatment, a decrement of α-SMA protein levels was detected, proving the potential of biofiber as a valid therapeutic treatment for HTSs in an established wound healing process.

Project description:We present a robust method to prepare thin oriented nematic liquid crystalline elastomer-polymer (LCE-polymer) core-sheath fibers. An electrospinning setup is utilized to spin a single solution of photo-crosslinkable low molecular weight reactive mesogens and a support polymer to form the coaxial LCE-polymer fibers, where the support polymer forms the sheath via in situ phase separation as the solvent evaporates. We discuss the effect of phase separation and compare two different sheath polymers (polyvinylpyrrolidone and polylactic acid), investigating optical and morphological properties of obtained fibers, as well as the shape changes upon heating. The current fibers show only irreversible contraction, the relaxation most likely being hindered by the presence of the passive sheath polymer, increasing in stiffness on cooling. If the sheath polymer can be removed while keeping the LCE core intact, we expect LCE fibers produced in this way to have potential to be used as actuators, for instance in soft robotics and responsive textiles.

Project description:Most of antimicrobial peptides interact with food components decreasing their activity, which limit their successful incorporation into packaging material, functional foods and edible films. The aim of this work was to develop a nisin carrier. Nanofibers of amaranth protein and pullulan (50:50) loaded with nisin were obtained by electrospinning. The nanofibers morphology was determined by scanning electron microscopy and fluorescent microscopy. The molecular interactions were characterized by infrared spectroscopy, X-ray diffraction, differential scanning calorimetry, and thermogravimetric analysis. The nisin loading efficiency as well as the antimicrobial activity against Leuconostoc mesenteroides were evaluated. The micrographs of the obtained materials exhibited smooth and continuous fibers with no defects characterized by diameters between 124 and 173 nm. The FTIR analysis showed intermolecular interactions mainly by hydrogen bonding. The electrospinning process improved the thermal properties of the polymeric mixture displacing the Tm peak to higher temperatures and increasing crystallinity. The antimicrobial activity of nisin in broth and agar against L. mesenteroides was maintained after incorporation into fibers. The results presented an outlook for the potential use of protein amaranth nanofibers when incorporating antimicrobials as a food preservation strategy.

Project description:Nano- and micro- fibers of conjugated polymer semiconductors are particularly interesting both for applications and for fundamental research. They allow an investigation into how electronic properties are influenced by size confinement and chain orientation within microstructures that are not readily accessible within thin films. Moreover, they open the way to many applications in organic electronics, optoelectronics and sensing. Electro-spinning, the technique subject of this review, is a simple method to effectively form and control conjugated polymer fibers. We provide the basics of the technique and its recent advancements for the formation of highly conducting and high mobility polymer fibers towards their adoption in electronic applications.

Project description:In this work, different nanocomposite electrospun fiber mats were obtained based on poly(e-caprolactone) (PCL) and reinforced with both organic and inorganic nanoparticles. In particular, on one side, cellulose nanocrystals (CNC) were synthesized and functionalized by "grafting from" reaction, using their superficial OH- group to graft PCL chains. On the other side, commercial chitosan, graphene as organic, while silver, hydroxyapatite, and fumed silica nanoparticles were used as inorganic reinforcements. All the nanoparticles were added at 1 wt% with respect to the PCL polymeric matrix in order to compare the different behavior of the woven no-woven nanocomposite electrospun fibers with a fixed amount of both organic and inorganic nanoparticles. From the thermal point of view, no difference was found between the effect of the addition of organic or inorganic nanoparticles, with no significant variation in the Tg (glass transition temperature), Tm (melting temperature), and the degree of crystallinity, leading in all cases to high crystallinity electrospun mats. From the mechanical point of view, the highest values of Young modulus were obtained when graphene, CNC, and silver nanoparticles were added to the PCL electrospun fibers. Moreover, all the nanoparticles used, both organic and inorganic, increased the flexibility of the electrospun mats, increasing their elongation at break.

Project description:Chitosan/poly(vinyl alcohol)/amino-functionalized montmorillonite nanocomposite electrospun membranes with enhanced adsorption capacity and thermomechanical properties were fabricated and utilized for the removal of a model cationic dye (Basic Blue 41). Effects of nanofiller concentrations (up to 3.0 wt %) on the morphology and size of the nanofibers as well as the porosity and thermomechanical properties of the nanocomposite membranes are studied. It is shown that the incorporation of the nanoclay particles with ∼10 nm lateral sizes into the polymer increases the size of the pores by about 80%. To demonstrate the efficiency of the adsorbents, the dye removal rate is investigated as a function of pH, adsorbent dosage, dye concentration, and nanofiller loading. The highest and fastest dye removal occurs for the nanofibrous membranes containing 2 wt % nanofiller, where about 80% of the cationic dye is removed after 15 min. This performance is at least 20% better than the pristine chitosan/poly(vinyl alcohol) membrane. The thermal stability and compression resistance of the nanocomposite membranes are found to be higher than those of the pristine membrane. In addition, reusability studies show that the dye removal performance of this nanocomposite membrane reduces by only about 5% over four cycles. The adsorption kinetics is explained by the Langmuir isotherm model and is expressed by a pseudo-second-order kinetic mechanism that determines a spontaneous chemisorption process. The results of this study provide a valuable perspective on the fabrication of high-performance, reusable, and efficient electrospun fibrous nanocomposite adsorbents.

Project description:Wetting of electrospun mats plays a huge role in tissue engineering and filtration applications. However, it is challenging to trace the interrelation between the wetting of individual nano-sized fibers and the macroscopic electrospun mat. Here we measured the wetting of different nylon-11 samples - solution-cast films, electrospun fibers deposited onto a substrate, and free-standing mats. With electrospun nylon-11 on aluminium foil, we traced the dependence of the wetting contact angle on the fibers' surface density (substrate coverage). When the coverage was low, the contact angle increased almost linearly with it. At ∼17-20% coverage, the contact angle achieved its maximum of 124 ± 7°, which matched the contact angle of a non-woven electrospun mat, 126 ± 2°. Our results highlight the importance of the outermost layer of fibers for the wetting of electrospun mats.
