Yield and Nutraceutical Value of Lettuce and Basil Improved by a Microbial Inoculum in Greenhouse Experiments.

ABSTRACT: Members of Bacillus spp. have been widely used to enrich the soil/root interface to provide plant growth promoting activities. A new isolate, namely to Bacillus sp. VWC18, has been tested under greenhouse conditions in lettuce (Lactuca sativa L.) pots at different concentrations (10³, 10⁵, 10⁷, and 10⁹ CFU·mL^-1) and application time (single inoculum at transplant and multiple inoculum every ten days) to evaluate the best application dose and frequency. Analysis of foliar yield, main nutrients, and minerals evidenced a significant response for all applications. The lowest (10³ CFU·mL^-1) and the highest doses (10⁹ CFU·mL^-1), applied every ten days until harvest, had the greatest efficacy; the nutrient yield (N, K, P, Na, Ca, Fe, Mg, Mn, Cu, and B) increased more than twice. A new randomized block design with three replicates was then performed in lettuce and basil (Ocinum basilicum L.), with the two best performing concentrations applied every ten days. In addition to previous analysis, root weight, chlorophyll, and carotenoids were also examined. Both experiments confirmed the previous results: inoculation of the substrate with Bacillus sp. VWC18 promoted plant growth, chlorophyll, and mineral uptake in both crop species. Root weight duplicated or triplicated compared to control plants, and chlorophyll concentration reached even higher values. Both parameters had a dose-dependent increase.

Project description:IntroductionMore than half of the world's population consumes rice as their primary food. The majority of rice production is concentrated in Asia, with the top 10 rice-growing countries accounting for 84% of the world's total rice cultivation. However, rice production is also strongly linked to environmental changes. Among all the global sources of greenhouse gas (GHG) emissions, paddy cultivation stands out as a significant contributor to global methane (CH4) and nitrous oxide (N2O) emissions. This contribution is expected to increase further with the projected increase of 28% in global rice output by 2050. Hence, modifications to rice management practices are necessary both to increase yield and mitigate GHG emissions.MethodsWe investigated the effect of seedling treatment, soil application, and foliar application of a methane-derived microbial biostimulant on grain yield and GHG emissions from rice fields over three seasons under 100% fertilizer conditions. Further, microbial biostimulant was also tested under 75% nitrogen (N) levels to demonstrate its effect on grain yield. To understand the mechanism of action of microbial biostimulant on crop physiology and yield, a series of physiological, transcript, and metabolite analyses were also performed.ResultsOur three-season open-field studies demonstrated a significant enhancement of grain yield, up to 39%, with a simultaneous reduction in CH4 (31%-60%) and N2O (34%-50%) emissions with the use of methane-derived microbial biostimulant. Under 75% N levels, a 34% increase in grain yield was observed with microbial biostimulant application. Based on the physiological, transcript, and metabolite analyses data, we were further able to outline the potential mechanisms for the diverse synergistic effects of methane-derived microbial biostimulant on paddy, including indole-3-acetic acid production, modulation of photosynthesis, tillering, and panicle development, ultimately translating to superior yield.ConclusionThe reduction in GHG emission and enhanced yield observed under both recommended and reduced N conditions demonstrated that the methane-derived biostimulant can play a unique and necessary role in the paddy ecosystem. The consistent improvements seen across different field trials established that the methane-derived microbial biostimulant could be a scalable solution to intensify rice productivity with a lower GHG footprint, thus creating a win-win-win solution for farmers, customers, and the environment.

Project description:Water scarcity is a worldwide problem, and in order to obtain plenty of production, agricultural irrigation water accounts for a large portion. Many studies have shown that the interaction of root microorganisms and soil can promote crop growth. Developing ways to reduce irrigation to maintain soil fertility and ensure crop yield by regulating the root microenvironment is an important research goal. Here, we developed a reasonable irrigation plan for eggplant cultivation in a solar greenhouse. The maximum theoretical amount of water demand during eggplant planting obtained from a previous study was used as the control (CK), and the irrigation in the treatments was reduced by 10, 20 and 30% relative to this amount. The 10% irrigation reduction treatment (T1) significantly improved soil nutrients and increased soil catalase, urease and alkaline phosphatase activities (p < 0.05). Further analysis of rhizosphere microorganisms revealed the highest richness and diversity of the microbial community under the T1 treatment, with Bacilli as the most abundant bacteria and Aspergillaceae as the most abundant fungi and lower relative abundances of Chloroflexi and Acidobacteria (p < 0.05). Changes in microbial community structure under the influence of different irrigation treatments resulted in improvements in rhizosphere N cycling and nutrient catabolism. The plant-microbe interactions led to significant increases in eggplant plant height, root vigour, root surface area, leaf chlorophyll a, leaf net photosynthetic rate, water use efficiency, transpiration rate, and stomatal conductance under the T1 treatment compared to the CK treatment; soluble sugar, soluble protein and free amino acid contents in eggplant fruit increased by 10.8, 12.3 and 6.7%, respectively; and yield increased by 3.9%. Our research proved that the 10% irrigation reduction treatment (T1) could improve microbial community richness and fruit yield, which would improve irrigation efficiency and cost reduction in agriculture.

Project description:Salinization poses a significant challenge in agriculture, exacerbated by anthropogenic global warming. Biostimulants, derived from living microorganisms or natural extracts, have emerged as valuable tools for conventional and organic agriculture. However, our understanding of the molecular mechanisms underlying the effects of biostimulants is very limited, especially in crops under real cultivation conditions. In this study, we adopted an integrative approach to investigate the effectiveness of the combined application of plant growth-promoting bacterium (Bacillus megaterium strain BM08) and a non-microbial biostimulant under control conditions (normal watering) and salt stress. After confirming the yield increase under both conditions, we investigated the molecular mechanisms underlying the observed effect by measuring a number of physiological parameters (i.e., lipid peroxidation, antioxidants, chlorophylls, total phenolics and phytohormone content), as well as RNA sequencing and primary metabolite analyses. Our findings reveal that the combined effect of the microbial and non-microbial biostimulants led to a decrease in the antioxidant response and an up-regulation of genes involved in cytokinin biosynthesis under salt stress conditions. This, in turn, resulted in a higher concentration of the bioactive cytokinin, isopentenyladenosine, in roots and leaves and an increase in γ-aminobutyric acid, a non-proteic amino acid related to abiotic stress responses. In addition, we observed a decrease in malic acid, along with an abscisic acid (ABA)-independent up-regulation of SR-kinases, a family of protein kinases associated with abiotic stress responses. Furthermore, we observed that the single application of the non-microbial biostimulant triggers an ABA-dependent response under salt stress; however, when combined with the microbial biostimulant, it potentiated the mechanisms triggered by the BM08 bacterial strain. This comprehensive investigation shows that the combination of two biostimulants is able to elicit a cytokinin-dependent response that may explain the observed yield increase under salt stress conditions.

Project description:Salinity in water and soil is one of the major environmental factors limiting the productivity of agronomic and horticultural crops. In basil (Ocimum basilicum L., Lamiaceae) and other Ocimum species, information on the plant response to mild salinity levels, often induced by the irrigation or fertigation systems, is scarce. In the present work, we tested the effectiveness of a microbial-based biostimulant containing two strains of arbuscular mycorrhiza fungi (AMF) and Trichoderma koningii in sustaining greenhouse basil yield traits, subjected to two mild salinity stresses (25 mM [low] and 50 mM [high] modulated by augmenting the fertigation osmotic potential with NaCl) compared to a non-stressed control. The impact of salinity stress was further appraised in terms of plant physiology, morphological ontogenesis and composition in polyphenols and volatile organic compounds (VOC). As expected, increasing the salinity of the solution strongly depressed the plant yield, nutrient uptake and concentration, reduced photosynthetic activity and leaf water potential, increased the Na and Cl and induced the accumulation of polyphenols. In addition, it decreased the concentration of Eucalyptol and β-Linalool, two of its main essential oil constituents. Irrespective of the salinity stress level, the multispecies inoculum strongly benefited plant growth, leaf number and area, and the accumulation of Ca, Mg, B, p-coumaric and chicoric acids, while it reduced nitrate and Cl concentrations in the shoots and affected the concentration of some minor VOC constituents. The benefits derived from the inoculum in term of yield and quality harnessed different mechanisms depending on the degree of stress. under low-stress conditions, the inoculum directly stimulated the photosynthetic activity after an increase of the Fe and Mn availability for the plants and induced the accumulation of caffeic and rosmarinic acids. under high stress conditions, the inoculum mostly acted directly on the sequestration of Na and the increase of P availability for the plant, moreover it stimulated the accumulation of polyphenols, especially of ferulic and chicoric acids and quercetin-rutinoside in the shoots. Notably, the inoculum did not affect the VOC composition, thus suggesting that its activity did not interact with the essential oil biosynthesis. These results clearly indicate that beneficial inocula constitute a valuable tool for sustaining yield and improving or sustaining quality under suboptimal water quality conditions imposing low salinity stress on horticultural crops.
