The identity of Sasaoblongula C.H.Hu (Poaceae, Bambusoideae, Arundinarieae): evidence from morphology and molecular data.

ABSTRACT: Sasaoblongula was described in 1987 based on a cultivated plant at the bamboo garden of Sun Yat-sen University. This species has two or three branches at the upper nodes, which differ from the rest of Sasa species that have a single branch per node. During the field trip to Baishi Town, Yunfu City, Guangdong Province in July 2021, one bamboo species with oblong foliage leaves was collected and matches the isotype. Then, our question was to test the identity of S.oblongula concerning other Sasa species based on morphology and molecular data. To do that, we sequenced the whole chloroplast genome of S.oblongula and did a phylogenetic analysis. Our morphological results indicate that the new collection is S.oblongula. The phylogenetic tree showed that S.oblongula is close to Pseudosasa, instead of Sasa species. Therefore, we transferred it to the genus Pseudosasa, and a revised description of P.oblongula is provided here.

Project description:The genus Gelidocalamus T. H. Wen, endemic to southern China, is a small but taxonomically problematic genus of Arundinarieae (Poaceae, Bambusoideae). During field work, a population of Gelidocalamus from Zixing, Hunan, was discovered, appearing to be distinct from our previously identified collection. Comparisons of the population of Zixing were performed by using scanning electron microscopy (SEM) and a plastid genome-based phylogeny. Morphologically, it was mostly similar to G.multifolius, but differed by culm leaf erect with densely white pubescence, apical branch sheath much longer than the internodes and foliage leaf larger. Phylogenetically, the new species was well-supported as a sister to the clade of G.multifolius + G.tessellatus, and the above three taxa were clustered in the Shibataea clade (IV) of Arundinarieae. Thus, the new species, formally named as Gelidocalamuszixingensis W.G.Zhang, G.Y.Yang & C.K.Wang, was described and illustrated herein.

The complete chloroplast genome of Pseudosasa usawae (Poaceae: Bambusoideae: Arundinarieae) in Taiwan.

Project description:Pseudosasa usawae is an endemic species in Taiwan, and grows at an altitude of 600-1200 m. In this study, we fully characterized the complete chloroplast genome of P. usawae. The complete chloroplast sequence was 139,660 bp, including large single-copy (LSC), small single-copy (SSC), and a pair of invert repeats (IR) region of 83,271, 12,803, and 21,793 bp. Besides, the plastid genome comprised a total of 129 genes, including protein-coding, tRNA, and rRNA genes as 83, 38, and 8 genes. Phylogenetic analysis reveals that P. usawae is closely associated with Phyllostachys genus clade, sister to the lineage of Phyllostachys.

The complete chloroplast genome and phylogenetic position of Thamnocalamus unispiculatus (Poaceae: Bambusoideae: Arundinarieae).

Project description:Thamnocalamus unispiculatus T.P.Yi & J.Y.Shi 2007 is an important bamboo species with significant ecological and economic value. This study presents the complete chloroplast genome sequence of T. unispiculatus. The sequence was 139,726 bp in length and exhibited a typical quadripartite structure, containing four regions: large single copy regions (LSC, 83,283 bp), small single copy regions (SSC, 12,851 bp) and a pair of inverted repeats (IRs, 21,726 bp). A total of 130 genes were annotated, including 86 protein-coding genes, 36 transfer RNA genes, and eight ribosomal RNA genes. Phylogenetic analysis indicated that T. unispiculatus and T. spathiflorus are sister species, supporting the conclusion that Thamnocalamus is a monophyletic group. The chloroplast genome of T. unispiculatus promotes the protection and exploration of biodiversity, phylogenetic relationships, and genetic research in Bambusoideae.

A new genus of temperate woody bamboos (Poaceae, Bambusoideae, Arundinarieae) from a limestone montane area of China.

Project description:Ampelocalamuscalcareus is a climbing and slender bamboo, known from south Guizhou, China. This species grows in broadleaved forests of limestone montane areas. Recent molecular phylogenetic analyses demonstrated that A.calcareus was sister to all other lineages of the tribe Arundinarieae rather than a member of Ampelocalamus. The morphological features and habitats of A.calcareus and related genera including Ampelocalamus, Drepanostachyum and Himalayacalamus were compared and discussed. The characteristics of the branch complements, nodes and foliage leaves distinguish A.calcareus from morphologically similar taxa. On the basis of molecular and morphological evidence, we propose to establish a new genus, Hsuehochloa, to accommodate A.calcareus and to honour the late Chinese bamboo taxonomist Chi-Ju Hsueh (Ji-Ru Xue). In addition, we describe the inflorescence of Hsuehochloa for the first time.

Genome skimming reveals the complete chloroplast genome of Ampelocalamus naibunensis (Poaceae: Bambusoideae: Arundinarieae) with phylogenomic implication.

Project description:Ampelocalamus naibunensis is one drooping bamboo with an important ornamental value endemic to Taiwan Island. To date, the genetic and genomic information of this species is little known. Here we characterized the complete chloroplast genome of A. naibunensis using genome skimming approach. The complete chloroplast genome is 139,860 bp, with a large single copy region (LSC) of 83,380 bp and a small single copy region (SSC) of 13,014 bp separated by a pair of inverted repeats (IRs) of 21,822 bp. The genome encodes a total of 129 genes, of which 111 are unique, containing 76 protein-coding genes, 4 ribosomal RNAs and 31 transfer RNAs. Sixteen distinct genes contain one or two introns, and the GC content of the cp genome is 38.9%. Phylogenomic analysis strongly supports the placement of A. naibunensis in the Chimonocalamus lineage (III), distantly related to A. calcareus (XI) within temperate woody bamboos.

Chimonobambusasangzhiensis (Poaceae, Bambusoideae), a new combination supported by morphological and molecular evidence.

Project description:This study elucidates the taxonomic position of Indosasasangzhiensis in considering whether it belongs to Indosasa or Chimonobambusa. Based on morphological and molecular phylogenetic evidence, our results explicitly indicated that I.sangzhiensis should be a member of Chimonobambusa, rather than Indosasa, and is a distinct species closely related to C.communis, C.opienensis and C.puberula. Thus, the new combination Chimonobambusasangzhiensis (B.M.Yang) N.H.Xia & Z.Y.Niu is made. A detailed description as well as two color plates of this species are also provided.

The identity of Dinochloa species and enumeration of Melocalamus (Poaceae: Bambusoideae) in China.

Project description:Three woody bamboo species collected in Hainan, China in 1940 have been described as Dinochloa based on vegetative specimens. However, the identity of these species has long been in doubt, largely because the vegetative phase in species of Dinochloa is morphologically similar to that in species of Melocalamus, a climbing or scrambling bamboo genus of the paleotropical woody bamboos (Poaceae: Bambusoideae) that consists of about 15 species and one variety. To determine the phylogenetic affinity of the three Dinochloa species from Hainan, we sampled almost all recognized Chinese species of Melocalamus and representative species of Dinochloa as well as other closely related genera, performed molecular phylogenetic analysis, and compared their morphology based on herbarium and fieldwork investigation. Our ddRAD data indicate that the three species from Hainan are closely related to Melocalamus, not Dinochloa. Morphological analysis showed that these three species have a climbing habit but do not grow spirally, their culm leaves have smooth bases, and there is a ring of powder and/or tomenta above and below the nodes. Taken together our findings indicate that the three species from Hainan originally published in Dinochloa should be transferred to Melocalamus, i.e., Melocalamus orenudus (McClure) D.Z. Li & J.X. Liu, Melocalamus puberulus (McClure) D.Z. Li & J.X. Liu, and Melocalamus utilis (McClure) D.Z. Li & J.X. Liu, respectively. This study concludes with an enumeration of Chinese species of Melocalamus, with a key to nine recognized species and one variety, and a lectotypification for M. compatiflorus.

Sinosasagracilis (Poaceae, Bambusoideae), a new combination supported by morphological and phylogenetic evidence.

Project description:The results of phylogenetic analysis, based on the whole chloroplast genome and morphological study support the transfer of a long ignored bamboo species, Sasagracilis, to the recently established genus, Sinosasa, in this study. Morphologically, this species differs from all the other known Sinosasa species by having very short (2-3 mm) foliage leaf inner ligules, which is unusual in this genus. A revised description of its morphology and colour photos are also provided.

Distribution of Holttumochloa (Poaceae: Bambusoideae) in China with description of a new species revealed by morphological and molecular evidence.

Project description:Holttumochloa has previously only been recorded from Malaysia. Here we describe and illustrate a new species, Holttumochloa hainanensis sp. nov., from the lowland montane forests of Diaoluo Mountain on the Island of Hainan, South China. Morphologically, H. hainanensis is similar to Holttumochloa korbuensis, but can be clearly distinguished from it in having larger culms covered by white wax, longer leaf blades, larger pseudospikelets and anthers. Furthermore, molecular phylogeny based on the nuclear gene GBSSI corroborates the identification of the new species and its affinity. The biogeographical significance of the new record of Holttumochloa in South China is also highlighted in this study.

Project description:Due to their specialised flowering biology where frequent or even annual flowering is uncommon, reproductive materials of bamboos are not always available, so hampering taxonomic interpretation and research into other aspects. Bambusacontracta, B.corniculata, B.cornigera and B.subtruncata were established only based on vegetative materials and flowering or fruiting material has been hitherto unknown. The floral morphology of these four species is described for the first time and, correspondingly, epitypes are designated to support a more complete interpretation of the species.
