Two new species of genus Leucoagaricus (Agaricaceae, Agaricales) from Pakistan.

ABSTRACT: The genus of basidiomycetous fungi, Leucoagaricus, occurs worldwide, from subtropical to boreal latitudes. Several collections of Leucoagaricus were made during mycological field trips conducted in different forests of Margalla, Pakistan. An integrative framework combining morphological and phylogenetic data was employed for their study. As a result, the two species La.margallensis and La.glareicolor are here described as new to science. Detailed macro- and micro-morphological descriptions, and a molecular phylogenetic reconstruction based on nrITS and LSU sequence data are provided and used to discriminate the new species from morphologically and phylogenetically close taxa. Whereas, our phylogenetic tree inference gave unequivocal support for the inclusion of these two species within the section Leucoagaricus.

Two new species of Micropsalliota (Agaricales, Agaricaceae) from subtropical regions of China.

Project description:Micropsalliota is a relatively small genus, with 97 names recorded in previous research. In this study, two new species of Micropsalliota, M.bispora and M.vulgaris, have been identified based on morphological and phylogenetic evidence from subtropical regions of China. Morphologically, M.bispora is characterized by very small basidiomata, cymbiform basidiospores up to 9.0 μm long, white to cream pileus with fawn to dull red center, and tibiiform cheilocystidia; M.vulgaris is recognized by small basidiomata, medium-sized spores, white to cream pileus, covered with reddish brown to dark brown fibrils, various cheilocystidia that are up to 60 μm long, and hyphae of fibrils exhibiting pale brown vacuolar pigmentation. Their distinct taxonomic status is confirmed by the positions of the two new species in 4-locus (ITS, LSU, rpb2, tef-1α) phylogenetic trees. Detailed descriptions and morphological photographs of two new species are presented. To aid in diagnosis, a key to 35 species of Micropsalliota in China is provided.

The genus Coprinellus (Basidiomycota; Agaricales) in Pakistan with the description of four new species.

Project description:Mushrooms with a thin-fleshed pileus that becomes plicate on opening, deliquescent lamellae and dark brown to blackish basidiospores are commonly called coprinoid mushrooms. The genus Coprinellus is one of the important lineages of coprinoid mushroom in the family Psathyrellaceae. Species-level taxonomy in Coprinellus is based mainly on the presence or absence and the structure of veil and cystidia on the pileus, of cystidia on the lamellae and on basidiospore morphology. In this study, four new species of Coprinellus (Co.campanulatus, Co.disseminatus-similis, Co.pakistanicus and Co.tenuis) are described from Pakistan. Species descriptions are based on morphological and molecular data. Phylogenetic analyses based on nuc rDNA ITS region show that the new species Co.campanulatus and Co.disseminatus-similis are clustered in a clade including members of section Micacei; Co.tenuis falls in a clade with members of section Domestici; and Co.pakistanicus recovered in a separate clade adjacent to other recently described clades of genus Coprinellus. Morpho-anatomical descriptions of the new species and comparison with closely allied taxa are provided. With this study, the number of known species of Coprinellus in Pakistan has reached eight.

Taxonomy and multi-gene phylogeny of Micropsalliota (Agaricales, Agaricaceae) with description of six new species from China.

Project description:Micropsalliota is a relatively small genus containing only 62 previously identified species. Here, we describe six new taxa of Micropsalliota based on morphological and phylogenetic analyses: M. minor, M. ovalispora, M. pseudodelicatula, M. rufosquarrosa, M. tenuipes, and M. wuyishanensis and a new record taxon to China. The first Maximum likelihood and Bayesian analyses of a three-gene dataset (ITS, LSU, and rpb2) separated the genus into 18 weakly to strongly supported major clades and subclades, but only a few subclades were synapomorphies. According to phylogenetic analyses, M. cornuta does not belong in Micropsalliota. A key to 20 species of Micropsalliota in China is provided.

Taxonomy and Phylogeny of Cystolepiota (Agaricaceae, Agaricales): New Species, New Combinations and Notes on the C. seminuda Complex.

Project description:Species of Cystolepiota are known as diminutive lepiotaceous fungi with a worldwide distribution. Previous studies revealed that Cystolepiota is not monophyletic and preliminary DNA sequence data from recent collections suggested that several new species exist. Based on multi-locus DNA sequence data (the nuc rDNA internal transcribed spacer region ITS1-5.8S-ITS2, ITS; the D1-D2 domains of nuc 28S rDNA, LSU; the most variable region of the second-largest subunit of RNA polymerase II, rpb2 and a portion of the translation-elongation factor 1-α. tef1), C. sect. Pulverolepiota forms a distinct clade separating from Cystolepiota. Therefore, the genus Pulverolepiota was resurrected and two combinations, P. oliveirae and P. petasiformis were proposed. With the integration of morphological characteristics, multi-locus phylogeny, and information on geography and habitat, two new species, viz. C. pseudoseminuda and C. pyramidosquamulosa, are described and C. seminuda was revealed to be a species complex containing at least three species, viz. C. seminuda, C. pseudoseminuda, and Melanophyllum eryei. In addition, C. seminuda was re-circumscribed and neo-typified based on recent collections.

Looking for Lepiotapsalion Huijser & Vellinga (Agaricales, Agaricaceae).

Project description:Lepiotapsalion is fully described based on a recent collection from Sardinia (Italy) and the holotype. NrITS- and nrLSU-based phylogeny demonstrates that sequences deposited in GenBank as "L.psalion" and generated from two Dutch and one Chinese collections are not conspecific with the holotype and represent two distinct, undescribed species. These species are here proposed as Lepiotarecondita sp. nov. and Lepiotasinorecondita ad int.

The genus Parasola in Pakistan with the description of two new species.

Project description:Parasola is a genus of small, veil-less coprinoid mushrooms in the family Psathyrellaceae (Agaricales). The genus is not well documented in Asia, specifically in Pakistan. In this study we describe two new species Parasola glabra and P. pseudolactea from Pakistan, based on morphological and molecular data. Phylogeny based on three DNA regions: nuc rDNA region encompassing the internal transcribed spacers 1 and 2 along with the 5.8S rDNA (ITS), nuc 28S rDNA D1-D2 domains (28S) and translation elongation factor 1α gene (TEF1α) show that the new taxa are clustered in a clade formed by the members of section Parasola of genus Parasola. Parasola glabra with grayish pileus, slightly depressed pileal disc, lamellae separated from the stipe by pseudocollarium, basidiospores 14.5-16.5 × 9.5-11.5 × 8.0-10.5 µm, in front view broadly ovoid to oblong, some with rhomboidal outline, in side view ellipsoid, with eccentric germ-pore of 1.5 µm diameter. Parasola pseudolactea with yellowish brown to dull brown pileus, disc indistinctly umbonate, lamellae free, pseudocollarium absent, basidiospores 13.5-14.5 × 10.5-12.0 × 9.5-10.5 µm, in face view rounded triangular to heart shaped, rarely ovoid to subglobose, in side view ellipsoid to oblong, with eccentric germ-pore of 1.5 µm diam. In addition to these new species, P. auricoma and P. lilatincta were also studied. Morphological descriptions for the new species and comparison with known Parasola species are provided. Our observations highlight the diversity of Parasola in northern Pakistan and further document the need for additional systematic focus on the region's fungi.

A study on the genus Candolleomyces (Agaricales: Psathyrellaceae) from Punjab, Pakistan.

Project description:Many basidiomata of the genus Candolleomyces were found on sandy and loamy soil from the Indus Riverbed, Kot Addu District. A phylogenetic study was conducted to examine the occurrence of Candolleomyces sindhudeltae sp. nov. using a combination of ITS and LSU regions. Our morphological, anatomical, and phylogenetic studies indicated the novelty of Candolleomyces sindhudeltae sp. nov. The distinguishing features of C. sindhudeltae are convex to campanulate and areolate pileus with scalloped to cracked cap margins, branched, and pale reddish lamellae, greenish-brown ellipsoid to ovoid basidiospores, polymorphic cheilo, and caulocystidia. The novel taxa formed independent phylogenetic relationships within the genus Candolleomyces. The addition of our new species to the genus Candolleomyces makes us confident that the genus was separated from Psathyrella accurately.

First record of the rare genus Typhrasa (Psathyrellaceae, Agaricales) from China with description of two new species.

Project description:Typhrasa is a rare genus that comprises two species and that has previously been reported only from Europe and North America. The present study expands the geographical scope of the genus by describing two new species - T. polycystis and T. rugocephala - from subtropical China. The new species are supported by morphological characteristics and phylogenetic analyses (ITS, LSU and tef-1α). The new species have very similar morphological characteristics and are 98% similar in their ITS region. However, T. rugocephala has two types of long gills at the same time, rarely fusiform pleurocystidia with rostrum. Detailed descriptions, colour photos, illustrations and a key to related species are presented in this paper.

Taxonomy and Phylogeny of Cystostereaceae (Agaricales, Basidiomycota): A New Genus, Five New Species, and Three New Combinations.

Project description:This paper aims to understand the species diversity, taxonomy, and phylogeny of Cystostereaceae (Agaricales), which is based primarily on material from East and Southeast Asia. Cystostereaceae is a small, understudied family of saprobes of woody plants with a worldwide distribution. Phylogenetic analyses of the LSU and ITS sequences revealed four distinct clades in the Cystostereaceae, representing the genera Crustomyces, Cystostereum, Effusomyces gen. nov., and Parvodontia. In addition, phylogenetic analyses showed that Cystidiodontia and Rigidotubus are synonyms of Crustomyces for their type of species nested within the Crustomyces clade. The new monotypic genus Effusomyces, based on specimens from Thailand, lacks any distinctive morphological features. Parvodontia, originally erected for a species from South America, is reported for the first time from Asia. The widely distributed and morphologically well-characterized Cystostereum is represented in East Asia by two new species: Cystostereum crassisporum and C. submurrayi. In addition, three new species, viz., Crustomyces albidus, Effusomyces thailandicus, and Parvodontia austrosinensis, are described and illustrated. Finally, three new combinations are proposed: Crustomyces isabellinus, C. laminiferus, and C. tephroleucus. A key to the genera and species of Cystostereaceae is provided. Our results proved that the species diversity of wood-decaying fungi in East and Southeast Asia is rich and suggested that more investigations and studies should be carried out in the future.
