Temperature changes in the root ecosystem affect plant functionality.

ABSTRACT: Climate change is increasing the frequency of extreme heat events that aggravate its negative impact on plant development and agricultural yield. Most experiments designed to study plant adaption to heat stress apply homogeneous high temperatures to both shoot and root. However, this treatment does not mimic the conditions in natural fields, where roots grow in a dark environment with a descending temperature gradient. Excessively high temperatures severely decrease cell division in the root meristem, compromising root growth, while increasing the division of quiescent center cells, likely in an attempt to maintain the stem cell niche under such harsh conditions. Here, we engineered the TGRooZ, a device that generates a temperature gradient for in vitro or greenhouse growth assays. The root systems of plants exposed to high shoot temperatures but cultivated in the TGRooZ grow efficiently and maintain their functionality to sustain proper shoot growth and development. Furthermore, gene expression and rhizosphere or root microbiome composition are significantly less affected in TGRooZ-grown roots than in high-temperature-grown roots, correlating with higher root functionality. Our data indicate that use of the TGRooZ in heat-stress studies can improve our knowledge of plant response to high temperatures, demonstrating its applicability from laboratory studies to the field.

Project description:BackgroundGreen roofs perform ecosystem services such as summer roof temperature reduction and stormwater capture that directly contribute to lower building energy use and potential economic savings. These services are in turn related to ecosystem functions performed by the vegetation layer such as radiation reflection and transpiration, but little work has examined the role of plant species composition and diversity in improving these functions.Methodology/principal findingsWe used a replicated modular extensive (shallow growing- medium) green roof system planted with monocultures or mixtures containing one, three or five life-forms, to quantify two ecosystem services: summer roof cooling and water capture. We also measured the related ecosystem properties/processes of albedo, evapotranspiration, and the mean and temporal variability of aboveground biomass over four months. Mixtures containing three or five life-form groups, simultaneously optimized several green roof ecosystem functions, outperforming monocultures and single life-form groups, but there was much variation in performance depending on which life-forms were present in the three life-form mixtures. Some mixtures outperformed the best monocultures for water capture, evapotranspiration, and an index combining both water capture and temperature reductions. Combinations of tall forbs, grasses and succulents simultaneously optimized a range of ecosystem performance measures, thus the main benefit of including all three groups was not to maximize any single process but to perform a variety of functions well.Conclusions/significanceEcosystem services from green roofs can be improved by planting certain life-form groups in combination, directly contributing to climate change mitigation and adaptation strategies. The strong performance by certain mixtures of life-forms, especially tall forbs, grasses and succulents, warrants further investigation into niche complementarity or facilitation as mechanisms governing biodiversity-ecosystem functioning relationships in green roof ecosystems.

Project description:The process and function that underlie the assembly of root-associated microbiomes may be strongly linked to the survival strategy of plants. However, the assembly and functional changes of root-associated microbial communities in different desert plants in natural desert ecosystems are still unclear. Thus, we studied the microbial communities and diversity of root endosphere (RE), rhizosphere soil (RS), and bulk soil (BS) among three representative desert plants (Alhagi sparsifolia, Tamarix ramosissima, and Calligonum caput-medusae) in three Xinjiang desert regions {Taklimakan (CL), Gurbantünggüt (MSW), and Kumtag (TLF)} in China. This study found that the soil properties {electrical conductivity (EC), soil organic carbon (SOC), total nitrogen (TN) and phosphorus (TP), available nitrogen (AN) and phosphorus (AP)} of C. caput-medusae were significantly lower than those of A. sparsifolia and T. ramosissima, while the root nutrients (TN and TP) of A. sparsifolia were significantly higher compared to C. caput-medusae and T. ramosissima. The beta diversity of bacteria and fungi (RE) among the three desert plants was significantly different. The common OTU numbers of bacteria and fungi in three compartments (RE, RS, and BS) of the three desert plants were ranked as RS > BS > RE. The bacterial and fungal (RE) Shannon and Simpson indexes of C. caput-medusae were significantly lower as compared to those of A. sparsifolia and T. ramosissima. Additionally, bacterial and fungal (RE and RS) node numbers and average degree of C. caput-medusae were lower than those found in A. sparsifolia and T. ramosissima. Root and soil nutrients collectively contributed to the composition of root-associated bacterial (RE, 12.4%; RS, 10.6%; BS, 16.6%) and fungal communities (RE, 34.3%; RS, 1.5%; BS, 17.7%). These findings demonstrate variations in the bacterial and fungal populations across different plant species with distinct compartments (RE, RS, and BS) in arid environments. More importantly, the study highlights how much soil and plant nutrients contribute to root-associated microbial communities.

Project description:Soil and microbial biomass carbon (C), nitrogen (N), and phosphorus (P) play an important role in soil nutrient dynamics in biogeochemical cycles of terrestrial ecosystems. However, increased human activities as a result of agricultural intensification on soil nutrients and microbial C:N:P stoichiometry are poorly understood in this fragile forest-savanna transition agroecosystem. This study aimed to (i) assess soil and microbial C, N, and P stoichiometry in different land use systems, and (ii) examine the effect of soil and microbial C, N, and P stoichiometry on soils susceptible to human-induced land use changes. A total of 82 composite soil samples at a depth of 0-20 cm were sampled from forest, savanna, grassland, fallow and cropland for laboratory analysis. The results revealed that the concentrations of C, N, and P were low in Fallow and Cropland compared to other land use systems. Analysis of variance in microbial C, N, and P stoichiometric ratios revealed a significant decreasing tendency compared to soil C:N, C:P and N:P ratios with no statistical significance (p < 0.05). The C:P and N:P ratios were low compared to the C:N ratio in land uses. A significant positive correlation was observed between MBC and MBN (0.95; p < 0.01), and with C and N (0.69; p < 0.01). There were significant interactive effects of land use on soil and microbial variables. The estimated microbial C:N:P stoichiometric ratios (21:2:1) were well constrained in the study area. The transition from Forest to Cropland resulted in 64%, 52%, and 71% reduction in C, N, and P, respectively. This implies that phosphorus is the main factor limiting productivity. The low availability of phosphorus in these tropical soils may have resulted in low C:P and N:P ratios. Therefore, we conclude that our results highlight the importance of phosphorus limitation on ratios of microbial C:P and N:P in landuse systems. Nutrient inputs such as fertilizers, manure and crop residues should be applied to croplands to improve soil and microbial C, N and P levels. Further, effects of land use on soil nutrient status and stoichiometry at 1-meter depth will be considered in our future work.
