Transcriptomic and metabolic studies on the role of inorganic and organic iodine compounds in lettuce plants.

ABSTRACT: Iodine (I) is considered a beneficial element or even micronutrient for plants. The aim of this study was to determine the molecular and physiological processes of uptake, transport, and metabolism of I applied to lettuce plants. KIO₃, KIO₃ + salicylic acid, 5-iodosalicylic acid and 3,5-diiodosalicylic acid were applied. RNA-sequencing was executed using 18 cDNA libraries constructed separately for leaves and roots from KIO₃, SA and control plants. De novo transcriptome assembly generated 1937.76 million sequence reads resulting in 27,163 transcripts with N50 of 1638 bp. 329 differentially expressed genes (DEGs) in roots were detected after application of KIO₃, out of which 252 genes were up-regulated, and 77 were down-regulated. In leaves, 9 genes revealed differential expression pattern. DEGs analysis indicated its involvement in such metabolic pathways and processes as: chloride transmembrane transport, phenylpropanoid metabolism, positive regulation of defense response and leaf abscission, and also ubiquinone and other terpenoid-quinone biosynthesis, protein processing in endoplasmic reticulum, circadian rhythm including flowering induction as well as a putative PDTHA (i.e. Plant Derived Thyroid Hormone Analogs) metabolic pathway. qRT-PCR of selected genes suggested their participation in the transport and metabolism of iodine compounds, biosynthesis of primary and secondary metabolites, PDTHA pathway and flowering induction.

Project description:A human's diet should be diverse and rich in vitamins, macro- and microelements essential for the proper functioning of the human body. Globally, a high percentage of the human population suffers from malnutrition, deficiencies of nutrients and vitamins also known as the problem of hidden hunger. This problem it is not only common in poor countries, but also occurs in developed countries. Iodine is a nutrient crucial for the proper functioning of the human and animal body. For plants, it is referred to as a beneficial element or even a microelement. The design of the biofortification experiment was determined on the basis of the interaction of iodine and vanadium (synergistic interaction in marine algae), where vanadium-dependent iodoperoxidase catalyzes apoplastic oxidation of iodine, resulting in high efficiency of iodine uptake and accumulation in brown algae (Laminaria digitate). Three independent experiments (Exp.) were carried out with the foliar application of vanadium (V) and iodine (I) compounds. The main differences between the experiments with the adapted proper corn biofortification method were the different application stage between the individual experiments, the application intervals and the dose of the iodine-vanadium compound. In each experiment, the accumulation of iodine and vanadium in the grain was several times lower than in the leaves. The combination iodine and vanadium significantly increased the accumulation of iodine in the grain in the case of applying V with inorganic iodine compounds, and a decrease in the accumulation of I after applying V with organic iodine compound -especially in Exp. No. 3. In grain, the highest content of I-, IO3- was in combination with the application of 2-iodobenzoic acid (products of its metabolism). In most of the tested combinations, vanadium stimulated the accumulation/synthesis of exogenous/endogenous 5-iodosalicylic acid (5ISA) and 2-iodobenzoic acid (2IBeA), respectively, and decreased the content of 2,3,5-triiodobenzoic acid (2,3,5-triIBeA) in leaves and grains. The tested compounds I and V and the combinations of their application had a diversified effect on the vitamin C content in the grains. Vanadium in the lower dose of 0.1 µM significantly increased the sugar content in the grain.

Project description:Sarracenia pitcher plants display interspecific differences in prey, so far only explained by pitcher morphology. We hypothesized that pitcher odours play a role in prey composition. We first compared odour and prey compositions among Sarracenia taxa grown together, forming a kinship gradient from S. purpurea known to capture primarily ants towards S. leucophylla known to capture many flying insects: S. purpurea, S. X mitchelliana, and S. X Juthatip soper & S. X leucophylla horticultural hybrids. We then measured several pitcher traits to disentangle the contributions of morphology and odour to prey variation. The pitcher odours were as diverse as those of generalist-pollinated flowers but with notable differences among taxa, reflecting their relatedness. VOC similarity analyses revealed taxon specificities, that mirrored those revealed by prey similarity analyses. S. X leucophylla stood out by being more specialised in flying insects like bees and moths and by releasing more monoterpenes known to attract flower visitors. S. X Juthatip soper trapped as many bees but fewer moths, sesquiterpenes contributing less to its scent. Ants and Diptera were the main prey of the other two with fatty-acid-derivative-dominated scents. Quantities of the different prey groups can be inferred 98% from quantities of the odour classes and pitcher dimensions. Two syndromes were revealed: ants associated with fatty-acid-derivatives and short pitchers; flying insects associated with monoterpenes, benzenoids and tall pitchers. In S. X leucophylla, emission rate of fatty-acid-derivatives and pitcher length explained most variation in ant captures; monoterpenes and pitcher length explained most variation in bee and moth captures; monoterpenes alone explained most variation in Diptera and wasp captures. Our results suggest that odours are key factors of the diet composition of pitcher plants. They support the hypothesis of perceptual exploitation of insect biases in carnivorous plants and provide new insights into the olfactory preferences of insect groups.
