Schiff Bases Functionalized with T-Butyl Groups as Adequate Ligands to Extended Assembly of Cu(II) Helicates.

ABSTRACT: The study of the inherent factors that influence the isolation of one type of metallosupramolecular architecture over another is one of the main objectives in the field of Metallosupramolecular Chemistry. In this work, we report two new neutral copper(II) helicates, [Cu₂(L¹)₂]·4CH₃CN and [Cu₂(L²)₂]·CH₃CN, obtained by means of an electrochemical methodology and derived from two Schiff-based strands functionalized with ortho and para-t-butyl groups on the aromatic surface. These small modifications let us explore the relationship between the ligand design and the structure of the extended metallosupramolecular architecture. The magnetic properties of the Cu(II) helicates were explored by Electron Paramagnetic Resonance (EPR) spectroscopy and Direct Current (DC) magnetic susceptibility measurements.

In Situ Assembled Polynuclear Zinc Oxo Clusters Using Modified Schiff Bases as Ligands.

Project description:A series of different cores and nuclearity zinc metal clusters 1-5 have been synthesized using Zn(ClO4)2·6H2O, Schiff-base primary ligands, and dibenzoyl methane (DBM) or monoethanolamine (MEA) as co-ligand in a room-temperature reaction. The structure of the complexes is characterized using single-crystal X-ray diffraction. Among them, (1) [Zn(L1)(DBM)] is mononuclear; (2) [Zn4(L2)2(DBM)4], (3) [Zn4(L2)4(H2O)2(ClO4)2]·2CH2Cl2, and (4) [Zn4(L3)2(DBM)4] have a cubane core; and (5) [Zn4(L4)4(MEA)2(ClO4)2] has a ladderlike core structure. Compounds 1-5 have also been characterized using UV-vis absorption and emission spectroscopies. For an in-depth understanding of the absorption spectra of 1 and 3, density functional theory (DFT) calculations have been performed, which suggest that the transitions correspond to the π → π* intraligand charge transfer (ILCT) transitions.

Spectroscopic Characterization and Biological Activity of Hesperetin Schiff Bases and Their Cu(II) Complexes.

Project description:The three Schiff base ligands, derivatives of hesperetin, HHSB (N-[2,3-dihydro-5,7-dihydroxy-2-(3-hydroxy-4-methoxyphenyl)chromen-4-ylidene]isonicotinohydrazide), HIN (N-[2,3-dihydro-5,7-dihydroxy-2-(3-hydroxy-4-methoxyphenyl)chromen-4-ylidene]benzhydrazide) and HTSC (N-[2,3-dihydro-5,7-dihydroxy-2-(3-hydroxy-4-methoxyphenyl)chromen-4-ylidene]thiosemicarbazide) and their copper complexes, CuHHSB, CuHIN, and CuHTSC were designed, synthesized and analyzed in terms of their spectral characterization and the genotoxic activity. Their structures were established using several methods: elemental analysis, FT-IR, UV-Vis, EPR, and ESI-MS. Spectral data showed that in the acetate complexes the tested Schiff bases act as neutral tridentate ligand coordinating to the copper ion through two oxygen (or oxygen and sulphur) donor atoms and a nitrogen donor atom. EPR measurements indicate that in solution the complexes keep their structures with the ligands remaining bound to copper(II) in a tridentate fashion with (O-, N, Oket) or (O-, N, S) donor set. The genotoxic activity of the compounds was tested against model tumour (HeLa and Caco-2) and normal (LLC-PK1) cell lines. In HeLa cells the genotoxicity for all tested compounds was noticed, for HHSB and CuHHSB was the highest, for HTSC and CuHTSC-the lowest. Generally, Cu complexes displayed lower genotoxicity to HeLa cells than ligands. In the case of Caco-2 cell line HHSB and HTSC induced the strongest breaks to DNA. On the other side, CuHHSB and CuHTSC induced the highest DNA damage against LLC-PK1.

Triaryl-substituted Schiff bases are high-affinity subtype-selective ligands for the estrogen receptor.

Project description:We have explored the isoelectronic replacement of the C═C double bond found at the core of many nonsteroidal estrogen ligands with a simple Schiff base (C═N). Di- and triaryl-substituted imine derivatives were conveniently prepared by the condensation of benzophenones with various anilines without the need for phenolic hydroxy protection. Most of these imines demonstrated high affinity for the estrogen receptors, which, in some cases exceeded that of estradiol. In cell-based assays, these imines profiled as ERα agonists but as ERβ antagonists, showing preferential reliance on the N-terminal activation function (AF1), which is more active in ERα. X-ray analysis revealed that the triaryl-imines distort the ligand-binding pocket in a new way: by controlling the separation of helices 3 and 11, which appears to alter the C-terminal AF2 surface that binds transcriptional coactivators. This work suggests that C═N for C═C substitution might be more widely considered as a general strategy for preparing drug analogues.

Cathecol and Naphtol Groups in Salphen-Type Schiff Bases for the Preparation of Polynuclear Complexes.

Project description:In this paper, we show a strategy to modify salphen-type Schiff base ligands with naphtol (SYML1) and pyrocathecol (2,3-dihydroxyphenyl) groups (SYML2), or a combination of both (ASYML). Each of these ligands can be used to obtain polynuclear metal complexes following two different strategies. One relies on using metals that are either too large for the N2O2 cavity or not fond of coordination number 4 and the other one relies on forcing the polynuclear species by adding functional groups to the hydroxybenzaldehayde in order to have extra coordination sites in the ligand. We report and characterize the mononuclear complexes SYML1-Cu and SYML1-Ce, along with the dinuclear complex SYML1-Fe and the tetranuclear species SYML2-Mn. The asymmetric ligand ASYML routinely hydrolyzes into the symmetric ligands in the reaction mixtures. SYML1-Fe displays a nearly linear Fe-O-Fe bridge with very strong antiferromagnetic coupling between the Fe(III) ions.

Phenoxyacetohydrazide Schiff bases: β-glucuronidase inhibitors.

Project description:Phenoxyacetohydrazide Schiff base analogs 1-28 have been synthesized and their in vitro β-glucouoronidase inhibition potential studied. Compounds 1 (IC50=9.20±0.32 µM), 5 (IC50=9.47±0.16 µM), 7 (IC50=14.7±0.19 µM), 8 (IC50=15.4±1.56 µM), 11 (IC50=19.6±0.62 µM), 12 (IC50=30.7±1.49 µM), 15 (IC50=12.0±0.16 µM), 21 (IC50=13.7±0.40 µM) and 22 (IC50=22.0±0.14 µM) showed promising β-glucuronidase inhibition activity, better than the standard (D-saccharic acid-1,4-lactone, IC50=48.4±1.25 µM).

Antimicrobial activity of silver sulfide quantum dots functionalized with highly conjugated Schiff bases in a one-step synthesis.

Project description:In the present paper, low-dimensional Ag2S QDs were fabricated for the first time, with four different dithiocarbazate derivative Schiff bases (SB) as capping agents in a one-pot synthesis. These SB-capped Ag2S QDs were almost spherical with an average size range of 4.0 to 5.6 nm, which is slightly smaller than conventional thioglycolic acid (TGA)-capped Ag2S QDs. We demonstrate that the growth of Gram-positive bacteria (Bacillus subtillus and Staphylococcus aureus), Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa), and a prevalent fungal pathogen (Candida albicans) are inhibited more when the bacterial and fungal cells were nurtured with the synthesized SB-Ag2S QDs, compared with TGA-Ag2S QDs or free unbound Schiff bases. The minimum inhibitory concentration (MIC) results confirmed that even low concentrations of SB-Ag2S QDs were able to inhibit bacterial (MIC 5-75 μg mL-1) and fungal growth (MIC 80-310 μg mL-1), and in some cases they performed better than streptomycin (8-25 μg mL-1). Lethality bioassay results confirmed that SB-Ag2S QDs were not toxic to brine shrimp (Artemia salina). The results show that capping agents are essential in the design of functional Ag2S QDs, and highlight that Schiff bases provide an excellent opportunity to optimize the biological activities of silver based QDs.

Effect of Protonation on Optical and Electrochemical Properties of Thiophene-Phenylene-Based Schiff Bases with Alkoxy Side Groups.

Project description:Three polyazomethines and their corresponding model compounds were protonated with trifluoroacetic acid, and its effect on their optical (UV-vis absorption and photoluminescence) properties and electrochemical behavior has been studied, in the context of the presence and elongation of alkoxy side groups. Moreover, the effect of environment dielectric constants (i.e., polarity of the solvent) was considered on the doping process. It has been proven that the presence of alkoxy side groups is necessary for protonation to occur, while unsubstituted compounds undergo hydrolysis to constitutive units. Acid doping of imines consisting of alkoxy side chains has resulted in a distinct bathochromic shift (>200 nm) of the low-energy absorption band. Even the length of alkyl chains has not affected the position of shifted bands; it has been observed that azomethines with smaller, methoxy side groups undergo the protonation process much faster than their octyloxy-substituted analogues, due to the absence of steric hindrance. The electrochemical studies of these alkoxy-substituted imines have indicated a better p-type behavior after protonation induced by the capability of the protonated form to easily oxidize in acetonitrile and to generate the native molecules. The environmental polarity has also had impact on the doping process, which took place only in low-polar media.

Cu(II) and Zn(II) Complexes of New 8-Hydroxyquinoline Schiff Bases: Investigating Their Structure, Solution Speciation, and Anticancer Potential.

Project description:We report the synthesis and characterization of three novel Schiff bases (L1-L3) derived from the condensation of 2-carbaldehyde-8-hydroxyquinoline with amines containing morpholine or piperidine moieties. These were reacted with CuCl2 and ZnCl2 yielding six new coordination compounds, with the general formula ML2, where M = Cu(II) or Zn(II) and L = L1-L3, which were all characterized by analytical, spectroscopic (Fourier transform infrared (FTIR), UV-visible absorption, nuclear magnetic resonance (NMR), or electron paramagnetic resonance (EPR)), and mass spectrometric techniques, as well as by single-crystal X-ray diffraction. In the solid state, two Cu(II) complexes, with L1 and L2, are obtained as dinuclear compounds, with relatively short Cu-Cu distances (3.146 and 3.171 Å for Cu2(L1)4 and Cu2(L2)4, respectively). The free ligands show moderate lipophilicity, while their complexes are more lipophilic. The pKa values of L1-L3 and formation constants of the complex (for ML and ML2) species were determined by spectrophotometric titrations, with the Cu(II) complexes showing higher stability than the Zn(II) complexes. EPR indicated the presence of several species in solution as pH varied and binding modes were proposed. The binding of the complexes to bovine serum albumin (BSA) was evaluated by fluorescence and circular dichroism (CD) spectroscopies. All complexes bind BSA, and as demonstrated by CD, the process takes several hours to reach equilibrium. The antiproliferative activity was evaluated in malignant melanoma cells (A375) and in noncancerous keratinocytes (HaCaT). All complexes display significant cytotoxicity (IC50 < 10 μM) but modest selectivity. The complexes show higher activity than the free ligands, the Cu(II) complexes being more active than the Zn(II) complexes, and approximately twice more cytotoxic than cisplatin. A Guava ViaCount assay corroborated the antiproliferative activity.

Memantine-Derived Schiff Bases as Transdermal Prodrug Candidates.

Project description:Condensation reactions of salicylaldehyde, 2-pyridinecarboxaldehyde, and pyridoxaldehyde with memantine (Me) produced novel memantine-derived Schiff bases (1-3). Speciation predictions and calculations of Log P, Log D, and of the percentage (%) of neutral species for (1-3) were carried out. In comparison with Me, the Schiff bases presented increased log P and log D in all cases and pH values, suggesting higher hydrophobicity. The determined solubilities in n-octanol were 34.7 mg/mL for memantine hydrochloride and 67.3 mg/mL for (3). According to the molecular weights and calculated logP, compounds (1-3) are suitable for transdermal administration, especially compound (3). In addition, hydrolysis of 3 with the release of pyridoxal, a daily cofactor in human metabolism, was observed. The results suggested that 3 is the most promising compound and that formation of the pyridoxal Schiff base with Me might be an effective strategy to obtain a prodrug candidate with increased lipophilicity, which would be able to passively cross biological barriers during transdermal delivery and might have applications in the treatment of Alzheimer's disease and other neurological disorders.
