Pretreatment methods for monitoring microplastics in soil and freshwater sediment samples: A comprehensive review.

ABSTRACT: This paper reviews the currently used pretreatment methods for microplastics (MPs) analysis in soil and freshwater sediments, primarily sample processing, pretreatment, and characterization methods for MPs analysis. In addition, analytical tools (e.g., lab instruments), MPs characteristics, and MPs quantity, are included in this review. Prior to pretreatment, soil and sediment samples are typically processed using sieving and drying methods, and a sample quantity of <50 g was mostly used for the pretreatment. Density separation was commonly performed before organic matter removal. Sodium chloride (NaCl) and zinc chloride (ZnCl₂) were most often used for density separation, and hydrogen peroxide (H₂O₂) oxidation was most frequently used to remove organic matter. Although advantages of each pretreatment method have been investigated, it is still challenging to determine a universal pretreatment method due to sample variability (e.g., sample characteristics). Furthermore, it is highly required to establish standard pretreatment methods that can be used for various environmental matrices, including air, water, and wastes as well as soil and sediment.

Project description:Understanding of the sources, fate, and impact of microplastics (MPs, < 5 mm) remains limited, particularly in freshwater environments, while limited comparability across available surveys hinders adequate monitoring and risk assessment of these contaminants. Here, the distribution of microscopic debris in an urban river close to the marine environment in the West of Scotland was investigated to assess concentration and distribution of primary and secondary MPs. Also, the efficiency of light and scanning electron microscopy with energy-dispersive spectroscopy (SEM-EDS) was evaluated for characterisation and quantification of MPs sized 2.8 mm-11 μm. Bank sediment samples were collected twice from the River Kelvin in Glasgow and were size-fractionated and processed for extraction of MPs by density separation. Sample MPs spiking and use of procedural blanks allowed the influence of processing on field data quality to be considered. Total abundances were 161-432 MPs kg-1 dry sediment, with fibres as the dominant type, comprising > 88% of total counts. Nevertheless, fibres in blanks suggest potential contributions from atmospheric contamination. Moreover, fibres concentrated mainly in fractions < 0.09 mm suggesting that their fate may be influenced by drivers of fine sediment dynamics in rivers. While no primary MPs were observed, metallic and glass pellets were present in high abundances in settled material and could be easily misidentified by visual inspection, demonstrating that compositional analysis is needed to avoid analytical errors from MP misidentification and overestimation. SEM-EDS allowed for a quick screening of plastic vs non-plastic pellets and improved identification of smaller fragments, whereas more advanced techniques are needed for proper identification of fibres. This study is the first to report on MPs in freshwater rivers in Scotland and suggests that diffuse sources of pollution may be delivering secondary MPs to the river. Their sources, fate, and risk in these systems will thus warrant further attention.

Project description:Although a number of different methods have been used to quantify soil bacteria, identifying the optimal method(s) for soil bacterial abundance is still in question. No single method exists for undertaking an absolute microbial count using culture-dependent methods (CDMs) or even culture-independent methods (CIMs). This study investigated soil storage and pretreatment methods for optimal bacterial counts. Appropriate storage temperature (4°C) and optimal pretreatment methods (sonication time for 3 min and centrifugation at 1400 g) were necessary to preserve bacterial cell viability and eliminate interference from soil particles. To better estimate soil bacterial numbers under various cellular state and respiration, this study also evaluated three CDMs (i.e., colony forming unit, spotting, and most probable number (MPN) and three CIMs (i.e., flow cytometry (FCM), epifluorescence microscopy (EM) count, and DNA quantitation). Each counting method was tested using 72 soil samples collected from a local arable farm site at three different depths (i.e., 10-20, 90-100, and 180-190 cm). Among all CDMs, MPN was found to be rapid, simple, and reliable. However, the number of bacteria quantified by MPN was 1-2 orders lower than that quantified by CIMs, likely due to the inability of MPN to count anaerobic bacteria. The DNA quantitation method appeared to overestimate soil bacterial numbers, which may be attributed to DNA from dead bacteria and free DNA in the soil matrix. FCM was found to be ineffective in counting soil bacteria as it was difficult to separate the bacterial cells from the soil particles. Dyes used in FCM stained the bacterial DNA and clay particles. The EM count was deemed a highly effective method as it provided information on soil mineral particles, live bacteria, and dead bacteria; however, it was a time-consuming and labor-intensive process. Combining both types of methods was considered the best approach to acquire better information on the characteristics of indigenous soil microorganisms (aerobic versus anaerobic, live versus dead).
