Persistent homological cell tracking technology.

ABSTRACT: In this paper, we develop a cell tracking method based on persistent homological figure detection technology. We apply our tracking method to 9 different time-series cell images and extract several kinds of cell movements. Being able to analyze various images with a single method allows researchers to systematically understand and compare different tracking data. Persistent homological cell tracking technology's 9 parameters all have clear meanings. Thus, researchers can decide the parameters not by black box trial-and-error but by the purpose of their analysis. We use model data with ground truth to see our method's performance. We compare persistent homological figure detection and cell tracking technology with Image-Pro, sure-foreground in watershed method, and cell detection methods in previous studies. We see that there are some cases where Image-Pro's tracking stops and requires manual plots, while our method does not require manual plots. We show that our technology includes sure-foreground and has more information, and can be applied to different types of data that previously needed different methods. We also show that our technology is powerful as a detection technology by applying the technology to 5 different types of cell images.

Project description:Radio frequency identification (RFID) has become a key technology in the logistics and management industry, thanks to distinctive features such as the low cost of RFID tags, and the easiness of the RFID tags' deployment and integration within the items to be tracked. In consequence, RFID plays a fundamental role in the so-called digital factory or 4.0 Industry, aiming to increase the level of automatization of industrial processes. In addition, RFID has also been found to be of great help in improving the tracking of patients, medicines, and medical assets in hospitals, where the digitalization of these operations improves their efficiency and safety. This contribution reviews the state-of-the-art of RFID for e-Health applications, describing the contributions to improve medical services and discussing the limitations. In particular, it has been found that a lot of effort has been put into software development, but in most of the cases a detailed study of the physical layer (that is, the characterization of the RFID signals within the area where the system is deployed) is not properly conducted. This contribution describes a basic RFID system for tracking and managing assets in hospitals, aiming to provide additional details about implementation aspects that must be considered to ensure proper functionality of the system. Although the scope of the RFID system described in this contribution is restricted to a small area of the hospital, the architecture is fully scalable to cover the needs of the different medical services in the hospital. Ultra high-frequency (UHF) RFID technology is selected over the most extended near-field communication (NFC) and high-frequency (HF) RFID technology to minimize hardware infrastructure. In particular, UHF RFID also makes the coverage/reading area conformation easier by using different kinds of antennas. Information is stored in a database, which is accessed from end-user mobile devices (tablets, smartphones) where the position and status of the assets to be tracked are displayed.

Project description:ImportanceObjectively measuring how Mohs defect reconstruction changes casual observer attention has important implications for patients and facial plastic surgeons.ObjectiveTo use eye-tracking technology to objectively measure the ability of Mohs facial defect reconstruction to normalize facial attention.Design, setting, and participantsThis observational outcomes study was conducted at an academic tertiary referral center from January to June 2016. An eye-tracking system was used to record how 82 casual observers directed attention to photographs of 32 patients with Mohs facial defects of varying sizes and locations before and after reconstruction as well as 16 control faces with no facial defects. Statistical analysis was performed from November 2018 to January 2019.Main outcomes and measuresFirst, the attentional distraction caused by facial defects was quantified in milliseconds of gaze time using eye tracking. Second, the eye-tracking data were analyzed using mixed-effects linear regression to assess the association of facial defect reconstruction with normalized facial attention.ResultsThe 82 casual observers (63 women and 19 men; mean [SD] age, 34 [12] years) viewed control faces in a similar and consistent fashion, with most attention (65%; 95% CI, 62%-69%) directed at the central triangle, which includes the eyes, nose, and mouth. The eyes were the most visually important feature, capturing a mean of 60% (95% CI, 57%-64%) of fixation time within the central triangle and 39% (95% CI, 36%-43%) of total observer attention. The presence of Mohs defects was associated with statistically significant alterations in this pattern of normal facial attention. The larger the defect and the more centrally a defect was located, the more attentional distraction was observed, as measured by increased attention on the defect and decreased attention on the eyes, ranging from 729 (95% CI, 526-931) milliseconds for small peripheral defects to 3693 (95% CI, 3490-3896) milliseconds for large central defects. Reconstructive surgery was associated with improved gaze deviations for all faces and with normalized attention directed to the eyes for all faces except for those with large central defects.Conclusions and relevanceMohs defects are associated with altered facial perception, diverting attention from valuable features such as the eyes. Reconstructive surgery was associated with normalized attentional distraction for many patients with cutaneous Mohs defects. These data are important to patients who want to know how reconstructive surgery could change the way people look at their face. The data also point to the possibility of outcomes prediction based on facial defect size and location before reconstruction. Eye tracking is a valuable research tool for outcomes assessment that lays the foundation for understanding how reconstructive surgery may change perception and normalize facial deformity.

Project description:PurposeStereopsis, the ability of humans to perceive depth through distinct visual stimuli in each eye, is foundational to autostereoscopic technology in computing. However, ensuring stable head position during assessments has been challenging. This study evaluated the utility of artificial intelligence (AI)-enhanced face tracking technology in overcoming this challenge by ensuring that each eye consistently receives its intended image.MethodsThe Lume Pad 2, an autostereoscopic tablet with AI-enhanced face tracking, was utilized to simulate quantitative parts of the Stereo Fly test and TNO Stereotests for contour and random dot stereopsis. The study recruited 30 children (14 males and 16 females, mean age of 9.2 ± 0.3 years, age range of 6-12 years) and 30 adults (10 males and 20 females, mean age of 29.4 ± 1.0 years, age range of 21-42 years) to assess the tablet's inter-session reliability. Agreement between conventional and the autostereoscopic tablet-simulated stereotests was tested in a larger group of 181 children (91 males and 90 females, mean age of 9.1 ± 0.4 years, age range of 6-12 years) and 160 adults (69 males and 91 females, mean age of 38.6 ± 2.1 years, age range of 21-65 years). Inter-session reliability and agreement were analyzed using weighted Kappa coefficient and non-parametric Bland-Altman analysis.ResultsThe autostereoscopic tablet demonstrated high inter-session reliability (κ all > 0.80), except for the simulated TNO Stereotest in adults, which demonstrated moderate inter-session reliability (κ = 0.571). Non-parametric Bland-Altman analysis revealed zero median differences, confirming consistent inter-session reliability. Similar patterns were observed in comparing AI-based and conventional methods, with both the weighted Kappa coefficient (κ all > 0.80) and non-parametric Bland-Altman analysis indicating significant agreement. The agreement between methodologies was confirmed by permissible differences, which were smaller than the minimum step range.ConclusionThe integration of AI-based autostereoscopic technology with sub-pixel precision demonstrates significant potential for clinical stereopsis measurements.
