Flexible multichannel electrodes for acute recording in nonhuman primates.

ABSTRACT: Flexible electrodes have demonstrated better biocompatibility than rigid electrodes in relieving tissue encapsulation and long-term recording. Nonhuman primates are closer to humans in their brains' structural and functional properties, thus making them more suitable than rodents as animal models for potential clinical usage. However, the application of flexible electrodes on nonhuman primates has rarely been reported. In the present study, a flexible multichannel electrode array for nonhuman primates was developed and implemented for extracellular recording in behaving monkeys. To minimize the window of durotomy for reducing possible risks, a guide-tube-compatible implantation solution was designed to deliver the flexible electrodes through the dura into the cortex. The proposed structure for inserting flexible electrodes was characterized ex vivo and validated in vivo. Furthermore, acute recording of multichannel flexible electrodes for the primates was performed. The results showed that the flexible electrodes and implantation method used in this study meet the needs of extracellular recording in nonhuman primates. Task-related neuronal activities with a high signal-to-noise ratio of spikes demonstrated that our whole device is currently a minimally invasive and clinically viable approach for extracellular recording.

Project description:Soft and conductive nanomaterials like carbon nanotubes, graphene, and nanowire scaffolds have expanded the family of ultraflexible microelectrodes that can bend and flex with the natural movement of the brain, reduce the inflammatory response, and improve the stability of long-term neural recordings. However, current methods to implant these highly flexible electrodes rely on temporary stiffening agents that temporarily increase the electrode size and stiffness thus aggravating neural damage during implantation, which can lead to cell loss and glial activation that persists even after the stiffening agents are removed or dissolve. A method to deliver thin, ultraflexible electrodes deep into neural tissue without increasing the stiffness or size of the electrodes will enable minimally invasive electrical recordings from within the brain. Here we show that specially designed microfluidic devices can apply a tension force to ultraflexible electrodes that prevents buckling without increasing the thickness or stiffness of the electrode during implantation. Additionally, these "fluidic microdrives" allow us to precisely actuate the electrode position with micron-scale accuracy. To demonstrate the efficacy of our fluidic microdrives, we used them to actuate highly flexible carbon nanotube fiber (CNTf) microelectrodes for electrophysiology. We used this approach in three proof-of-concept experiments. First, we recorded compound action potentials in a soft model organism, the small cnidarian Hydra. Second, we targeted electrodes precisely to the thalamic reticular nucleus in brain slices and recorded spontaneous and optogenetically evoked extracellular action potentials. Finally, we inserted electrodes more than 4 mm deep into the brain of rats and detected spontaneous individual unit activity in both cortical and subcortical regions. Compared to syringe injection, fluidic microdrives do not penetrate the brain and prevent changes in intracranial pressure by diverting fluid away from the implantation site during insertion and actuation. Overall, the fluidic microdrive technology provides a robust new method to implant and actuate ultraflexible neural electrodes.

Project description:BackgroundThe development of wearable health monitoring systems is garnering tremendous interest in research, technology and commercial applications. Their ability of providing unique capabilities in continuous, real-time, and non-invasive tracking of the physiological markers of users can provide insights into the performance and health of individuals. Electrocardiogram (ECG) signals are of particular interest, as cardiovascular disease is the leading cause of death globally. Monitoring heart health and its conditions such as ventricular disturbances and arrhythmias can be achieved through evaluating various features of ECG such as R-peaks, QRS complex, T-wave, and P-wave. Despite recent advances in biosensors for wearable applications, most of the currently available solutions rely solely on a single system attached to the body, limiting the ability to obtain reliable and multi-location biosignals. However, in engineering systems, sensor fusion, which is the optimal integration and processing of data from multiple sensors, has been a common theme and should be considered for wearables. In recent years, due to an increase in the availability and variety of different types of sensors, the possibility of achieving sensor fusion in wearable systems has become more attainable. Sensor fusion in multi-sensing systems results in significant enhancements of information inferences compared to those from systems with a sole sensor. One step towards the development of sensor fusion for wearable health monitoring systems is the accessibility to multiple reliable electrophysiological signals, which can be recorded continuously.ResultsIn this paper, we develop a textile-based multichannel ECG band that has the ability to measure ECG from multiple locations on the waist. As a proof of concept, we demonstrate that ECG signals can be reliably obtained from different locations on the waist where the shape of the QRS complex is nearly comparable with recordings from the chest using traditional gel electrodes. In addition, we develop a probabilistic approach-based on prediction and update strategies-to detect R-peaks from noisy textile data in different statuses, including sitting, standing, and jogging. In this approach, an optimal search method is utilized to detect R-peaks based on the history of the intervals between previously detected R-peaks. We show that the performance of our probabilistic approach in R-peak detection is significantly better than that based on Pan-Tompkins and optimal-threshold methods.ConclusionA textile-based multichannel ECG band was developed to track the heart rate changes from multiple locations on the waist. We demonstrated that (i) the ECG signal can be detected from different locations on the waist, and (ii) the accuracy of the detected R-peaks from textile sensors was improved by using our proposed probabilistic approach. Despite the limitations of the textile sensors that might compromise the quality of ECG signals, we anticipate that the textile-based multichannel ECG band can be considered as an effective wearable system to facilitate the development of sensor fusion methodology for pervasive and non-invasive health monitoring through continuous tracking of heart rate variability (HRV) from the waist. In addition, from the commercialization point of view, we anticipate that the developed band has the potential to be integrated into garments such as underwear, bras or pants so that individuals can use it on a daily basis.

Project description:BackgroundIn terms of structure and function, the skull is one of the most complicated organs in the body. It is also one of the most important parts in terms of developmental and evolutionary origins. This complexity makes it difficult to obtain evolutionary assessments if, as is usually the case with fossils, only part of the skull is available. For this reason this study involves a set of comparisons whereby the smallest functional units are studied first, and these built up, through a triple-nested hierarchical design, into more complex anatomical regions and eventually into the skull-as-a-whole. This design has been applied to macaques (Macaca) in order to reveal patterns of variation at the different levels. The profiles of such variation have been obtained both within and between species. This has lead to a search for the skull parts that have undergone similar selection pressures during evolution and comparable development patterns in both ontogeny and phylogeny.ResultsMorphometric analysis (Principal Components) was used to obtain these profiles of species and sex separations based on 77 cranial variables from 11 species of macaques. The results showed that 7 functional units could be aggregated into three functionally reasonable anatomical regions on the basis of similarities in profiles. These were: the masticatory apparatus containing mandible, lower teeth and upper teeth, the face as a whole combining maxilla (actually lower face) and upper face, and the cranium as a whole involving cranium and calvaria. Twenty-six variables were finally selected for analyzing the morphology of the whole skull. This last showed an overall profile similar to that revealed in the masticatory apparatus but also contained additional information pertaining to individual species and species-groups separations.ConclusionsThe study provides a model for carrying out analysis of species separations and sex variation simultaneously. Through this design it seems possible to see cranio-dental elements that may result from similar developmental processes, have similar functional adaptations, and show an appropriately integrated structure morphologically. This study also implies that the biological information drawn from part of skull alone, e.g. as in studies of incomplete fossils may provide misleading information.
