Ammonolytic transfer dehydrogenation of amines and amides: a versatile method to valorize nitrogen compounds to nitriles.

ABSTRACT: The dehydrogenation of amines has been identified as an efficient method for nitrile synthesis. At present, this approach is restricted to (oxidative) dehydrogenations of primary amines, most often with specialized homogeneous catalysts. In this work, amines were transfer dehydrogenated to nitriles using simple and cheap alkenes (e.g. ethylene or propene) as hydrogen scavengers. The scope was expanded to secondary amines, tertiary amines and even aldehydes. Additional nitrogen is built in from NH₃. The versatility of the process was proven by coupling it to the ammonolysis of secondary amides. This enabled us to recycle long-chain polyamides (LCPA) into monomeric compounds, i.e. α,ω-amidonitriles and dinitriles. Reactions were performed with a recyclable heterogeneous Pt catalyst, at 200 °C and with limited addition of NH₃ and ethylene. High yields of up to 94% were obtained for the corresponding nitriles.

Electrochemical Dehydrogenation Pathways of Amines to Nitriles on NiOOH.

Project description:Nitriles are highly important synthetic intermediates with applications in a wide variety of organic reactions including production of pharmaceuticals, fine chemicals, and agricultural chemicals. Thus, developing effective green routes to oxidize amines to nitriles is of great interest. One promising method to achieve the oxidation of primary amines to nitriles is through electrochemical oxidation on NiOOH electrodes. This reaction has long been thought to occur through an indirect mechanism consisting of a series of potential independent hydrogen atom transfer steps to catalytic Ni3+ sites in NiOOH, which reduces NiOOH to Ni(OH)2. The role of the applied potential in this mechanism is simply to regenerate NiOOH by oxidizing Ni(OH)2. In this work, we demonstrate that a second, potential-dependent pathway recently found to apply to alcohol and aldehyde oxidation on NiOOH and consisting of potential-dependent hydride transfer to Ni4+ sites is the dominant pathway for the oxidation of amines using propylamine and benzylamine as model systems. After qualitatively and quantitatively examining the contributions of indirect and potential-dependent oxidation pathways to amine oxidation on NiOOH, we also examine the effect the amine concentration, solution pH, applied bias, and deuterium substitution have on the two pathways, further clarifying their mechanisms and exploring what factors control their rate. This work provides a comprehensive understanding of the mechanism of primary amine oxidation on NiOOH.

Pd-CuFe Catalyst for Transfer Hydrogenation of Nitriles: Controllable Selectivity to Primary Amines and Secondary Amines.

Project description:A multicomponent nanocatalyst system was fabricated for the transfer hydrogenation of nitrile compounds. This catalyst system contains palladium, copper, and iron, which are supported on the magnetite nanospheres, and the loading of palladium could be at the parts per million level. Palladium and copper contribute to the transformation of nitrile, and the product distribution highly depends on the alloying of Fe to Cu. The nitriles could be converted to primary amine by the Pd-Cu catalyst in the absence of Fe, whereas in the presence of Fe the products are secondary amines with high selectivity. This could be attributed to the electronic modulation of iron to copper. A variety of nitriles have been transformed to the corresponding primary or secondary amines with high selectivity, and the TOF reaches 2,929 hr-1 for Pd. Furthermore, the catalyst could be recycled by an external magnetic field and reused five times without severe activity loss.

Photoinduced Remote Functionalization of Amides and Amines Using Electrophilic Nitrogen Radicals.

Project description:The selective functionalization of C(sp3 )-H bonds at distal positions to functional groups is a challenging task in synthetic chemistry. Reported here is a photoinduced radical cascade strategy for the divergent functionalization of amides and protected amines. The process is based on the oxidative generation of electrophilic amidyl radicals and their subsequent transposition by 1,5-H-atom transfer, resulting in remote fluorination, chlorination and, for the first time, thioetherification, cyanation, and alkynylation. The process is tolerant of most common functional groups and delivers useful building blocks that can be further elaborated. The utility of this strategy is demonstrated through the late-stage functionalization of amino acids and a dipeptide.

The PM6-FGC Method: Improved Corrections for Amines and Amides.

Project description:Recently, we reported a new approach to develop pairwise analytical corrections to improve the description of noncovalent interactions, by approximate methods of electronic structures, such as semiempirical quantum mechanical (SQM) methods. In particular, and as a proof of concept, we used the PM6 Hamiltonian and we named the method PM6-FGC, where the FGC acronym, corresponding to Functional Group Corrections, emphasizes the idea that the corrections work for specific functional groups rather than for individual atom pairs. The analytical corrections were derived from fits to B3LYP-D3/def2-TZVP (reference). PM6 interaction energy differences, evaluated for a reduced set of small bimolecular complexes, were chosen as representatives of saturated hydrocarbons, carboxylic, amine and, tentatively, amide functional groups. For the validation, the method was applied to several complexes of well-known databases, as well as to complexes of diglycine and dialanine, assuming the transferability of amine group corrections to amide groups. The PM6-FGC method showed great potential but revealed significant inaccuracies for the description of some interactions involving the -NH2 group in amines and amides, caused by the inadequate selection of the model compound used to represent these functional groups (an NH3 molecule). In this work, methylamine and acetamide are used as representatives of amine and amide groups, respectively. This new selection leads to significant improvements in the calculation of noncovalent interactions in the validation set.

Chemoselective α,β-Dehydrogenation of Saturated Amides.

Project description:We report a method for the selective α,β-dehydrogenation of amides in the presence of other carbonyl moieties under mild conditions. Our strategy relies on electrophilic activation coupled to in situ selective selenium-mediated dehydrogenation. The α,β-unsaturated products were obtained in moderate to excellent yields, and their synthetic versatility was demonstrated by a range of transformations. Mechanistic experiments suggest formation of an electrophilic SeIV species.

One-pot method for the synthesis of 1-aryl-2-aminoalkanol derivatives from the corresponding amides or nitriles.

Project description:We have identified a novel one-pot method for the synthesis of β-amino alcohols, which is based on C-H bond hydroxylation at the benzylic α-carbon atom with a subsequent nitrile or amide functional group reduction. This cascade process uses molecular oxygen as an oxidant and sodium bis(2-methoxyethoxy)aluminum hydride as a reductant. The substrate scope was examined on 30 entries and, although the respective products were provided in moderate yields only, the above simple protocol may serve as a direct and powerful entry to the sterically congested 1,2-amino alcohols that are difficult to prepare by other routes. The plausible mechanistic rationale for the observed results is given and the reaction was applied to a synthesis of a potentially bioactive target.

Oxidative 1,2-carboamination of alkenes with alkyl nitriles and amines toward γ-amino alkyl nitriles.

Project description:Difunctionalization of alkenes has become a powerful tool for quickly increasing molecular complexity in synthesis. Despite significant progress in the area of alkene difunctionalization involving the incorporation of a nitrogen atom across the C-C double bonds, approaches for the direct 1,2-carboamination of alkenes to produce linear N-containing molecules are scarce and remain a formidable challenge. Here we describe a radical-mediated oxidative intermolecular 1,2-alkylamination of alkenes with alkyl nitriles and amines involving C(sp3)-H oxidative functionalization catalysed by a combination of Ag2CO3 with iron Lewis acids. This three-component alkene 1,2-alkylamination method is initiated by the C(sp3)-H oxidative radical functionalization, which enables one-step formation of two new chemical bonds, a C-C bond and a C-N bond, to selectively produce γ-amino alkyl nitriles.

A versatile method for the identification of senolytic compounds.

Project description:The increased burden of senescent cells is as a well-established hallmark of aging and age-related diseases. This finding sparked significant interest in the identification of molecules capable of selectively eliminating senescent cells, so-called senolytics. Here, we fine-tuned a method for the identification of senolytics that is compatible with high-content fluorescence microscopy. We used spectral detector imaging to measure the emission spectrum of unlabeled control or senescent cells. We observed that senescent cells exhibited higher levels of autofluorescence than their non-senescent counterparts, particularly in the cytoplasmic region. Building on this result, we devised a senolytic assay based on co-culturing quiescent and senescent cells, fluorescently tagged in the nuclear region through the overexpression of H2B-GFP and H2B-RFP, respectively. We validated this approach by showing that first generation senolytics were effective in reducing the number of RFP+ nuclei leaving the count of GFP+ nuclei unaffected. The result was confirmed by flow cytometry analysis of nuclei isolated from these quiescent-senescent cell co-cultures. We found that this system enables to capture cell type-specific effects of senolytics as in the case of fisetin, which kills senescent Mouse Embryonic Fibroblasts but not senescent human melanoma SK-MEL-103 cells. This approach is amenable to genetic and chemical screening for the discovery of senolytic compounds in that it overcomes the limitations of current methods, which rely upon costly chemical reagents or fluorescence microscopy using cells labeled with fluorescent cytoplasmic probes that overlap with the autofluorescence signal emitted by senescent cells.

Efficient Dehydrogenation of Amines and Carbonyl Compounds Catalyzed by a Tetranuclear Ruthenium-mu-Oxo-mu-Hydroxo-Hydride Complex.

Project description:The tetranuclear ruthenium-mu-oxo-mu-hydroxo-hydride complex {[(PCy(3))(CO)RuH](4)(mu(4)-O)(mu(3)-OH)(mu(2)-OH)} (1) was found to be a highly effective catalyst for the transfer dehydrogenation of amines and carbonyl compounds. For example, the initial turnover rate of the dehydrogenation of 2-methylindoline was measured to be 1.9 s(-1) with the TON of 7950 after 1 h at 200 degrees C. The extensive H/D scrambling patterns observed from the dehydrogenation reaction of indoline-N-d(1) and indoline-alpha-d(2) suggest a monohydride mechanistic pathway with the C-H bond activation rate-limiting step.
