Project description:The lens system in the human eye is able to best focus light from only one distance at a time.Therefore, many objects in the natural environment are not imaged sharply on the retina. Furthermore, light from objects in the environment is subject to the particular aberrations of the observer's lens system (e.g., astigmatism and chromatic aberration). We refer to blur created by the observer's optics as "natural" or "defocus" blur as opposed to "on-screen" blur created by software on a display screen. Although blur discrimination has been studied extensively, human ability to discriminate defocus blur in images of natural scenes has not been systematically investigated. Here, we measured discrimination of defocus blur for a collection of natural image patches, sampled from well-focused photographs. We constructed a rig capable of presenting stimuli at three physical distances simultaneously. In Experiment 1, subjects viewed monocularly two simultaneously presented natural image patches through a 4-mm artificial pupil at ±1° eccentricity. The task was to identify the sharper patch. Discrimination thresholds varied substantially between stimuli but were correlated between subjects. The lowest thresholds were at or below the lowest thresholds ever reported. In a second experiment, we paralyzed accommodation and retested a subset of conditions from Experiment 1. A third experiment showed that removing contrast as a cue to defocus blur had only a modest effect on thresholds. Finally, we describe a simple masking model and evaluate how well it can explain our experimental results and the results from previous blur discrimination experiments.

Project description:PurposeThis study aims to examine the short-term peripheral choroidal thickness (PChT) response to signed defocus blur, both with and without native peripheral aberrations. This examination will provide insights into the role of peripheral aberration in detecting signs of defocus.MethodsThe peripheral retina (temporal 15°) of the right eye was exposed to a localized video stimulus in 11 young adults. An adaptive optics system induced 2D myopic or hyperopic defocus onto the stimulus, with or without correcting native peripheral ocular aberrations (adaptive optics [AO] or NoAO defocus conditions). Choroidal scans were captured using Heidelberg Spectralis OCT at baseline, exposure (10, 20, and 30 minutes), and recovery phases (4, 8, and 15 minutes). Neural network-based automated MATLAB segmentation program measured PChT changes from OCT scans, and statistical analysis evaluated the effects of different optical conditions over time.ResultsDuring the exposure phase, NoAO myopic and hyperopic defocus conditions exhibited distinct bidirectional PChT alterations, showing average thickening (10.0 ± 5.3 µm) and thinning (-9.1 ± 5.5 µm), respectively. In contrast, induced AO defocus conditions did not demonstrate a significant change from baseline. PChT recovery to baseline occurred for all conditions. The unexposed fovea did not show any significant ChT change, indicating a localized ChT response to retinal blur.ConclusionsWe discovered that the PChT response serves as a marker for detecting peripheral retinal myopic and hyperopic defocus blur, especially in the presence of peripheral aberrations. These findings highlight the significant role of peripheral oriented blur in cueing peripheral defocus sign detection.

Project description:From measurements of wavefront aberrations in 16 emmetropic eyes, we calculated where objects in the world create best-focused images across the central 27\(^\circ\) (diameter) of the retina. This is the retinal conjugate surface. We calculated how the surface changes as the eye accommodates from near to far and found that it mostly maintains its shape. The conjugate surface is pitched top-back, meaning that the upper visual field is relatively hyperopic compared to the lower field. We extended the measurements of best image quality into the binocular domain by considering how the retinal conjugate surfaces for the two eyes overlap in binocular viewing. We call this binocular extension the blur horopter. We show that in combining the two images with possibly different sharpness, the visual system creates a larger depth of field of apparently sharp images than occurs with monocular viewing. We examined similarities between the blur horopter and its analog in binocular vision: the binocular horopter. We compared these horopters to the statistics of the natural visual environment. The binocular horopter and scene statistics are strikingly similar. The blur horopter and natural statistics are qualitatively, but not quantitatively, similar. Finally, we used the measurements to refine what is commonly referred to as the zone of clear single binocular vision.

Project description:Electrophysiological population signals contain oscillatory and non-oscillatory aperiodic (1/frequency-like) components. So far research has largely focused on oscillatory activity, and only recently, interest in aperiodic population activity has gained momentum. Accordingly, while the cortical correlation structure of oscillatory population activity has been characterized, little is known about the correlation of aperiodic neuronal activity. To address this, we investigated aperiodic neuronal population activity in the human brain using resting-state magnetoencephalography (MEG). We combined source-analysis, signal orthogonalization and irregular-resampling auto-spectral analysis (IRASA) to systematically characterize the cortical distribution and correlation of aperiodic neuronal activity. We found that aperiodic population activity is robustly correlated across the cortex and that this correlation is spatially well structured. Furthermore, we found that the cortical correlation structure of aperiodic activity is similar but distinct from the correlation structure of oscillatory neuronal activity. Anterior cortical regions showed the strongest differences between oscillatory and aperiodic correlation patterns. Our results suggest that correlations of aperiodic population activity serve as robust markers of cortical network interactions. Furthermore, our results show that aperiodic and oscillatory signal components provide non-redundant information about large-scale neuronal correlations. This may reflect at least partly distinct neuronal mechanisms underlying and reflected by oscillatory and aperiodic neuronal population activity.

Project description:Global form and motion sensitivity undergo long development in childhood with motion sensitivity rather than form being impaired in a number of childhood disorders and both impaired in adult clinical populations. This suggests extended development and vulnerability of extrastriate cortical areas associated with global processing. However, in some developmental and clinical populations, it remains unclear to what extent impairments might reflect deficits at earlier stages of visual processing, such as reduced visual acuity and contrast sensitivity. To address this, we investigated the impact of degraded spatial vision on cortical global form and motion processing in healthy adults. Loss of high spatial frequencies was simulated using a diffuser to blur the stimuli. Participants completed behavioral and EEG tests of global form and motion perception under three levels of blur. For the behavioral tests, participants' form and motion coherence thresholds were measured using a two-alternative, forced-choice procedure. Steady-state visual evoked potentials were used to measure cortical responses to changes in the coherence of global form and motion stimuli. Both global form and global motion perception were impaired with increasing blur as measured by elevated behavioral thresholds and reduced cortical responses. However, form thresholds showed greater impairment in both behavioral and EEG measures than motion thresholds at the highest levels of blur. The results suggest that high spatial frequencies play an important role in the perception of both global form and motion but are especially significant for global form. Overall, the results reveal complex interactions between low-level factors and global visual processing, highlighting the importance of taking these factors into account when investigating extrastriate function in low vision populations.

Project description:We present hierarchical spherical deformation for a group-wise shape correspondence to address template selection bias and to minimize registration distortion. In this work, we aim at a continuous and smooth deformation field to guide accurate cortical surface registration. In conventional spherical registration methods, a global rigid alignment and local deformation are independently performed. Motivated by the composition of precession and intrinsic rotation, we simultaneously optimize global rigid rotation and non-rigid local deformation by utilizing spherical harmonics interpolation of local composite rotations in a single framework. To this end, we indirectly encode local displacements by such local composite rotations as functions of spherical locations. Furthermore, we introduce an additional regularization term to the spherical deformation, which maximizes its rigidity while reducing registration distortion. To improve surface registration performance, we employ the second order approximation of the energy function that enables fast convergence of the optimization. In the experiments, we validate our method on healthy normal subjects with manual cortical surface parcellation in registration accuracy and distortion. We show an improved shape correspondence with high accuracy in cortical surface parcellation and significantly low registration distortion in surface area and edge length. In addition to validation, we discuss parameter tuning, optimization, and implementation design with potential acceleration.

Project description:Because refractive development is governed largely by the retina, we analyzed the retinal transcriptome in chicks wearing a spectacle lens, a well-established means to induce refractive errors, to identify gene expression alterations and to develop novel mechanistic hypotheses about refractive development. One-week-old white Leghorn chicks wore a unilateral spectacle lens of + or –15 diopters for 6 hours or 3 days (n=6 for each condition). With total RNA from the retina/retinal pigment epithelium (RPE), Affymetrix Chicken GeneChips were used to compare gene expression levels between lens-wearing and contralateral control eyes. Normalized microarray signal intensities were evaluated by an analysis of variance approach. Selected differentially expressed genes were validated by qPCR in biologically independent samples.

Project description:In the context of ocular development and eye growth regulation, retinal defocus and/or image contrast appear key variables although the nature of the signal(s) relayed from the retina to the sclera remains poorly understood. Nonetheless, under optimal visual conditions, eye length is brought into alignment with its optical power to achieve approximate emmetropia, through appropriate adjustment to eye growth. The retinal pigment epithelium (RPE), which lies between the retina and choroid/sclera, appears to play a crucial role in this process. In the investigations reported here, we used a chick model system to assess the threshold duration of exposure to lens-imposed defocus and form-deprivation necessary for conversion of evoked retinal signals into changes in BMP gene expression in the RPE. Our study provides evidence for the following: 1) close-loop, optical defocus-guided (negative and positive lenses) bidirectional BMP gene expression regulation, 2) open-loop, form-deprivation (diffusers)-induced down-regulation of BMP gene expression, and 3) early, transient up-regulation of BMP gene expression in response to both types of lens and diffuser applications. The critical exposure for accurately encoding retinal images as biological signals at the level of the RPE is in the order of minutes to hours, depending on the nature of the visual manipulations.

Project description:PurposeThe retina plays an important regulatory role in ocular growth. To screen for new retinal candidate genes that could be involved in the inhibition of ocular growth, we used chick microarrays to analyze the changes in retinal mRNA expression after myopic defocus was imposed by positive lens wear.MethodsFour male white leghorn chicks, aged nine days, wore +6.9D spectacle lenses over both eyes for 24 h. Four untreated age-matched male chicks from the same batch served as controls. The chicks were euthanized, and retinas from both eyes of each chick were pooled. RNA was isolated and labeled cRNA was prepared. These samples were hybridized to Affymetrix GeneChip Chicken Genome arrays with more than 28,000 characterized genes. After comparison of multiple normalization methods, GC-RMA and a false-discovery rate of 6% was chosen for normalization of the data. The expression of 16 candidate genes was further studied, using semiquantitative real-time RT-PCR. In addition, the expression of the mRNA of some of these candidate genes was assessed in chicks that wore either +6.9D lenses for 4 h or -7D lenses for 24 h.Results123 transcripts were found to be differentially expressed (p<0.05; at least 1.5-fold change in expression level), with an absolute mean fold-change of 1.97+/-1.16 (mean+/-standard deviation). Nine of the sixteen genes that were examined by real-time RT-PCR were validated. Regardless of whether positive or negative lenses were worn, six of these nine genes were regulated in the same direction after 24 h: arginyltransferase 1 (ATE1), E74-like factor 1 (ELF1), growth factor receptor-bound protein 2 (GRB2), SHQ1 homolog (S. cerevisiae) (SHQ1), spectrin, beta, non-erythrocytic 1 (SPTBN1), prepro-urotensin II-related peptide (pp-URP). Three genes responded differently to positive and negative lens treatment after 24 h: ATP-binding cassette, sub-family C, member 10 (ABCC10), CD226 molecule (CD226) and oxysterol binding protein 2 (OSBP2).ConclusionsThe validated genes that were regulated only by myopic defocus may represent elements in a pathway generating a "stop-signal" for eye growth. Some of the genes identified in this study have so far not been described in the retina. Further investigation of their function may improve the understanding of the signaling cascades in emmetropization. More general, published microarray data are variable among different animal models (mouse, chick, monkeys), tissues (retina, retina/retinal pigment epithelium), treatments (diffusers, lenses, lid-suture), as well as different treatment durations (hours, days), and comparisons remain difficult. That only a small number of common genes were found emphasizes the need for careful normalization of the experimental parameters.

Project description:Studies on Parkinson's disease (PD) are becoming very popular on multidisciplinary platforms. The development of predictable telemonitored early detection models has become closely related to many different research areas. The aim of this article is to develop a visual performance test that can examine the effects of Parkinson's disease on the visual cortex, which can be a subtitle scoring test in UPDRS. However, instead of showing random images and asking for discrepancies between them, it is expected that the questions to be asked to patients should be provable in the existing cortex models, should be deduced between the images, and produce a reference threshold value to compare with the practical results. In a developed test, horizontal and vertical motion blur orientation was applied to natural image samples, and then neural outputs were produced by representing three (original-horizontal-vertical) image groups with the Layer 4 (L4) cortex model. This image representation is then compared with a filtering model which is very similar to thalamus' functionality. Thus, the linear problem-solving performance of the L4 cortex model is also addressed in the study. According to the obtained classification results, the L4 model produces high-performance success rates compared to the thalamic model, which shows the adaptation power of the visual cortex on the image pattern differences. In future studies, developed motion-based visual tests are planned to be applied to PD patient groups/controls, and their performances with mathematical threshold values will be examined.
