Working towards a conservation plan for fish parasites: Cyprinid parasites from the south African cape fold freshwater ecoregion as a case study.

ABSTRACT: The preservation of the world's biodiversity for future generations has been a global objective for many years, with the establishment of the International Union for Conservation of Nature (IUCN) Red List of Threatened Species in 1964. However, the conservation of parasites is a more recent development and, due to the difficulty in obtaining data and studying some of the parasitic species, comes its own series of challenges. Using parasites of cyprinid hosts (one critically endangered, one endangered and three near threatened) collected from South Africa's Cape Fold freshwater ecoregion (CF) as a case study, this paper discusses the challenges and possible solutions for implementing a fish parasite conservation project. Novel data on the fish parasites (1819 metazoan parasite individuals, representing the Acanthocephala, Cestoda, Copepoda, Digenea, Monogenea and Nematoda) of the CF are provided from the five endemic hosts, Cheilobarbus serra (Peters, 1864), Labeobarbus seeberi (Gilchrist et Thompson, 1913), Pseudobarbus phlegethon (Barnard, 1938), Sedercypris calidus (Barnard, 1938), and Sedercypris erubescens (Skelton, 1974). Conservation statuses for selected parasite taxa are also proposed based on the conservation statuses of the fish hosts, according to the Conservation Assessment Methodology for Animal Parasites (CAMAP).

Project description:The Eastern Cape redfin, Pseudobarbus afer, has long been considered to be a single widespread and variable species occurring in multiple isolated river systems in the Cape Fold Ecoregion (CFE) at the southern tip of Africa. Mitochondrial cytochrome b and control region sequence data of individuals from populations currently assigned to Pseudobarbus afer across the species' distribution range revealed existence of four deeply divergent taxonomic units: (i) the Mandela lineage confined to the Sundays, Swartkops and Baakens river systems, (ii) the Krom lineage endemic to the Krom River system, (iii) the St Francis lineage occurring in the Gamtoos and adjacent river systems, and (iv) the Forest lineage occurring in several coastal river systems from the Tsitsikamma to the Klein Brak River system. The Forest lineage is closely related to Pseudobarbus phlegethon from the Olifants River system on the west coast of South Africa, suggesting that it does not belong to Pseudobarbus afer s.l. Herein we focus on the three lineages within the Pseudobarbus afer s.l. complex and provide new diagnosis for Pseudobarbus afer s.s (Mandela lineage), revalidate Pseudobarbus senticeps (Krom lineage) as a distinct species, and describe a new species Pseudobarbus swartzi (St Francis lineage). The three species exhibit subtle differences, which explains why they were previously considered to represent a single variable and widespread species. Pseudobarbus senticeps differs from both Pseudobarbus afer and Pseudobarbus swartzi by having fewer (i.e. larger) scales (25-33, mode 29 lateral line scale series; 10-12, mode 11 circumpeduncular scales) and presence of a lateral stripe which terminates in a conspicuous triangular blotch at the base of the caudal fin. Long barbels which reach or surpass the vertical through the posterior edge of the eye further separate Pseudobarbus senticeps from Pseudobarbus afer s.s. which possesses simple short barbels which do not reach the vertical through the posterior margin of the eye. Pseudobarbus afer s.s differs from Pseudobarbus swartzisp. n. by possession of fewer scale rows along the lateral line (29-35, mode 32 vs 34-37, mode 36 in Pseudobarbus swartzi), fewer scales around the caudal peduncle (12-16, mode 12 vs 13-17, mode 16 in Pseudobarbus swartzi) and a distinct mesh or net-like pigmentation pattern on latero-ventral scales.

Project description:BackgroundHost specificity in parasites represents the extent to which a parasite's distribution is limited to certain host species. Considering host-specific parasites of primarily freshwater fish (such as gill monogeneans), their biogeographical distribution is essentially influenced by both evolutionary and ecological processes. Due to the limited capacity for historical dispersion in freshwater fish, their specific coevolving parasites may, through historical host-parasite associations, at least partially reveal the historical biogeographical routes (or historical contacts) of host species. We used Dactylogyrus spp., parasites specific to cyprinid fish, to infer potential historical contacts between Northwest African and European and Asian cyprinid faunas. Using phylogenetic reconstruction, we investigated the origin(s) of host-specific Dactylogyrus spp. parasitizing Northwest African and Iberian cyprinid species.ResultsIn accordance with hypotheses on the historical biogeography of two cyprinid lineages in Northwest Africa, Barbini (Luciobarbus) and Torini (Carasobarbus), we demonstrated the multiple origins of Northwest African Dactylogyrus. Dactylogyrus spp. of Carasobarbus spp. originated from Asian cyprinids, while Dactylogyrus spp. of Luciobarbus spp. originated from European cyprinids. This indicates the historical Northern route of Dactylogyrus spp. dispersion to Northwest African Luciobarbus species rather than the Southern route, which is currently widely accepted for Luciobarbus. In addition, both Northwest African cyprinid lineages were also colonized by Dactylogyrus marocanus closely related to Dactylogyrus spp. parasitizing African Labeo spp., which suggests a single host switch from African Labeonini to Northwest African Luciobarbus. We also demonstrated the multiple origins of Dactylogyrus spp. parasitizing Iberian Luciobarbus species. One Iberian Dactylogyrus group was phylogenetically closely related to Dactylogyrus of Moroccan Carasobarbus, while the second was related to Dactylogyrus of Moroccan Luciobarbus.ConclusionsOur study confirms the different origins of two Northwest African cyprinid lineages. It suggests several independent historical contacts between European Iberian Luciobarbus and two lineages of Northwest African cyprinids, these contacts associated with host switches of Dactylogyrus parasites.

Project description:BackgroundMany fish species have been introduced in wild ecosystems around the world to provide food or leisure, deliberately or from farm escapes. Some of those introductions have had large ecological effects. The north American native rainbow trout (Oncorhynchus mykiss Walbaum, 1792) is one of the most widely farmed fish species in the world. It was first introduced in Spain in the late 19th century for sport fishing (Elvira 1995) and nowadays is used there for both fishing and aquaculture. On the other hand, the European native brown trout (Salmo trutta L.) is catalogued as vulnerable in Spain. Detecting native and invasive fish populations in ecosystem monitoring is crucial, but it may be difficult from conventional sampling methods such as electrofishing. These techniques encompass some mortality, thus are not adequate for some ecosystems as the case of protected areas. Environmental DNA (eDNA) analysis is a sensitive and non-invasive method that can be especially useful for rare and low-density species detection and inventory in water bodies.MethodsIn this study we employed two eDNA based methods (qPCR and nested PCR-RFLP) to detect salmonid species from mountain streams within a protected area, The Biosphere Reserve and Natural Park of Redes (Upper Nalón Basin, Asturias, Northern Spain), where brown trout is the only native salmonid. We also measured some habitat variables to see how appropriate for salmonids the area is. The sampling area is located upstream impassable dams and contains one rainbow trout fish farm.ResultsEmploying qPCR methodology, brown trout eDNA was detected in all the nine sampling sites surveyed, while nested PCR-RFLP method failed to detect it in two sampling points. Rainbow trout eDNA was detected with both techniques at all sites in the Nalón River' (n1, n2 and n3). Salmonid habitat units and water quality were high from the area studied.DiscussionIn this study, a high quantity of rainbow trout eDNA was found upstream and downstream of a fish farm located inside a Biosphere Reserve. Unreported escapes from the fish farm are a likely explanation of these results. Since salmonid habitat is abundant and the water quality high, the establishment of rainbow trout populations would be favored should escapes occur. Environmental DNA has here proved to be a valuable tool for species detection in freshwater environments, and the probe-based qPCR highly sensitive technique for detection of scarce species. We would recommend this method for routine monitoring and early detection of introduced species within natural reserves.
