Electrochemical Nitrogen Fixation for Green Ammonia: Recent Progress and Challenges.

ABSTRACT: Ammonia, a key feedstock used in various industries, has been considered a sustainable fuel and energy storage option. However, NH₃ production via the conventional Haber-Bosch process is costly, energy-intensive, and significantly contributing to a massive carbon footprint. An electrochemical synthetic pathway for nitrogen fixation has recently gained considerable attention as NH₃ can be produced through a green process without generating harmful pollutants. This review discusses the recent progress and challenges associated with the two relevant electrochemical pathways: direct and indirect nitrogen reduction reactions. The detailed mechanisms of these reactions and highlight the recent efforts to improve the catalytic performances are discussed. Finally, various promising research strategies and remaining tasks are presented to highlight future opportunities in the electrochemical nitrogen reduction reaction.

Project description:Photocatalytic nitrogen reduction reaction (NRR) to ammonia holds a great promise for substituting the traditional energy-intensive Haber-Bosch process, which entails sunlight as an inexhaustible resource and water as a hydrogen source under mild conditions. Remarkable progress has been achieved regarding the activation and solar conversion of N2 to NH3 with the rapid development of emerging photocatalysts, but it still suffers from low efficiency. A comprehensive review on photocatalysts covering tungsten and related metals as well as their broad ranges of alloys and compounds is lacking. This article aims to summarize recent advances in this regard, focusing on the strategies to enhance the photocatalytic performance of tungsten and related metal semiconductors for the NRR. The fundamentals of solar-to-NH3 photocatalysis, reaction pathways, and NH3 quantification methods are presented, and the concomitant challenges are also revealed. Finally, we cast insights into the future development of sustainable NH3 production, and highlight some potential directions for further research in this vibrant field.

Recent Progress in Electrochemical Immunosensors.

Project description:Biosensors used for medical diagnosis work by analyzing physiological fluids. Antibodies have been frequently used as molecular recognition molecules for the specific binding of target analytes from complex biological solutions. Electrochemistry has been introduced for the measurement of quantitative signals from transducer-bound analytes for many reasons, including good sensitivity. Recently, numerous electrochemical immunosensors have been developed and various strategies have been proposed to detect biomarkers. In this paper, the recent progress in electrochemical immunosensors is reviewed. In particular, we focused on the immobilization methods using antibodies for voltammetric, amperometric, impedimetric, and electrochemiluminescent immunosensors.

Recent Progress in Electrochemical HbA1c Sensors: A Review.

Project description:This article reviews recent progress made in the development of electrochemical glycated hemoglobin (HbA1c) sensors for the diagnosis and management of diabetes mellitus. Electrochemical HbA1c sensors are divided into two categories based on the detection protocol of the sensors. The first type of sensor directly detects HbA1c by binding HbA1c on the surface of an electrode through bio-affinity of antibody and boronic acids, followed by an appropriate mode of signal transduction. In the second type of sensor, HbA1c is indirectly determined by detecting a digestion product of HbA1c, fructosyl valine (FV). Thus, the former sensors rely on the selective binding of HbA1c to the surface of the electrodes followed by electrochemical signaling in amperometric, voltammetric, impedometric, or potentiometric mode. Redox active markers, such as ferrocene derivatives and ferricyanide/ferrocyanide ions, are often used for electrochemical signaling. For the latter sensors, HbA1c must be digested in advance by proteolytic enzymes to produce the FV fragment. FV is electrochemically detected through catalytic oxidation by fructosyl amine oxidase or by selective binding to imprinted polymers. The performance characteristics of HbA1c sensors are discussed in relation to their use in the diagnosis and control of diabetic mellitus.

Searching for the Rules of Electrochemical Nitrogen Fixation.

Project description:Li-mediated ammonia synthesis is, thus far, the only electrochemical method for heterogeneous decentralized ammonia production. The unique selectivity of the solid electrode provides an alternative to one of the largest heterogeneous thermal catalytic processes. However, it is burdened with intrinsic energy losses, operating at a Li plating potential. In this work, we survey the periodic table to understand the fundamental features that make Li stand out. Through density functional theory calculations and experimentation on chemistries analogous to lithium (e.g., Na, Mg, Ca), we find that lithium is unique in several ways. It combines a stable nitride that readily decomposes to ammonia with an ideal solid electrolyte interphase, balancing reagents at the reactive interface. We propose descriptors based on simulated formation and binding energies of key intermediates and further on hard and soft acids and bases (HSAB principle) to generalize such features. The survey will help the community toward electrochemical systems beyond Li for nitrogen fixation.

Control of nitrogen fixation and ammonia excretion in Azorhizobium caulinodans.

Project description:Due to the costly energy demands of nitrogen (N) fixation, diazotrophic bacteria have evolved complex regulatory networks that permit expression of the catalyst nitrogenase only under conditions of N starvation, whereas the same condition stimulates upregulation of high-affinity ammonia (NH3) assimilation by glutamine synthetase (GS), preventing excess release of excess NH3 for plants. Diazotrophic bacteria can be engineered to excrete NH3 by interference with GS, however control is required to minimise growth penalties and prevent unintended provision of NH3 to non-target plants. Here, we tested two strategies to control GS regulation and NH3 excretion in our model cereal symbiont Azorhizobium caulinodans AcLP, a derivative of ORS571. We first attempted to recapitulate previous work where mutation of both PII homologues glnB and glnK stimulated GS shutdown but found that one of these genes was essential for growth. Secondly, we expressed unidirectional adenylyl transferases (uATs) in a ΔglnE mutant of AcLP which permitted strong GS shutdown and excretion of NH3 derived from N2 fixation and completely alleviated negative feedback regulation on nitrogenase expression. We placed a uAT allele under control of the NifA-dependent promoter PnifH, permitting GS shutdown and NH3 excretion specifically under microaerobic conditions, the same cue that initiates N2 fixation, then deleted nifA and transferred a rhizopine nifAL94Q/D95Q-rpoN controller plasmid into this strain, permitting coupled rhizopine-dependent activation of N2 fixation and NH3 excretion. This highly sophisticated and multi-layered control circuitry brings us a step closer to the development of a "synthetic symbioses" where N2 fixation and NH3 excretion could be specifically activated in diazotrophic bacteria colonising transgenic rhizopine producing cereals, targeting delivery of fixed N to the crop while preventing interaction with non-target plants.

Electrochemical Reduction of Nitrogen to Ammonia Using Zinc Telluride.

Project description:Electrosynthesis of ammonia (NH3), an important constituent molecule of various commercial fertilizers, is a promising and sustainable alternative strategy compared with the century-old Haber-Bosch process. Herein, zinc telluride (ZnTe) is demonstrated as an efficient electrocatalyst for reducing nitrogen (N2) under ambient conditions to NH3. In this simple chemical strategy, Zn preferentially binds N2 over hydrogen (H2), and Te, by virtue of its superior electronic properties, enhances the electrocatalytic activity of ZnTe. The analysis of the X-ray diffraction data using the Bravais-Friedel-Donnay-Harker (BFDH) theory predicted a crystal geometry with the active electrocatalytic sites predominantly confined to the (111) planes of ZnTe. The preferential binding of nitrogen (N2; adsorption energy = -0.043 eV) over hydrogen (H2, adsorption energy = -0.028 eV) to Zn on the (111) plane of ZnTe is further confirmed by density functional theory. The ZnTe catalyst is observed to be stable in the acidic medium and delivers a very high yield of NH3 (19.85 μg/h-1 mgcat -1) and a Faradaic efficiency of 6.24% at -0.6 V (versus RHE). Additional verification experiments do not reveal the formation of side products (such as NH2-NH2) during N2 reduction by ZnTe. Further, density functional theory calculations strongly predict that the electrocatalytic reduction of N2 to NH3 by ZnTe preferentially occurs via the alternate pathway.

Human lung development: recent progress and new challenges.

Project description:Recent studies have revealed biologically significant differences between human and mouse lung development, and have reported new in vitro systems that allow experimental manipulation of human lung models. At the same time, emerging clinical data suggest that the origins of some adult lung diseases are found in embryonic development and childhood. The convergence of these research themes has fuelled a resurgence of interest in human lung developmental biology. In this Review, we discuss our current understanding of human lung development, which has been profoundly influenced by studies in mice and, more recently, by experiments using in vitro human lung developmental models and RNA sequencing of human foetal lung tissue. Together, these approaches are helping to shed light on the mechanisms underlying human lung development and disease, and may help pave the way for new therapies.

Lung neuroendocrine neoplasms: recent progress and persistent challenges.

Project description:This review summarizes key recent developments relevant to the pathologic diagnosis of lung neuroendocrine neoplasms, including carcinoids, small cell lung carcinoma (SCLC), and large cell neuroendocrine carcinoma (LCNEC). Covered are recent insights into the biological subtypes within each main tumor type, progress in pathological diagnosis and immunohistochemical markers, and persistent challenging areas. Highlighted topics include highly proliferative carcinoids and their distinction from small cell and large cell neuroendocrine carcinomas (NECs), the evolving role of Ki67, the update on the differential diagnosis of NEC to include thoracic SMARCA4-deficient undifferentiated tumors, the recent data on SCLC transcriptional subtypes with the emergence of POU2F3 as a novel marker for the diagnosis of SCLC with low/negative expression of standard neuroendocrine markers, and the update on the diagnosis of LCNEC, particularly in biopsies. There has been remarkable recent progress in the understanding of the genetic and expression-based profiles within each type of lung neuroendocrine neoplasm, and it is hoped that these insights will enable the development of novel diagnostic, prognostic, and predictive biomarkers to aid in the pathologic assessment of these tumors in the future.

Discovering new medicines targeting helicases: challenges and recent progress.

Project description:Helicases are ubiquitous motor proteins that separate and/or rearrange nucleic acid duplexes in reactions fueled by adenosine triphosphate (ATP) hydrolysis. Helicases encoded by bacteria, viruses, and human cells are widely studied targets for new antiviral, antibiotic, and anticancer drugs. This review summarizes the biochemistry of frequently targeted helicases. These proteins include viral enzymes from herpes simplex virus, papillomaviruses, polyomaviruses, coronaviruses, the hepatitis C virus, and various flaviviruses. Bacterial targets examined include DnaB-like and RecBCD-like helicases. The human DEAD-box protein DDX3 is the cellular antiviral target discussed, and cellular anticancer drug targets discussed are the human RecQ-like helicases and eIF4A. We also review assays used for helicase inhibitor discovery and the most promising and common helicase inhibitor chemotypes, such as nucleotide analogues, polyphenyls, metal ion chelators, flavones, polycyclic aromatic polymers, coumarins, and various DNA binding pharmacophores. Also discussed are common complications encountered while searching for potent helicase inhibitors and possible solutions for these problems.
