Impact of a Whey Protein Hydrolysate Treated by Electrodialysis with Ultrafiltration Membrane on the Development of Metabolic Syndrome and the Modulation of Gut Microbiota in Mice.

ABSTRACT: The development of Metabolic Syndrome (MetS) affects a large number of people around the world and represents a major issue in the field of health. Thus, it is important to implement new strategies to reduce its prevalence, and various approaches are currently under development. Recently, an eco-friendly technology named electrodialysis with ultrafiltration membrane (EDUF) was used successfully for the first time at a semi-industrial scale to produce three fractions concentrated in bioactive peptides (BPs) from an enzymatically hydrolyzed whey protein concentrate (WPC): the initial (F1), the final (F2) and the recovery fraction (F3), and it was demonstrated in vitro that F3 exhibited interesting DPP-IV inhibitory effects. Therefore, the present study aimed to evaluate the effect of each fraction on in vivo models of obesity. A daily dose of 312.5 mg/kg was administered to High Fat/High Sucrose diet (HFHS) induced C57BL6/J mice for eight weeks. The physiological parameters of each group and alterations of their gut microbiota by the fractions were assessed. Little effect of the different fractions was demonstrated on the physiological state of the mice, probably due to the digestion process of the BP content. However, there were changes in the gut microbiota composition and functions of mice treated with F3.

Project description:Acid saccharification of hemicelluloses offers promising pathways to sustainably diversify the revenue of the lignocellulose biorefinery industry. Electrodialysis to separate inorganic acids from acid hydrolysate in the hemicellulose saccharification process could realize the recovery of sulfuric acid, and significantly reduced the chemical consumption than the traditional ion exchange resins method. In this work, the deacidification of corncob acid hydrolysate was conducted by a homemade electrodialysis apparatus. The results showed that: (1) more than 99% of acid can be removed through the electrodialysis process; (2) A non-negligible membrane fouling occurred during the electrodialysis process, which aggravated with the repeated batch running The final global system resistance rose from 15.8 Ω (1st batch) to 43.9 Ω (10th batch), and the treatment ending time was delayed from 120 min (1st batch) to 162 min (10th batch); (4) About 90% of protein, 70% of ferulate acid, and 80% of p-coumarate acid precipitated from the corncob acid hydrolysate during the electrodialysis process. The zeta potential of corncob acid hydrolysate changed from a positive value to a negative value, and an isoelectric point around pH 2.3 was reached. HSQC, FTTR, and GPC, along with SEM and EDS analysis, revealed that the fouling layers mostly consisted of hydrolysates of protein and lignin. The result of HSQC indicated that the membrane foulant may exist in the form of lignin-carbohydrate complexes, as the lignin component of the membrane foulant is in the form of p-coumarate and ferulate. From the result of FTIR, a strong chemical bonding, such as a covalent linkage, existed between the lignin and protein in the membrane foulant. Throughout the electrodialysis process, the increased pH decreased the stability of colloidal particles, including lignin and proteins. Destabilized colloidal particles started to self-aggregate and form deposits on the anion exchange membrane's surface. Over time, these deposits covered the entire membrane surface and the spaces between the membranes. Eventually, they attached to the surface of the cation exchange membrane. In the end, a suggestion to control and minimize membrane fouling in this process was discussed: lower pH as a process endpoint and a post-treatment method.

Project description:Berberine is an isoquinoline alkaloid found in plants. It presents a wide range of pharmacological activities, including anti-inflammatory and antioxidant properties, despite a low oral bioavailability. Growing evidence suggests that the gut microbiota is the target of berberine, and that the microbiota metabolizes berberine to active metabolites, although little evidence exists in the specific species involved in its therapeutic effects. This study was performed to detail the bidirectional interactions of berberine with the broiler chicken gut microbiota, including the regulation of gut microbiota composition and metabolism by berberine and metabolization of berberine by the gut microbiota, and how they contribute to berberine-mediated effects on gut health. As previous evidence showed that high concentrations of berberine may induce dysbiosis, low (0.1 g/kg feed), middle (0.5 g/kg feed) and high (1 g/kg feed) doses were here investigated. Low and middle doses of in-feed berberine stimulated potent beneficial bacteria from the Lachnospiraceae family in the large intestine of chickens, while middle and high doses tended to increase villus length in the small intestine. Plasma levels of the berberine-derived metabolites berberrubine, thalifendine and demethyleneberberine were positively correlated with the villus length of chickens. Berberrubine and thalifendine were the main metabolites of berberine in the caecum, and they were produced in vitro by the caecal microbiota, confirming their microbial origin. We show that members of the genus Blautia could demethylate berberine into mainly thalifendine, and that this reaction may stimulate the production of short-chain fatty acids (SCFAs) acetate and butyrate, via acetogenesis and cross-feeding respectively. We hypothesize that acetogens such as Blautia spp. are key bacteria in the metabolization of berberine, and that berberrubine, thalifendine and SCFAs play a significant role in the biological effect of berberine.
