Just Keep Rolling?-An Encompassing Review towards Accelerated Vaccine Product Life Cycles.

ABSTRACT: In light of the recent pandemic, several COVID-19 vaccines were developed, tested and approved in a very short time, a process that otherwise takes many years. Above all, these efforts have also unmistakably revealed the capacity limits and potential for improvement in vaccine production. This review aims to emphasize recent approaches for the targeted rapid adaptation and production of vaccines from an interdisciplinary, multifaceted perspective. Using research from the literature, stakeholder analysis and a value proposition canvas, we reviewed technological innovations on the pharmacological level, formulation, validation and resilient vaccine production to supply bottlenecks and logistic networks. We identified four main drivers to accelerate the vaccine product life cycle: computerized candidate screening, modular production, digitized quality management and a resilient business model with corresponding transparent supply chains. In summary, the results presented here can serve as a guide and implementation tool for flexible, scalable vaccine production to swiftly respond to pandemic situations in the future.

Project description:Substantial amounts of post-consumer scrap are exported to other regions or lost during recovery and remelting, and both export and losses pose a constraint to desires for having regionally closed material cycles. To quantify the challenges and trade-offs associated with closed-loop metal recycling, we looked at the material cycles from the perspective of a single material unit and trace a unit of material through several product life cycles. Focusing on steel, we used current process parameters, loss rates, and trade patterns of the steel cycle to study how steel that was originally contained in high quality applications such as machinery or vehicles with stringent purity requirements gets subsequently distributed across different regions and product groups such as building and construction with less stringent purity requirements. We applied MaTrace Global, a supply-driven multiregional model of steel flows coupled to a dynamic stock model of steel use. We found that, depending on region and product group, up to 95% of the steel consumed today will leave the use phase of that region until 2100, and that up to 50% can get lost in obsolete stocks, landfills, or slag piles until 2100. The high losses resulting from business-as-usual scrap recovery and recycling can be reduced, both by diverting postconsumer scrap into long-lived applications such as buildings and by improving the recovery rates in the waste management and remelting industries. Because the lifetimes of high-quality (cold-rolled) steel applications are shorter and remelting occurs more often than for buildings and infrastructure, we found and quantified a tradeoff between low losses and high-quality applications in the steel cycle. Furthermore, we found that with current trade patterns, reduced overall losses will lead to higher fractions of secondary steel being exported to other regions. Current loss rates, product lifetimes, and trade patterns impede the closure of the steel cycle.

Project description:BackgroundGlobal vaccination coverage rates have remained around 85% for the past several years. Increasing immunization coverage rates requires an effective cold chain to maintain vaccine potency. Remote temperature monitoring (RTM) technology for vaccine refrigerators has shown promise for improving the ability of supply systems to maintain optimal temperature conditions to ensure potent vaccines reach the end users.MethodsA pilot study of RTM technology and data use teams was implemented in 36 study sites in Kenya. Data were collected at baseline and endline points over a 3-month baseline and 7-month implementation period. Data included 44 qualitative interviews, process logs, meeting minutes from data use team meetings, and quantitative temperature and power data from the RTM devices.ResultsThe ability of cold chain equipment to maintain World Health Organization-recommended temperatures in study sites improved markedly between the baseline and implementation periods, resulting in an improvement in total time spent in the correct range from 83.9% in the baseline period to 90.9% in the intervention period and an improvement in time spent in the too cold range from 6.5% to 1.5%. Friedman tests revealed that differences in time spent in the correct range and time spent in the too cold range during the course of the study were statistically significant (P<.001 and P=.04, respectively). Qualitative and quantitative data suggest that this improvement was due to a combination of improved responsiveness to temperature excursions at the facility level, resulting from SMS alarms for temperature excursion periods, and improved ability at the management level to recognize and address recurring problems.ConclusionThe combination of using RTM technology with a structured data review process by a management team is a promising approach for improving cold chain outcomes. Future research examining the added value of each of the technological and behavioral components separately is needed.

Project description:BackgroundAntipredator behaviors are theoretically subjected to a balance by which their display should be minimized when their benefits do not outweigh their costs. Such costs may be not only energetic, but also entail a reduction in the time available for other fitness-enhancing behaviors. However, these behaviors are only beneficial under predation risk. Therefore, antipredator behaviors are predicted to be maximized under strong predation risk. Moreover, predation pressure can differ among individuals according to traits such as sex or body size, if these traits increase vulnerability. Antipredator behaviors are expected to be maximized in individuals whose traits make them more conspicuous to predators. However, how sex, body size and antipredator behaviors interact is not always understood.MethodsIn this work, I tested the interaction between sex, body size and antipredator behavior in the common pill woodlouse (Armadillidium vulgare), which conglobate (i.e., they roll up their bodies almost conforming a sphere that conceals their appendages) in response to predator attacks. Specifically, I tested whether latency to unroll after a standardized mechanical induction was greater in animals exposed to predator chemical cues (toad feces) than in conspecifics exposed to cues of non-predatory animals (rabbits) or no chemical cues whatsoever (distilled water), incorporating sex and body mass in the analyses.ResultsIn agreement with my prediction, latency to unroll was greater in individuals exposed to predator chemical cues. In other words, these animals engage in conglobation for longer under perceived predator vicinity. However, this result was only true for males. This sexual dimorphism in antipredator behavior could result from males being under greater predation risk than females, thus having evolved more refined antipredator strategies. Indeed, males of this species are known to actively search for females, which makes them more prone to superficial ground mobility, and likely to being detected by predators. Body size was unrelated to latency to unroll. As a whole, these results support the hypothesis that antipredator behavior is tuned to predator cues in a way consistent with a balance between costs and benefits, which might differ between the sexes.
