Fecal Microbiota and Diet Composition of Buryatian Horses Grazing Warm- and Cold-Season Grass Pastures.

ABSTRACT: The Buryatian horse is an ancient breed and, as an indigenous breed, they have unique adaptive abilities to use scarce pastures, graze in winter, and survive in harsh conditions with minimal human care. In this study, fecal microbiota of Buryatian horses grazing in the warm and cold seasons were investigated using NGS technology on the Illumina MiSeq platform. We hypothesized that the composition of microbial communities in the feces of horses maintained on pasture would change in the different seasons, depending on the grass availability and different plant diets. We conducted microhistological fecal studies of horse diet composition on steppe pasture. The alpha diversity analysis showed horses had a more abundant and diverse gut microbiota in summer. There were significant effects on the beta diversity of microbial families, which were clustered by the warm and cold season in a principal coordinate analysis (PCoA), with 45% of the variation explained by two principal coordinates. This clustering by season was further confirmed by the significant differences observed in the relative abundances of microbial families and genera. The obtained results can serve as an experimental substantiation for further study of the impact of pasture grasses, which have a pharmacological effect, on the diversity of the gut microbiome and horse health.

Project description:BackgroundDiet is a key driver of equine hindgut microbial community structure and composition. The aim of this study was to characterize shifts in the fecal microbiota of grazing horses during transitions between forage types within integrated warm- (WSG) and cool-season grass (CSG) rotational grazing systems (IRS). Eight mares were randomly assigned to two IRS containing mixed cool-season grass and one of two warm-season grasses: bermudagrass [Cynodon dactylon (L.) Pers.] or crabgrass [Digitaria sanguinalis (L.) Scop.]. Fecal samples were collected during transitions from CSG to WSG pasture sections (C-W) and WSG to CSG (W-C) on days 0, 2, 4, and 6 following pasture rotation and compared using 16S rRNA gene sequencing.ResultsRegardless of IRS or transition (C-W vs. W-C), species richness was greater on day 4 and 6 in comparison to day 0 (P < 0.05). Evenness, however, did not differ by day. Weighted UniFrac also did not differ by day, and the most influential factor impacting β-diversity was the individual horse (R2 ≥ 0.24; P = 0.0001). Random forest modeling was unable to accurately predict days within C-W and W-C, but could predict the individual horse based on microbial composition (accuracy: 0.92 ± 0.05). Only three differentially abundant bacterial co-abundance groups (BCG) were identified across days within all C-W and W-C for both IRS (W ≥ 126). The BCG differing by day for all transitions included amplicon sequence variants (ASV) assigned to bacterial groups with known fibrolytic and butyrate-producing functions including members of Lachnospiraceae, Clostridium sensu stricto 1, Anaerovorax the NK4A214 group of Oscillospiraceae, and Sarcina maxima. In comparison, 38 BCG were identified as differentially abundant by horse (W ≥ 704). The ASV in these groups were most commonly assigned to genera associated with degradation of structural carbohydrates included Rikenellaceae RC9 gut group, Treponema, Christensenellaceae R-7 group, and the NK4A214 group of Oscillospiraceae. Fecal pH also did not differ by day.ConclusionsOverall, these results demonstrated a strong influence of individual horse on the fecal microbial community, particularly on the specific composition of fiber-degraders. The equine fecal microbiota were largely stable across transitions between forages within IRS suggesting that the equine gut microbiota adjusted at the individual level to the subtle dietary changes imposed by these transitions. This adaptive capacity indicates that horses can be managed in IRS without inducing gastrointestinal dysfunction.

Project description:Land management by seasonal migratory herding, traditionally implemented by pastoralists, has allowed semi-arid rangeland ecosystems to remain productive and resilient to highly erratic, seasonal rainfall patterns. Changing pastoralist practices and rainfall patterns due to climate change have the potential to negatively influence the resilience of rangelands. To test the impact of different communal rangeland practices and increasing frequency of disturbance on the rangelands' vegetation, we established a clipping experiment in three rangeland management types: rainy season rangeland, dry season rangeland and seasonal exclosures, in the Maasai Steppe, northern Tanzania. Across these rangeland management types, we tested two clipping frequencies monthly vs. once per growing period and either fenced or open to grazing (herbivory) over two consecutive growing periods. We found that rainy season rangeland exhibited lowest regrowth rates and highest proportion of bare ground across herbivory, clipping frequency and growing period. When rainfall was low, seasonal exclosures showed lowest proportion of bare ground across herbivory and clipping frequency and plots that were seasonally clipped and excluded herbivores generated significantly more grass biomass, higher grass regrowth rates and lowest proportion of bare ground compared to all other treatments. Excluding herbivores had little impact on the vegetation biomass and cover but had a positive impact on grass regrowth rates when rainfall was high. Seasonal clipping allowed for lower proportion of bare ground, and in most cases higher regrowth rates and higher biomass compared to monthly clipping. We conclude that traditional seasonal migratory herding is a relevant concept to sustain rangeland productivity and resilience under increasing intensity of disturbance if grazing pressure on rainy season rangelands is adapted to rainfall conditions and sufficient resting time after heavy defoliation is granted. Dry season grazing (including rainy season resting) proved to be the most sustainable concept to maintain regrowth and control erosion.

Project description:Gastrointestinal strongyles are a major threat to horses' health and welfare. Given that strongyles inhabit the same niche as the gut microbiota, they may interact with each other. These beneficial or detrimental interactions are unknown in horses and could partly explain contrasted susceptibility to infection between individuals. To address these questions, an experimental pasture trial with 20 worm-free female Welsh ponies (10 susceptible (S) and 10 resistant (R) to parasite infection) was implemented for 5 months. Fecal egg counts (FEC), hematological and biochemical data, body weight and gut microbiological composition were studied in each individual after 0, 24, 43, 92 and 132 grazing days. R and S ponies displayed divergent immunological profiles and slight differences in microbiological composition under worm-free conditions. After exposure to natural infection, the predicted R ponies exhibited lower FEC after 92 and 132 grazing days, and maintained higher levels of circulating monocytes and eosinophils, while lymphocytosis persisted in S ponies. Although the overall gut microbiota diversity and structure remained similar during the parasite infection between the two groups, S ponies exhibited a reduction of bacteria such as Ruminococcus, Clostridium XIVa and members of the Lachnospiraceae family, which may have promoted a disruption of mucosal homeostasis at day 92. In line with this hypothesis, an increase in pathobionts such as Pseudomonas and Campylobacter together with changes in several predicted immunological pathways, including pathogen sensing, lipid metabolism, and activation of signal transduction that are critical for the regulation of immune system and energy homeostasis were observed in S relative to R ponies. Moreover, S ponies displayed an increase in protozoan concentrations at day 92, suggesting that strongyles and protozoa may contribute to each other's success in the equine intestines. It could also be that S individuals favor the increase of these carbohydrate-degrading microorganisms to enhance the supply of nutrients needed to fight strongyle infection. Overall, this study provides a foundation to better understand the mechanisms that underpin the relationship between equines and natural strongyle infection. The profiling of horse immune response and gut microbiota should contribute to the development of novel biomarkers for strongyle infection.

Project description:Climate seasonality is a predominant constraint on the lifecycles of species in alpine and polar biomes. Assessing the response of these species to climate change thus requires taking into account seasonal constraints on populations. However, interactions between seasonality, weather fluctuations, and population parameters remain poorly explored as they require long-term studies with high sampling frequency. This study investigated the influence of environmental covariates on the demography of a corvid species, the alpine chough Pyrrhocorax graculus, in the highly seasonal environment of the Mont Blanc region. In two steps, we estimated: (1) the seasonal survival of categories of individuals based on their age, sex, etc., (2) the effect of environmental covariates on seasonal survival. We hypothesized that the cold season-and more specifically, the end of the cold season (spring)-would be a critical period for individuals, and we expected that weather and individual covariates would influence survival variation during critical periods. We found that while spring was a critical season for adult female survival, it was not for males. This is likely because females are dominated by males at feeding sites during snowy seasons (winter and spring), and additionally must invest energy in egg production. When conditions were not favorable, which seemed to happen when the cold season was warmer than usual, females probably reached their physiological limits. Surprisingly, adult survival was higher at the beginning of the cold season than in summer, which may result from adaptation to harsh weather in alpine and polar vertebrates. This hypothesis could be confirmed by testing it with larger sets of populations. This first seasonal analysis of individual survival over the full life cycle in a sedentary alpine bird shows that including seasonality in demographic investigations is crucial to better understand the potential impacts of climate change on cold ecosystems.
