Nickel-Imidazolium Low Transition Temperature Mixtures with Lewis-Acidic Character.

ABSTRACT: Low transition temperature mixtures (LTTMs) are a new generation of solvents that have found extensive application in organic synthesis. The interactions between the components often generate highly activated, catalytically active species, thus opening the possibility of using LTTMs as catalysts, rather than solvents. In this work, we introduce a nickel-based imidazolium LTTM, study its thermal behavior and explore its catalytic activity in the solvent-free allylation of heterocycles with allylic alcohols. This system is effective in this reaction, affording the corresponding products in excellent yield without the need for additional purifications, thus resulting in a very environmentally friendly protocol.

Freezing-Tolerant Supramolecular Adhesives from Tannic Acid-Based Low-Transition-Temperature Mixtures.

Project description:Natural polyphenols like tannic acid (TA) have recently emerged as multifunctional building blocks for designing advanced materials. Herein, we show the benefits of having TA in a dynamic liquid state using low-transition-temperature mixtures (LTTMs) for developing freezing-tolerant glues. TA was combined with betaine or choline chloride to create LTTMs, which direct the self-assembly of guanosine into supramolecular viscoelastic materials with high adhesion. Molecular dynamics simulations showed that the structural properties of the material are linked to strong hydrogen bonding in TA-betaine and TA-choline chloride mixtures. Notably, long-term and repeatable adhesion was achieved even at -196 °C due to the binding ability of TA's catechol and gallol units and the mixtures' glass transition temperature. Additionally, the adhesives demonstrated injectability and low toxicity against fibroblasts in vitro. These traits reveal the potential of these systems as bioadhesives for tissue repair, opening new avenues for creating multifunctional soft materials with bioactive properties.

Nickel-Catalyzed Decarbonylative Alkylation of Aroyl Fluorides Assisted by Lewis-Acidic Organoboranes.

Project description:Herein, nickel-catalyzed decarbonylative C-F bond alkylation of aroyl fluorides with organoboron reagents is reported. Aroyl fluorides are more chemically stable than the corresponding aroyl chlorides and can be readily synthesized from the corresponding carboxylic acids. The fluoronickel intermediate formed via oxidative addition interacts with Lewis-acidic trialkylboranes, and the subsequent decarbonylative alkylation proceeds. This new synthetic methodology allows 1,2-bifunctionalization of aromatic carboxylic acids via palladium-catalyzed ortho-C-H arylation. In addition, an unprecedented 1,4-nickel migration on ortho-arylated aroyl fluorides was observed. As a demonstration of the synthetic utility of the present reaction, the sequential 1-alkyl-2-arylation of 3-hydroxy-2-naphthoic acid was accomplished via chemoselective alkylation at a fluorocarbonyl moiety and the subsequent C-O bond arylation at an acetoxy group.

Predicting Thermal Decomposition Temperature of Binary Imidazolium Ionic Liquid Mixtures from Molecular Structures.

Project description:Ionic liquids (ILs) have been regarded as "designer solvents" because of their satisfactory physicochemical properties. The 5% onset decomposition temperature (T d,5%onset) is one of the most conservative but reliable indicators for characterizing the possible fire hazard of engineered ILs. This study is devoted to develop a quantitative structure-property relationship model for predicting the T d,5%onset of binary imidazolium IL mixtures. Both in silico design and data analysis descriptors and norm index were employed to encode the structural characteristics of binary IL mixtures. The subset of optimal descriptors was screened by combining the genetic algorithm with the multiple linear regression method. The resulting optimal prediction model was a four-variable multiple linear equation, with the average absolute error (AAE) for the external test set being 12.673 K. The results of rigorous model validations also demonstrated satisfactory model robustness and predictivity. The present study would provide a new reliable approach for predicting the thermal stability of binary IL mixtures.

Acidic extracellular pH promotes epithelial mesenchymal transition in Lewis lung carcinoma model.

Project description:BackgroundEpithelial mesenchymal transition (EMT) is thought to be an essential feature of malignant tumor cells when they spread into the stroma. Despite the extracellular acidity of tumor tissues, the effect of acidic extracellular pH (pH e ) on EMT in carcinoma models, including the Lewis lung carcinoma (LLC) model, remains unclear.MethodsHigh and low metastatic LLC variants were generated by repeated tail vein injection of metastatic cells. DMEM/F12 medium, which has been supplemented with 15 mM HEPES, 4 mM phosphoric acid, and 1 g/L NaHCO3 and adjusted to the desire pH with HCl or NaOH, was used for cell culture. EMT marker gene expression was determined by quantitative reverse transcription-polymerase chain reaction. Migration and invasion activities were analyzed by wound healing assay and the Boyden chamber assay through Matrigel®, respectively.ResultsLow metastatic variant LLCm1 cells showed a cobble-stone like morphology at pH e 7.4. At pH e 6.8, however, their morphology became fibroblastic, similar in shape to high metastatic variant LLCm4 cells. Steady state levels of matrix metalloproteinase-9 (Mmp9) mRNA were induced by acidic pH e , maximizing at pH 6.8, with the levels of Mmp9 mRNA higher in LLCm4 than in LLCm1 cells. Both variants showed decreased levels of E-cadherin and increased levels of vimentin at pH e 6.8. Acidic pH e also induced expression of mRNAs encoding the E-cadherin repressors, Zeb2, Twist1 and Twist2, as well as enhancing cell motility and in vitro invasion through Matrigel®.ConclusionsAcidic pH e can induce EMT in some types of carcinoma.

Therapeutic Liquid Formulations Based on Low Transition Temperature Mixtures for the Incorporation of Anti-Inflammatory Drugs.

Project description:Most nonsteroidal anti-inflammatory drugs (NSAIDs) present poor aqueous solubility, impairing their efficiency in physiological media. In this context, Low Transition Temperature Mixtures (LTTMs) are a promising platform to overcome drugs' poor solubility, forming therapeutic liquid formulations. In this work, the LTTMs of citric acid:L-arginine:water (C:A:W) and glycerol:sorbitol (G:S) were studied in terms of their features and assessed in terms of their ability to increase the solubility of six NSAIDs in physiological media. The physicochemical properties of LTTMs were characterized by state-of-art techniques commonly used for these systems. The cytotoxicity of G:S was also evaluated in L929 mouse fibroblasts and the viscosity, polarity, and pH properties of the studied mixtures were related to the solubility of NSAIDs. The pH and polarity were the parameters that most influenced the drugs' solubility. Ibuprofen, naproxen, ketoprofen, indomethacin, and flurbiprofen did not present any solubility improvement in the formulations tested. However, concentrated mixtures of C:A:W or G:S in the physiologic-mimicked media (PBS) rendered a celecoxib solubility 4 and 5 times higher than PBS, respectively. These therapeutic liquid formulations of celecoxib in C:A:W or G:S can be a promising tool to increase celecoxib's therapeutic efficiency in local applications.

Temperature-dependent reentrant phase transition of RNA-polycation mixtures.

Project description:Liquid-liquid phase separation (LLPS) of multivalent biopolymers is a ubiquitous process in biological systems and is of importance in bio-mimetic soft matter design. The phase behavior of biomolecules, such as proteins and nucleic acids, is typically encoded by the primary chain sequence and regulated by solvent properties. One of the most important physical modulators of LLPS is temperature. Solutions of proteins and/or nucleic acids have been shown to undergo liquid-liquid phase separation either upon cooling (with an upper critical solution temperature, UCST) or upon heating (with a lower critical solution temperature, LCST). However, many theoretical frameworks suggest the possibility of more complex temperature-dependent phase behaviors, such as an hourglass or a closed-loop phase diagram with concurrent UCST and LCST transitions. Here, we report that RNA-polyamine mixtures undergo a reentrant phase separation with temperature. Specifically, at low temperatures, RNA-polyamine mixtures form a homogenous phase. Increasing the temperature leads to the formation of RNA-polyamine condensates. A further increase in temperature leads to the dissolution of condensates, rendering a reentrant homogenous phase. This dual-response phase separation of RNA is not unique to polyamines but also observed with short cationic peptides. The immiscibility gap is controlled by the charge of the polycation, salt concentration, and mixture composition. Based on the existing theories of complex coacervation, our results point to a complex interplay between desolvation entropy, ion-pairing, and electrostatic interactions in dictating the closed-loop phase behavior of RNA-polycation mixtures.

Geometrically Constrained Cationic Low-Coordinate Tetrylenes: Highly Lewis Acidic σ-Donor Ligands in Catalytic Systems.

Project description:A novel non-innocent ligand class, namely cationic single-centre ambiphiles, is reported in the phosphine-functionalised cationic tetrylene Ni0 complexes, [PhR DippENi(PPh3 )3 ]+ (4 a/b (Ge) and 5 (Sn); PhR Dipp={[Ph2 PCH2 SiR2 ](Dipp)N}- ; R=Ph, i Pr; Dipp=2,6-i Pr2 C6 H3 ). The inherent electronic nature of low-coordinate tetryliumylidenes, combined with the geometrically constrained [N-E-Ni] bending angle enforced by the chelating phosphine arm in these complexes, leads to strongly electrophilic EII centres which readily bind nucleophiles, reversibly in the case of NH3 . Further, the GeII centre in 4 a/b readily abstracts the fluoride ion from [SbF6 ]- to form the fluoro-germylene complex PhR DippGe(F)Ni(PPh3 )3 9, despite this GeII centre simultaneously being a σ-donating ligand towards Ni0 . Alongside the observed catalytic ability of 4 and 5 in the hydrosilylation of alkynes and alkenes, this forms an exciting introduction to a multi-talented ligand class in cationic single-centre ambiphiles.

Anthracene + Pyrene Solid Mixtures: Eutectic and Azeotropic Character.

Project description:To better characterize the thermodynamic behavior of a binary polycyclic aromatic hydrocarbon mixture, thermochemical and vapor pressure experiments were used to examine the phase behavior of the anthracene (1) + pyrene (2) system. A solid-liquid phase diagram was mapped for the mixture. A eutectic point occurs at 404 K at x(1) = 0.22. A model based on eutectic formation can be used to predict the enthalpy of fusion associated with the mixture. For mixtures that contain x(1) < 0.90, the enthalpy of fusion is near that of pure pyrene. This and X-ray diffraction results indicate that mixtures of anthracene and pyrene have pyrene-like crystal structures and energetics until the composition nears that of pure anthracene. Solid-vapor equilibrium studies show that mixtures of anthracene and pyrene form solid azeotropes at x(1) of 0.03 and 0.14. Additionally, mixtures at x(1) = 0.99 sublime at the vapor pressure of pure anthracene, suggesting that anthracene behavior is not significantly influenced by x(2) = 0.01 in the crystal structure.

Lewis Acidic Boranes, Lewis Bases, and Equilibrium Constants: A Reliable Scaffold for a Quantitative Lewis Acidity/Basicity Scale.

Project description:A quantitative Lewis acidity/basicity scale toward boron-centered Lewis acids has been developed based on a set of 90 experimental equilibrium constants for the reactions of triarylboranes with various O-, N-, S-, and P-centered Lewis bases in dichloromethane at 20 °C. Analysis with the linear free energy relationship log KB =LAB +LBB allows equilibrium constants, KB , to be calculated for any type of borane/Lewis base combination through the sum of two descriptors, one for Lewis acidity (LAB ) and one for Lewis basicity (LBB ). The resulting Lewis acidity/basicity scale is independent of fixed reference acids/bases and valid for various types of trivalent boron-centered Lewis acids. It is demonstrated that the newly developed Lewis acidity/basicity scale is easily extendable through linear relationships with quantum-chemically calculated or common physical-organic descriptors and known thermodynamic data (ΔH BF3 ). Furthermore, this experimental platform can be utilized for the rational development of borane-catalyzed reactions.
