Janus particle-engineered structural lipiodol droplets for arterial embolization.

ABSTRACT: Embolization (utilizing embolic materials to block blood vessels) has been considered one of the most promising strategies for clinical disease treatments. However, the existing embolic materials have poor embolization effectiveness, posing a great challenge to highly efficient embolization. In this study, we construct Janus particle-engineered structural lipiodol droplets by programming the self-assembly of Janus particles at the lipiodol-water interface. As a result, we achieve highly efficient renal embolization in rabbits. The obtained structural lipiodol droplets exhibit excellent mechanical stability and viscoelasticity, enabling them to closely pack together to efficiently embolize the feeding artery. They also feature good viscoelastic deformation capacities and can travel distally to embolize finer vasculatures down to 40 μm. After 14 days post-embolization, the Janus particle-engineered structural lipiodol droplets achieve efficient embolization without evidence of recanalization or non-target embolization, exhibiting embolization effectiveness superior to the clinical lipiodol-based emulsion. Our strategy provides an alternative approach to large-scale fabricate embolic materials for highly efficient embolization and exhibits good potential for clinical applications.

Project description:The advancement of imaging systems has significantly ameliorated various technologies, including Intelligence Surveillance Reconnaissance Systems and Guidance Systems, by enhancing target detection, recognition, identification, positioning, and tracking capabilities. These systems can be countered by deploying obscurants like smoke, dust, or fog to hinder visibility and communication. However, these counter-systems affect the visibility of both sides of the cloud. In this sense, this manuscript introduces a new concept of a smoke cloud composed of engineered Janus particles to conceal the target image on one side while providing clear vision from the other. The proposed method exploits the unique scattering properties of Janus particles, which selectively interact with photons from different directions to open up the possibility of asymmetric imaging. This approach employs a model that combines a genetic algorithm with Discrete Dipole Approximation to optimize the Janus particles' geometrical parameters for the desired scattering properties. Moreover, we propose a Monte Carlo-based approach to calculate the image formed as photons pass through the cloud, considering highly asymmetric particles, such as Janus particles. The effectiveness of the cloud in disguising a target is evaluated by calculating the Probability of Detection (PD) and the Probability of Identification (PID) based on the constructed image. The optimized Janus particles can produce a cloud where it is possible to identify a target more than 50% of the time from one side (PID > 50%) while the target is not detected more than 50% of the time from the other side (PD < 50%). The results demonstrate that the Janus particle-engineered smoke enables asymmetric imaging with simultaneous concealment from one side and clear visualization from the other. This research opens intriguing possibilities for modern obscurant design and imaging systems through highly asymmetric and inhomogeneous particles besides target detection and identification capabilities in challenging environments.

Project description:Computed tomography (CT) is the standard method to evaluate Lipiodol deposition after transarterial embolization (TAE) for a long period. However, iodine but not Lipiodol can be observed on CT. A minimally invasive other method to detect Lipiodol has been needed to evaluate accurate evaluation after procedure. The purpose of this study was to evaluate the efficacy of using the rate of change in sound velocity caused by ultrasonic heating to reflect Lipiodol accumulation after TAE in a rat liver tumor model. We analyzed the association of this developed technique with CT images and histological findings. Eight rats bearing N1S1 cells were prepared. After confirmation of tumor development in a rat liver, Lipiodol was injected via the hepatic artery. Seven days after TAE, CT scan and sound velocity changes caused by ultrasonic heating were measured, and then the rats were sacrificed. An ultrasonic pulse-echo method was used to measure the sound velocity. The temperature coefficient of the sound velocity in each treated tumor was evaluated and compared with the mean CT value and the histological Lipiodol accumulation ratio. Pearson's correlation coefficients were calculated to assess the correlation between the measured values. The correlation coefficient (r) of the mean CT value and histological Lipiodol accumulation ratio was 0.835 (p = 0.010), which was considered statistically significant. Also, those of the temperature coefficient of the sound velocity and the histological Lipiodol accumulation ratio were statistically significant (r = 0.804; p = 0.016). To our knowledge, this is the first study that reported the efficacy of ultrasonic heating to detect Lipiodol accumulation in rat liver tumors after TAE. Our results suggest that the rate of change in sound velocity caused by ultrasonic heating can be used to evaluate Lipiodol accumulation in liver tumors after TAE, and thus could represent an alternative to CT in this application. This new innovative technique is easy to treat and less invasive in terms of avoiding radiation compared with CT.
