Adult Mouse Leptomeninges Exhibit Regional and Age-related Cellular Heterogeneity Implicating Mental Disorders.

ABSTRACT: The leptomeninges envelop the central nervous system (CNS) and contribute to cerebrospinal fluid (CSF) production and homeostasis. We analyzed the meninges overlying the anterior or posterior forebrain in the adult mouse by single nuclear RNA-sequencing (snucRNA-seq). This revealed regional differences in fibroblast and endothelial cell composition and gene expression. Surprisingly, these non-neuronal cells co-expressed genes implicated in neural functions. The regional differences changed with aging, from 3 to 18 months. Cytokine analysis revealed specific soluble factor production from anterior vs posterior meninges that also altered with age. Secreted factors from the leptomeninges from different regions and ages differentially impacted the survival of anterior or posterior cortical neuronal subsets, neuron morphology, and glia proliferation. These findings suggest that meningeal dysfunction in different brain regions could contribute to specific neural pathologies. The disease-associations of meningeal cell genes differentially expressed with region and age were significantly enriched for mental and substance abuse disorders.

Project description:Sortilin is a member of the vacuolar protein sorting 10 protein (VPS10P) domain receptor family, which carries out signal transduction and protein transport in cells. Sortilin serves as the third, G-protein uncoupled, receptor of neurotensin that can modulate various brain functions. More recent data indicate an involvement of sortilin in mood disorders, dementia and Alzheimer-type neuropathology. However, data regarding the normal pattern of regional and cellular expression of sortilin in the human brain are not available to date. Using postmortem adult human brains free of neuropathology, the current study determined sortilin immunoreactivity (IR) across the entire brain. Sortilin IR was broadly present in the cerebrum and subcortical structures, localizing to neurons in the somatodendritic compartment, but not to glial cells. In the cerebrum, sortilin IR exhibited differential regional and laminar patterns, with pyramidal, multipolar and polymorphic neurons in cortical layers II-VI, hippocampal formation and amygdaloid complex more distinctly labeled relative to GABAergic interneurons. In the striatum and thalamus, numerous small-to-medium sized neurons showed light IR, with a small group of large sized neurons heavily labeled. In the midbrain and brainstem, sortilin IR was distinct in neurons at the relay centers of descending and ascending neuroanatomical pathways. Dopaminergic neurons in the substantia nigra, cholinergic neurons in the basal nuclei of Meynert and noradrenergic neurons in the locus coeruleus co-expressed strong sortilin IR in double immunofluorescence. In comparison, sortilin IR was weak in the olfactory bulb and cerebellar cortex, with the mitral and Purkinje cells barely visualized. A quantitative analysis was carried out in the lateral, basolateral, and basomedial nuclei of the amygdaloid complex, as well as cortical layers II-VI, which established a positive correlation between the somal size and the intensity of sortilin IR among labeled neurons. Together, the present study demonstrates a predominantly neuronal expression of sortilin in the human brain with substantial regional and cell-type variability. The enriched expression of sortilin in pyramidal, dopaminergic, noradrenergic and cholinergic neurons suggests that this protein may be particularly required for signal transduction, protein trafficking and metabolic homeostasis in populations of relatively large-sized projective neurons.

Project description:IntroductionCommon mental disorders represent psychiatric co-morbidity in medical illness, which leads to poor adherence to treatment, increased exposure to diagnostic procedures and the cost of treatment, longer hospital stay, and increasing the risk of complications that result in morbidity and mortality among patients admitted to non-psychiatric wards. There is a dearth of evidence related to the prevalence of common mental disorders and associated factors among adult patients admitted to non-psychiatric wards, particularly in the study area. This study aimed to assess the prevalence of common mental disorders and associated factors among adult patients admitted to non-psychiatric wards of public hospitals in the Harari region, eastern Ethiopia.MethodsAn institutional-based cross-sectional study was conducted among 640 randomly selected patients admitted to non-psychiatric wards from November 15 to December 15, 2022. A systematic random sampling technique was employed to select the study participants. Data were collected by interviewer-administered structured and semi-structured questionnaires. Self-report questionnaire (SRQ-20) was used to assess the presence of common mental disorders. The collected data were entered into Epi-data version 3.1 and exported to STATA version 14 for analysis. Bivariable and multivariable logistic regression were used to evaluate the association between independent and the outcome variable. Variables with a p-value < 0.05 were taken as statistically significant with an adjusted odds ratio and 95% confidence interval.ResultsThe prevalence of common mental disorders among adult patients admitted to non-psychiatric wards was found to be 45.3%, with a 95% CI: of 41.3-49.2. Age 41-51 years (AOR = 1.732, 95% CI: 1.030, 2.913), age 51 and above (AOR = 2.429, 95% CI: 1.515, 3.894), staying at hospital for 1-2 weeks (AOR = 1.743, 95% CI: 1.065, 2.853), staying at hospital for more than 4 weeks (AOR = 2.12, 95% CI: 1.77, 3.29), history of mental illness (AOR = 5.841, 95% CI: 2.274, 15.004), stressful life events (AOR = 1.876, 95% CI: 1.206, 2.9196), current substance use (AOR = 1.688, 95% CI: 1.75, 2.650), and poor social support (AOR = 2.562, 95% CI:1.166, 5.629) were factors significantly associated with common mental disorders.ConclusionThe prevalence of common mental disorders among patients admitted to non-psychiatric wards was high. It appears to be significantly associated with age, length of hospital stay, history of mental illness, stressful life events, current substance use, and social support. The study suggested that patients who are admitted to non-psychiatric wards should be screened for common mental disorders and its associated factors as part of routine inpatient care.
