Effects of Algicidal Macrophyte Metabolites on Cyanobacteria, Microcystins, Other Plankton, and Fish in Microcosms.

ABSTRACT: To control harmful algae blooms (HABs), methods based on natural mechanisms are now required. We investigated the effects of an algicide derived from macrophyte metabolites, namely mixtures of gallic, tetradecanoic, heptanoic, and octanoic acids (1:1:1:1 mass ratio, a total concentration of 14 mg/L), on the biomass of cyanobacteria and other plankton and the production of microcystins under experimental conditions. Two types of microcosms have been created: simple (microalgae, cyanobacteria, and zooplankton) and complex (microalgae, cyanobacteria, zooplankton, and planktivorous fish). We observed the dynamics of the phytoplankton structure, the concentrations of microcystins and chlorophyll-a, hydrochemistry, and the status of zooplankton and fish in both types of microcosms with and without algicide for one month (from 19 July to 19 August 2021). The introduction of algicide caused changes in phytoplankton structure, a drop in cyanobacterial biomass, and a decrease in the total concentration of microcystins. Surprisingly, the contributions of the most toxic microcystins (LR form) were higher in both types of microcosms exposed to algicide than in microcosms without algicide. The inhibitory effect on the cyanobacterial biomass was most significant in complex ecosystems (containing fish), while it was only observed at the end of the exposure in simple ecosystems. Not only algicide but also phytoplankton consumed by fish and zooplankton, as well as nutrient excretory activity by both consumers, seem to have impact on cyanobacterial biomass. This study found that the using chemical substances similar to macrophyte metabolites can help regulate HABs and cyanotoxins. However, the results differ depending on ecosystem type.

Project description:Plankton communities consist of complex microbial consortia that change over time. These fluctuations can be only partially explained by limiting resources. Biotic factors such as herbivores and pathogens also contribute to the control of algal blooms. Here we address the effects of algicidal bacteria on a natural plankton community in an indoor enclosure experiment. The algicidal bacteria, introduced into plankton taken directly from the North Sea during a diatom bloom, caused the rapid decline of the bloom-forming Chaetoceros socialis within only 1 day. The haptophyte Phaeocystis, in contrast, is resistant to the lytic bacteria and could benefit from the removal of the competitor, as indicated by an onset of a bloom in the treated enclosures. This cascading effect caused by the bacterial pathogen accelerated the succession of Phaeocystis, which bloomed with a delay of only several weeks in the in situ waters at Helgoland Roads in the North Sea. The algicidal bacteria can thus modulate the community within the limits of the abiotic and biotic conditions of the local environment. Implications of our findings for plankton ecosystem functioning are discussed.IMPORTANCE Plankton communities change on a seasonal basis in temperate systems, with distinct succession patterns; this is mainly due to algal species that have their optimal timing relative to environmental conditions. We know that bacterial populations are also instrumental in the decay and termination of phytoplankton blooms. Here, we describe algicidal bacteria as modulators of this important species succession. Upon treatment of a natural plankton consortium with an algicidal bacterium, we observed a strong shift in the phytoplankton community structure, compared to controls, resulting in formation of a succeeding Phaeocystis bloom. Blooms of this alga have a substantial impact on global biogeochemical and ecological cycles, as they are responsible for a substantial proportion of primary production during spring in the North Sea. We propose that one of the key factors influencing such community shifts may be algicidal bacteria.

Project description:UnlabelledCopper sulfate (CuSO4) has been widely used as an algicide to control harmful cyanobacterial blooms (CyanoHABs) in freshwater lakes. However, there are increasing concerns about this application, due mainly to the general toxicity of CuSO4 to other aquatic species and its long-term persistence in the environment. This study reported the isolation and characterization of two natural algicidal compounds, i.e., tryptamine and tryptoline, from Streptomyces eurocidicus JXJ-0089. At a concentration of 5 μg/ml, both compounds showed higher algicidal efficiencies than CuSO4 on Microcystis sp. FACHB-905 and some other harmful cyanobacterial strains. Tryptamine and tryptoline treatments induced a degradation of chlorophyll and cell walls of cyanobacteria. These two compounds also significantly increased the intracellular oxidant content, i.e., superoxide anion radical (O2 (-)) and malondialdehyde (MDA), but reduced the activity of intracellular reductants, i.e., superoxide dismutase (SOD), of cyanobacteria. Moreover, tryptamine and tryptoline treatments significantly altered the internal and external contents of microcystin-LR (MC-LR), a common cyanotoxin. Like CuSO4, tryptamine and tryptoline led to releases of intracellular MC-LR from Microcystis, but with lower rates than CuSO4 Tryptamine and tryptoline (5 μg/ml) in cyanobacterial cultures were completely degraded within 8 days, while CuSO4 persisted for months. Overall, our results suggest that tryptamine and tryptoline could potentially serve as more efficient and environmentally friendly alternative algicides than CuSO4 in controlling harmful cyanobacterial blooms.ImportanceCyanobacterial harmful algal blooms (CyanoHABs) in aquatic environments have become a worldwide problem. This study reports the isolation and characterization of two natural algicidal compounds from a streptomycete strain, Streptomyces eurocidicus JXJ-0089. Our results suggest that the identified algicides could potentially serve as more efficient and environmentally friendly alternative algicides than CuSO4 in controlling harmful cyanobacterial blooms.
