Recent advances in enantioselective ring-opening polymerization and copolymerization.

ABSTRACT: Precisely controlling macromolecular stereochemistry and sequences is a powerful strategy for manipulating polymer properties. Controlled synthetic routes to prepare degradable polyester, polycarbonate, and polyether are of recent interest due to the need for sustainable materials as alternatives to petrochemical-based polyolefins. Enantioselective ring-opening polymerization and ring-opening copolymerization of racemic monomers offer access to stereoregular polymers, specifically enantiopure polymers that form stereocomplexes with improved physicochemical and mechanical properties. Here, we highlight the state-of-the-art of this polymerization chemistry that can produce microstructure-defined polymers. In particular, the structures and performances of various homogeneous enantioselective catalysts are presented. Trends and future challenges of such chemistry are discussed.

Carbodiimide Ring-Opening Metathesis Polymerization.

Project description:Controlled incorporation of nitrogen into macromolecular skeletons is a long-standing challenge whose resolution would enable the preparation of soft materials with the scalability of man-made plastics and functionality of Nature's proteins. Nylons and polyurethanes notwithstanding, nitrogen-rich polymer backbones remain scarce, and their synthesis typically lacks precision. Here we report a strategy that begins to address this limitation founded on a mechanistic discovery: ring-opening metathesis polymerization (ROMP) of carbodiimides followed by carbodiimide derivatization. An iridium guanidinate complex was found to initiate and catalyze ROMP of N-aryl and N-alkyl cyclic carbodiimides. Nucleophilic addition to the resulting polycarbodiimides enabled the preparation of polyureas, polythioureas, and polyguanidinates with varied architectures. This work advances the foundations of metathesis chemistry and opens the door to systematic investigations of structure-folding-property relationships in nitrogen-rich macromolecules.

Functional Degradable Polymers by Radical Ring-Opening Copolymerization of MDO and Vinyl Bromobutanoate: Synthesis, Degradability and Post-Polymerization Modification.

Project description:The synthesis of vinyl bromobutanoate (VBr), a new vinyl acetate monomer derivative obtained by the palladium-catalyzed vinyl exchange reaction between vinyl acetate (VAc) and 4-bromobutyric acid is reported. The homopolymerization of this new monomer using the RAFT/MADIX polymerization technique leads to the formation of novel well-defined and controlled polymers containing pendent bromine functional groups able to be modified via postpolymerization modification. Furthermore, the copolymerization of vinyl bromobutanoate with 2-methylene-1,3-dioxepane (MDO) was also performed to deliver a range of novel functional degradable copolymers, poly(MDO-co-VBr). The copolymer composition was shown to be able to be tuned to vary the amount of ester repeat units in the polymer backbone, and hence determine the degradability, while maintaining a control of the final copolymers' molar masses. The addition of functionalities via simple postpolymerization modifications such as azidation and the 1,3-dipolar cycloaddition of a PEG alkyne to an azide is also reported and proven by (1)H NMR spectroscopy, FTIR spectroscopy, and SEC analyses. These studies enable the formation of a novel class of hydrophilic functional degradable copolymers using versatile radical polymerization methods.

Cationic-anionic synchronous ring-opening polymerization.

Project description:Chemical reactions with incompatible mechanisms (such as nucleophilic reactions and electrophilic reactions, cationic polymerization and anionic polymerization) are usually difficult to perform simultaneously in one-pot. In particular, synchronous cationic-anionic polymerization has been an important challenge in the field of polymer synthesis due to possible coupling termination of both chain ends. We recently found that such terminal couplings can be significantly inhibited by a bismuth salt with a strong nucleophilic anion (e.g., BiCl3) and disclosed the mechanism. Accordingly, we propose a cationic-anionic polymerization (CAP) method where cationic ring-opening polymerization (CROP) of 2-oxazolines (Ox) and anionic ring-opening polymerization (AROP) of cyclic esters (CE) can be initiated sequentially and propagated simultaneously in one-pot, using bismuth salts as the initial initiators, to afford a multifunctional copolymer polyoxazoline-block-polyester (POx-b-PCE). Furthermore, a block copolymer PAPOZ20-b-PCL5 synthesized by CAP can self-assemble into micellar aggregates, which exhibit excellent intrinsic antibacterial activities without loading any extra antibiotic components. Overall, such a CAP method opens new avenues for synthesizing multi-component copolymers and biomaterials.

Michael Addition-Elimination Ring-Opening Polymerization.

Project description:A cyclic thioenone system capable of controlled ring-opening polymerization (ROP) is presented that leverages a reversible Michael addition-elimination (MAE) mechanism. The cyclic thioenone monomers are easy to access and modify and for the first time incorporate the dynamic reversibility of MAE with chain-growth polymerization. This strategy features mild polymerization conditions, tunable functionalities, controlled molecular weights (Mn), and narrow dispersities. The obtained polythioenones exhibit excellent optical transparency and good mechanical properties and can be depolymerized to recover the original monomers. Density functional theory (DFT) calculations of model reactions offer insights into the role of monomer conformation in the polymerization process, as well as explaining divergent reactivity observed in seven-membered thiepane (TP) and eight-membered thiocane (TC) ring systems. Collectively, these findings demonstrate the feasibility of MAE mechanisms in ring-opening polymerization and provide important guidelines toward future monomer designs.

Degradable Polyampholytes from Radical Ring-Opening Copolymerization Enhance Cellular Cryopreservation.

Project description:Macromolecular cryoprotectants based on polyampholytes are showing promise as supplemental cryoprotectants alongside conventional DMSO-based freezing. Here we exploit radical ring-opening (ter)polymerization to access ester-containing cryoprotective polyampholytes, which were shown to be degradable. Using a challenging cell monolayer cryopreservation model, the degradable polyampholytes were found to enhance post-thaw recovery when supplemented into DMSO. This demonstrates that degradable macromolecular cryoprotectants can be developed for application in biotechnology and biomedicine.

Synthesis of PNVP-Based Copolymers with Tunable Thermosensitivity by Sequential Reversible Addition⁻Fragmentation Chain Transfer Copolymerization and Ring-Opening Polymerization.

Project description:Through the reversible addition⁻fragmentation chain transfer (RAFT) copolymerization of 3-ethyl-1-vinyl-2-pyrrolidone (C₂NVP) and N-vinylpyrrolidone (NVP), a series of well-defined P(C₂NVP-co-NVP) copolymers were synthesized (Mn = ca. 8000 to 16,000 and Mw/Mn <1.5) by using a difunctional chain transfer agent, S-(1-methyl-4-hydroxyethyl acetate) O-ethyl xanthate (MHEX). Copolymerizing kinetics and different monomer ratio in feeds were conducted to study the apparent monomer reaction rate and reactivity ratios of NVP and C₂NVP, which indicated similar reaction rates and predominantly ideal random copolymers for the two monomers. The Tgs of the obtaining P(C₂NVP-co-NVP) copolymers significantly corresponded to not only molecular weights MWs but also copolymer compositions. These copolymers presented characteristic lower critical solution temperatures (LCST) behavior. We then studied the cloud points (CPs) of the copolymers with varying MWs and compositions. With different MWs, the CPs were linearly decreased from ca. 51 to 45 °C. With different compositions, the CPs of the copolymers decreased from ca. 48 to 29 °C with C₂NVP content (i.e., from 60.8 to 89.9 mol %). Fitting the CPs by the theoretical equation, the result illustrated that the introduction of more hydrophobic units of C₂NVP suppressed the hydrophilic interaction between the polymer chain and water. We then successfully proceeded the chain extension through the ring-opening polymerization (ROP) of ε-caprolactone (CL) to the synthesis of a novel P(C₂NVP-co-NVP)-b-PCL amphiphilic block copolymer (Mn,NMR = 14,730 and Mw/Mn = 1.59). The critical micelle concentration (CMC) of the block copolymer had a value of ca. 1.46 × 10-4 g/L. The block copolymer micelle was traced by dynamic light scattering (DLS), obtaining thermosensitive behaviors with a particle size of ca. 240 nm at 25 °C and ca. 140 nm at 55 °C, respectively.

Photoinitiated Cationic Ring-Opening Polymerization of Octamethylcyclotetrasiloxane.

Project description:Photochemical techniques have recently been revitalized as they can readily be adapted to different polymerization modes to yield a wide range of complex macromolecular structures. However, the implementation of the photoinduced cationic methods in the polymerization of cyclic siloxane monomers has scarcely been investigated. Octamethylcyclotetrasiloxane (D4) is an important monomer for the synthesis of polydimethylsiloxane (PDMS) and its copolymers. In this study, the cationic ring-opening polymerization (ROP) of D4, initiated by diphenyl iodonium hexafluorophosphate (DPI), has been studied. Both direct and indirect initiating systems acting at broad wavelength using benzophenone and pyrene were investigated. In both systems, photochemically generated protonic acids and silylium cations are responsible for the polymerization. The kinetics of the polymerization are followed by viscosimetry and GPC analyses. The reported approach may overcome the problems associated with conventional methods and therefore represents industrial importance for the fabrication of polysiloxanes.

Thermodynamic Presynthetic Considerations for Ring-Opening Polymerization.

Project description:The need for polymers for high-end applications, coupled with the desire to mimic nature's macromolecular machinery fuels the development of innovative synthetic strategies every year. The recently acquired macromolecular-synthetic tools increase the precision and enable the synthesis of polymers with high control and low dispersity. However, regardless of the specificity, the polymerization behavior is highly dependent on the monomeric structure. This is particularly true for the ring-opening polymerization of lactones, in which the ring size and degree of substitution highly influence the polymer formation properties. In other words, there are two important factors to contemplate when considering the particular polymerization behavior of a specific monomer: catalytic specificity and thermodynamic equilibrium behavior. This perspective focuses on the latter and undertakes a holistic approach among the different lactones with regard to the equilibrium thermodynamic polymerization behavior and its relation to polymer synthesis. This is summarized in a monomeric overview diagram that acts as a presynthetic directional cursor for synthesizing highly specific macromolecules; the means by which monomer equilibrium conversion relates to starting temperature, concentration, ring size, degree of substitution, and its implications for polymerization behavior are discussed. These discussions emphasize the importance of considering not only the catalytic system but also the monomer size and structure relations to thermodynamic equilibrium behavior. The thermodynamic equilibrium behavior relation with a monomer structure offers an additional layer of complexity to our molecular toolbox and, if it is harnessed accordingly, enables a powerful route to both monomer formation and intentional macromolecular design.

Poly(thioester) by Organocatalytic Ring-Opening Polymerization.

Project description:Organocatalysts typically used for the ring-opening polymerization (ROP) of cyclic ester monomers are applied to a thiolactone, ε-thiocaprolactone (tCL). In the absence of an H-bond donor, a nucleophilic polymerization mechanism is proposed. Despite the decreased ability of thioesters and thiols (versus esters and alcohols) to H-bond, H-bonding organocatalysts-a thiourea in combination with an H-bond accepting base-are also effective for the ROP of tCL. The increased nucleophilicity of thiols (versus alcohols) is implicated in the increased Mw/Mn of the poly(thiocaprolactone) versus poly(caprolactone), but deleterious transesterification is suppressed in the presence of a thiourea. The thioester monomer, tCL, is shown to be thermodynamically similar to ε-caprolactam but kinetically similar to ε-caprolactone.
