Azetidine synthesis by La(OTf)₃-catalyzed intramolecular regioselective aminolysis of cis-3,4-epoxy amines.

ABSTRACT: Azetidine is a prevalent structural motif found in various biologically active compounds. In this research paper, we report La(OTf)₃-catalyzed intramolecular regioselective aminolysis of cis-3,4-epoxy amines to afford azetidines. This reaction proceeded in high yields even in the presence of acid-sensitive and Lewis basic functional groups.

Project description:A series of highly functionalized carbocycles was efficiently prepared via the selective cyclisation of unsaturated acetals and ketals in the presence of only 1 mol% of Bi(iii) or Fe(iii) triflates as the catalysts at room temperature, with yields ranging from 60 to 90%. With Bi(OTf)3 catalysis, α,β-unsaturated ether carbocycles are formed selectively, whereas with the Fe(OTf)3 system, a cycloisomerisation to carbocyclic diethers is mainly obtained. This acetal/olefin cyclisation could be run at a multi-gram scale and compound 2c could be obtained on a 300 gram-scale with a yield of 69% after precipitation in hexane.

Temperature dependence of turnover in a Sc(OTf)3-catalyzed intramolecular Schmidt reaction.

Project description:The intramolecular Schmidt reaction of ketones and tethered azides is an efficient method for the generation of amides and lactams. This reaction is catalyzed by Lewis acids, which tightly bind the strongly basic amide product and result in product inhibition. We report herein conditions to achieve a catalytic Schmidt reaction using substoichiometric amounts of the heat-stable Lewis acid Sc(OTf)3. This species was shown to effectively release products of the Schmidt reaction in a temperature-dependent fashion. Thus, heat was able to promote catalyst turnover. A brief substrate scope was conducted using these conditions.

Regioselective Cleavage of 3,4-Epoxy Alcohols with Substituted Alkynylaluminum Reagents: Application to the Stereoselective Synthesis of Polypropionates.

Project description:The reaction of 3-O-substituted propynyl aluminum reagents with a TIPS-protected 2,3-epoxy alcohol and several diastereomeric 2-methyl-3,4-epoxy alcohols offers a convenient synthetic approach for the subsequent preparation and epoxidation of allylic alcohols. The yields are low to moderate and the regioselectivity (internal vs external attack) similar to that of the standard diethylpropynyl aluminum reagent. The TMS-acetylide alane reagent gives improved yields and good regioselectivity, and constitutes the reagent of choice for the cleavage of the hindered epoxides. This attractive methodology is applied to the elaboration of two advanced polypropionate precursor fragments.

Synthesis of stereotetrads by regioselective cleavage of diastereomeric MEM-protected 2-methyl-3,4-epoxy alcohols with diethylpropynyl aluminum.

Project description:The regioselectivity of the epoxide ring opening of 2-methyl-3,4-epoxy alcohols with diethylpropynylalane has been studied as a function of the C1 alcohol protecting group. An efficient selective method was developed using MEM as the protecting group. The reaction proceeded in a highly regioselective manner providing the useful 1,3-diol motif. The undesired 1,4-diol product produced by some free alcohol diastereomers was not observed. This highly stereoselective method provides access to termini-differentiated stereotetrads, which are essential building bocks for polypropionate synthesis.

Project description:The first enantioselective aminolysis of aromatic trans-2,3-epoxy sulfonamides has been accomplished, which was efficiently catalyzed by a Gd-N,N'-dioxide complex. Under the directing effect of the sulfonamide moiety the ring-opening reaction proceeded selectively at the C-3 position in a highly enantioselective manner furnishing various Ts- and SES-protected 3-amino-3-phenylpropan-2-olamines as products.

Regioselective α-Cyanation of Unprotected Alicyclic Amines.

Project description:Secondary alicyclic amines are converted to α-aminonitriles via addition of TMSCN to their corresponding imines, intermediates that are produced in situ via the oxidation of amine-derived lithium amides with simple ketone oxidants. Amines with an existing α-substituent undergo regioselective α'-cyanation even if the C-H bonds at that site are less activated. Amine α-arylation can be combined with α'-cyanation to generate difunctionalized products in a single operation.

Project description:The first catalytic, highly C3-selective, stereosepecific ring-opening reaction of 2,3-epoxy alcohols and 2,3-epoxy sulfonamides has been accomplished. This process was efficiently promoted by W-salts, and the developed method was applicable to various epoxides with diverse N- and O-nucleophiles affording the products in good to excellent yields (up to 95%) and generally with high regioselectivities (C3:C2 up to >99:1).
