Phenotypic and genetic dissection of the contents of important metallic elements in hybrid rice grown in cadmium-contaminated paddy fields.

ABSTRACT: Rice (Oryza sativa L.) is a staple food that feeds over half of the world's population, and the contents of metallic elements in rice grain play important roles in human nutrition. In this study, the contents of important metallic elements were determined by ICP-OES, and included cadmium (Cd), zinc (Zn), manganese (Mn), copper (Cu), iron (Fe), nickel (Ni), calcium (Ca), and magnesium (Mg) in brown rice, in the first node from the top (Node 1), in the second node from the top (Node 2), and in roots of 55 hybrids and their parental lines. The heritability of metallic element contents (MECs), the general combining ability (GCA) for MEC, and the correlation between MECs in different organs/tissues of hybrids were also analyzed. The results indicated that: (1) there was a positive correlation between the contents of Cd and Zn in nodes and roots, but a negative correlation between the contents of Cd and Zn in brown rice of the hybrids(2) the GCA for MECs can be used to evaluate the ability of the parental lines to improve the metal contents in brown rice of the hybrids(3) the contents of Cd, Zn, Ca, and Mg in brown rice were mainly affected by additive genetic effects(4) the restorer lines R2292 and R2265 can be used to cultivate hybrids with high Zn and low Cd contents in the brown rice.

Project description:Endophytic bacteria play potentially important roles in the processes of plant adaptation to the environment. Understanding the composition and dynamics of endophytic bacterial communities under heavy metal (HM) stress can reveal their impacts on host development and stress tolerance. In this study, we investigated root endophytic bacterial communities of different rice cultivars grown in a cadmium (Cd)-contaminated paddy field. These rice cultivars are classified into low (RBQ, 728B, and NX1B) and high (BB and S95B) levels of Cd-accumulating capacity. Our metagenomic analysis targeting 16S rRNA gene sequence data reveals that Proteobacteria, Firmicutes, Actinobacteria, Acidobacteria, Bacteroidetes, and Spirochaetes are predominant root endophytic bacterial phyla of the five rice cultivars that we studied. Principal coordinate analysis shows that the developmental stage of rice governs a larger source of variation in the bacterial communities compared to that of any specific rice cultivar or of the root Cd content. Endophytic bacterial communities during the reproductive stage of rice form a more highly interconnected network and exhibit higher operational taxonomic unit numbers, diversities, and abundance than those during the vegetative stage. Forty-five genera are significantly correlated with Cd content in rice root, notably including positive-correlating Geobacter and Haliangium; and negative-correlating Pseudomonas and Streptacidiphilus. Furthermore, Phylogenetic Investigation of Communities by Reconstruction of Unobserved States analysis shows that functional pathways, such as biosynthesis of siderophore and type II polyketide products, are significantly enhanced during the reproductive stage compared to those during the vegetative stage under Cd stress. The isolated endophytic bacteria from the Cd-contaminated rice roots display high Cd resistance and multiple traits that may promote plant growth, suggesting their potential application in alleviating HM stress on plants. This study describes in detail for the first time the assemblage of the bacterial endophytomes of rice roots under Cd stress and may provide insights into the interactions among endophytes, plants, and HM contamination.

Project description:Cadmium (Cd) and arsenic (As) contamination risk in paddy soils has raised global concern. In order to scientifically and objectively evaluate the bioavailability of soil Cd, As and the risk of Cd or As threshold in contaminated farmland, this study was conducted to investigate different types of extractants for their potential extraction efficiency of Cd and As. Soils from two different parent materials in Hunan, Yueyang and Yiyang, typical double-cropping rice production areas in the south of China, were used as test soils. The extraction capabilities of 10 extractants (ultrapure water, 0.1 mol/L HCl, 1.0 mol/L NH4OAc, CaCl2-DTPA, 0.01 mol/L CaCl2, 0.1 mol/L CaCl2, 0.5 mol/L NaH2PO4, 0.05 mol/L NaHCO3, 0.1 mol/L NaNO3, 0.1 mol/L HNO3), were compared for their extraction capabilities of soil available Cd and As. Meanwhile, the content of Cd and As in plants issues and grains of rice was monitored during harvest, and the Cd, As content correlation between extracted forms and rice was analyzed. The results showed that the HCl, CaCl2, HNO3, and CaCl2-DTPA solutions exhibited high extraction efficiency for Cd (42.2-88.4 %); for As, NaH2PO4, HCl, and HNO3 have the extraction efficiency (0.85-23.4 %). The concentration of Cd extracted by 0.01 mol/L CaCl2 was significantly positively correlated with Cd levels in rice. The potential risk extraction threshold of CaCl2 in sandy loam soil was 0.178 mg/kg, while it was 0.312 mg/kg in clay soil. The concentration of As extracted by CaCl2-DTPA and 0.05 mol/L NaHCO3 in clay soil was significantly positively correlated with As levels in rice, the potential risk extraction thresholds were 0.115 mol/L and 0.106 mg/kg, respectively. These investigations indicated that the heavy metals extraction methods by 0.01 mol/L CaCl2, CaCl2-DTPA, and 0.05 mol/L NaHCO3 could reflect the Cd and As pollution degree in farmland and suggest their potential to serve as methods for assessing the risk of Cd and As pollution in sandy loam and clay paddy soil.

Project description:Co-cropping is an eco-friendly strategy to improve the phytoremediation capacity of plants growing in soils contaminated with heavy metals such as cadmium (Cd). This study was conducted to investigate the effects of co-cropping Indian mustard (Brassicajuncea) and silage maize (Zeamays) and applying peat on the phytoremediation of a Cd-contaminated acid paddy soil via characterizing plant growth and Cd uptake in pot experiments. There were six planting patterns (Control: no plants; MI-2 and MI-4: mono-cropping of Indian mustard at low and high densities, respectively; MS: mono-cropping of silage maize; CIS-2 and CIS-4: co-cropping of Indian mustard at low and high densities with silage maize, respectively) and two application rates of peat (NP: 0; WP: 30 g kg-1). When Indian mustard and silage maize were co-cropped, the shoot biomass of Indian mustard plants per pot was significantly (p < 0.05) lower than that obtained in the mono-cropping systems, with a substantial reduction (55-72%) in the same plant density group. The shoot biomass of silage maize plants in the mono-cropping systems did not differ significantly from that in the co-cropping systems regardless of the density of Indian mustard. The growth-promoting effect of the peat application was more pronounced in Indian mustard than silage maize. Under the low density of Indian mustard, the co-cropping systems significantly (p < 0.05) decreased Cd uptake by silage maize. Additionally, soil amendment with peat significantly (p < 0.05) increased shoot Cd removal rate and Cd translocation factor value in the co-cropping systems. Taken together, the results demonstrated that silage maize should be co-cropped with Indian mustard at an appropriate density in Cd-polluted soils to achieve simultaneous remediation of Cd-contaminated soils (via Indian mustard) and production of crops (here, silage maize). Peat application was shown to promote the removal of Cd from soil and translocation of Cd into shoots and could contribute to enhanced phytoremediation of Cd-contaminated acid paddy soil.
