Project description:Photochemical transformations enable exquisite spatiotemporal control over biochemical processes; however, methods for reliable manipulations of biomolecules tagged with biocompatible photo-sensitive reporters are lacking. Here we created a high-affinity binder specific to a photolytically removable caging group. We utilized chemical modification or genetically encoded incorporation of noncanonical amino acids to produce proteins with photocaged cysteine or selenocysteine residues, which were used for raising a high-affinity monoclonal antibody against a small photoremovable tag, 4,5-dimethoxy-2-nitrobenzyl (DMNB) group. Employing the produced photocage-selective binder, we demonstrate selective detection and immunoprecipitation of a variety of DMNB-caged target proteins in complex biological mixtures. This combined orthogonal strategy permits photocage-selective capture and light-controlled traceless release of target proteins for a myriad of applications in nanoscale assays.

Project description:In this work, we describe the synthesis, characterization, and ultimate in vivo assessment of second-generation insulin photoactivated depot (PAD) materials. These are the first to use visible light to stimulate insulin release and have an in vivo performance that is 28-fold improved relative to first-generation materials. This improvement is due to two major factors linked to the utilized chemistry: (1) we have incorporated the coumarin photocleavable group, which increases the photorelease wavelength into the visible range, enhancing tissue penetration of the light; (2) phototoggling of insulin solubility is produced by linking three insulin molecules to a central bridge via light cleaved groups, and not by bonding to a large polymer. The resulting trimer is, therefore, highly dense (87% insulin dry w/w) but retains the insolubility required of the approach. Only after irradiation with visible light is native, soluble insulin is released from the dermal depot. This high density increases the amount and ease of insulin release, as the density of photolytic groups is 10-20-fold higher than in polymer-based first-generation materials. We have synthesized new azide-terminated coumarin linkers that we react with the amine groups of insulin. Using mass spectrometry methods, we identify the sites of reaction and purify individual isomers, which we demonstrate have in vitro photolysis rates that are within a factor of 2 of each other. We then reacted these terminal azide groups with a tridentate strained alkyne linker. We show that the resulting insulin trimer is highly insoluble, but can be milled into injectable particles that release insulin only in response to light from a 406 nm light source. Finally, we demonstrate that these materials have a significantly improved in vivo performance, releasing 28-fold more insulin on a per energy basis than first-generation materials.

Project description:Fluorescent dithienylethene-based photochromic materials have been attracting considerable attention owing to their wide applications in biological and materials sciences. However, the limitations of detrimental UV irradiation for photocyclization, short emission lifetime, and inefficient photoresponsive speed still need to be addressed. Herein, a novel dithienylethene photochromic molecule, BFBDTE, has been prepared by the incorporation of a difluoroboron β-diketonate (BF2bdk) unit. The strong electron acceptor BF2bdk not only reduces the energy gap of the open isomer, ensuring visible light-controlled fluorescence switching, but also promotes intersystem crossing for the generation of thermally activated delayed fluorescence (TADF). Upon alternating irradiation with green and NIR light, BFBDTE presents a rare example of photochromism, fluorescence and TADF switching in various polar solvents and a poly(methyl methacrylate) (PMMA) film. Meanwhile, it shows rapid and well repeatable cyclization (12 s) and cycloreversion reactions (20 s) in PMMA, accompanied by fast TADF switching within 11 s. Furthermore, photo-electrochemical measurements reveal a remarkable on-off photoelectronic response (photocurrent density ratio: I light/I dark = 684) between the open- and closed-form of BFBDTE. These remarkable merits make BFBDTE promising for photoswitchable molecular devices, optical memory storage systems, NIR detectors, and photoelectric switching.

Project description:We present a method for using dynamic light scattering in the single-scattering limit to measure the viscoelastic moduli of soft materials. This microrheology technique only requires a small sample volume of 12 μL to measure up to six decades in time of rheological behavior. We demonstrate the use of dynamic light scattering microrheology (DLSμR) on a variety of soft materials, including dilute polymer solutions, covalently-crosslinked polymer gels, and active, biological fluids. In this work, we detail the procedure for applying the technique to new materials and discuss the critical considerations for implementing the technique, including a custom analysis script for analyzing data output. We focus on the advantages of applying DLSμR to biologically relevant materials: breast cancer cells encapsulated in a collagen gel and cystic fibrosis sputum. DLSμR is an easy, efficient, and economical rheological technique that can guide the design of new polymeric materials and facilitate the understanding of the underlying physics governing behavior of naturally derived materials.

Project description:In the field of materials science, dynamic molecular crystals have attracted significant attention as a novel class of energy-transducing materials. However, their development into becoming fully functional actuators remains somewhat limited. This study focuses on one family of dynamic crystalline materials and delves into exploring the efficiency of conversion of light energy to mechanical work. A simple setup is designed to determine a set of performance indices of anthracene-based crystals as an exemplary class of dynamic molecular crystals. The ability of these crystals to reversibly bend due to dimerization is realistically assessed from the perspective of the envisaged soft robotics applications, where wireless photomechanical grippers manipulate and assemble microscopic objects driven and controlled by light instead of lines and motors. The approach described here not only guides the quantification of responsive molecular crystals' actuation potential but also aims to attract an interdisciplinary interest to further develop this class of materials into controllable all-organic actuating elements to be used in microrobotics for engineering or biomedicine.

Project description:Soft materials can self-assemble into highly structured phases that replicate at the mesoscopic scale the symmetry of atomic crystals. As such, they offer an unparalleled platform to design mesostructured materials for light and sound. Here, we present a bottom-up approach based on self-assembly to engineer 3D photonic and phononic crystals with topologically protected Weyl points. In addition to angular and frequency selectivity of their bulk optical response, Weyl materials are endowed with topological surface states, which allow for the existence of one-way channels, even in the presence of time-reversal invariance. Using a combination of group-theoretical methods and numerical simulations, we identify the general symmetry constraints that a self-assembled structure has to satisfy to host Weyl points and describe how to achieve such constraints using a symmetry-driven pipeline for self-assembled material design and discovery. We illustrate our general approach using block copolymer self-assembly as a model system.

Project description:Truncated green fluorescent protein (GFP) with the 11th β-strand removed is potentially interesting for bioconjugation, imaging, and the preparation of semisynthetic proteins with novel spectroscopic or functional properties. Surprisingly, the truncated GFP generated by removing the 11th strand, once refolded, does not reassemble with a synthetic peptide corresponding to strand 11 but does reassemble following light activation. The mechanism of this process has been studied in detail by absorption, fluorescence, and Raman spectroscopy. The chromophore in this refolded truncated GFP is found to be in the trans configuration. Upon exposure to light a photostationary state is formed between the trans and cis conformations of the chromophore, and only truncated GFP with the cis configuration of the chromophore binds the peptide. A kinetic model describing the light-activated reassembly of this split GFP is discussed. This unique light-driven reassembly is potentially useful for controlling protein-protein interactions.

Project description:Hybrid hydrogels based on self-assembling low-molecular-weight gelator (LMWG) DBS-CONHNH2 (DBS = 1,3;2,4-dibenzylidene-d-sorbitol) and crosslinked polymer gelator (PG) PEGDM (poly(ethyleneglycol) dimethacrylate) are reported, and an active pharmaceutical ingredient (naproxen, NPX) is incorporated. The use of PEGDM as PG enhances the mechanical stiffness of the hybrid gel (G' increases from 400 to 4500 Pa) - the LMWG enhances its stability to very high frequency. Use of DBS-CONHNH2 as LMWG enables interactions with NPX and hence allows pH-mediated NPX release - the PG network is largely orthogonal and only interferes to a limited extent. Use of photo-activated PEGDM as PG enables spatially-resolved photo-patterning of robust hybrid gel domains within a preformed LMWG network - the presence of the LMWG enhances the spatial resolution. The photo-patterned multi-domain gel retains pH-mediated NPX release properties and directionally releases NPX into a compartment of higher pH. The two components within these hybrid PG/LMWG hydrogels therefore act largely independently of one another, although they do modify each others properties in subtle ways. Hybrid hydrogels capable of spatially controlled unidirectional release have potential applications in tissue engineering and drug-delivery.

Project description:Photolabile protecting groups (or "photocages") enable precise spatiotemporal control of chemical functionality and facilitate advanced biological experiments. Extant photocages exhibit a simple input-output relationship, however, where application of light elicits a photochemical reaction irrespective of the environment. Herein, we refine and extend the concept of photolabile groups, synthesizing the first Ca(2+) -sensitive photocage. This system functions as a chemical coincidence detector, releasing small molecules only in the presence of both light and elevated [Ca(2+) ]. Caging a fluorophore with this ion-sensitive moiety yields an "ion integrator" that permanently marks cells undergoing high Ca(2+) flux during an illumination-defined time period. Our general design concept demonstrates a new class of light-sensitive material for cellular imaging, sensing, and targeted molecular delivery.

Project description:Polymer topology dictates dynamic and mechanical properties of materials. For most polymers, topology is a static characteristic. In this article, we present a strategy to chemically trigger dynamic topology changes in polymers in response to a specific chemical stimulus. Starting with a dimerized PEG and hydrophobic linear materials, a lightly cross-linked polymer, and a cross-linked hydrogel, transformations into an amphiphilic linear polymer, lightly cross-linked and linear random copolymers, a cross-linked polymer, and three different hydrogel matrices were achieved via two controllable cross-linking reactions: reversible conjugate additions and thiol-disulfide exchange. Significantly, all the polymers, before or after topological changes, can be triggered to degrade into thiol- or amine-terminated small molecules. The controllable transformations of polymeric morphologies and their degradation herald a new generation of smart materials.
